ABSTRACT: An improved synthesis of a rhodamine boronic acid indicator is reported. This compound is used in an optimized data collection protocol for wavelength- and time-dependent selectivity of sugars such as fructose and ribose derivatives. One indicator is thus used to selectively distinguish structurally related sugar analytes.

Detecting Tumor Antigen-Specific T Cells via Interaction-Dependent Fucosyl-Biotinylation.

Project description:Re-activation and clonal expansion of tumor-specific antigen (TSA)-reactive T cells are critical to the success of checkpoint blockade and adoptive transfer of tumor-infiltrating lymphocyte (TIL)-based therapies. There are no reliable markers to specifically identify the repertoire of TSA-reactive T cells due to their heterogeneous composition. We introduce FucoID as a general platform to detect endogenous antigen-specific T cells for studying their biology. Through this interaction-dependent labeling approach, intratumoral TSA-reactive CD4+, CD8+ T cells, and TSA-suppressive CD4+ T cells can be detected and separated from bystander T cells based on their cell-surface enzymatic fucosyl-biotinylation. Compared to bystander TILs, TSA-reactive TILs possess a distinct T cell receptor (TCR) repertoire and unique gene features. Although exhibiting a dysfunctional phenotype, TSA-reactive CD8+ TILs possess substantial capabilities of proliferation and tumor-specific killing. Featuring genetic manipulation-free procedures and a quick turnover cycle, FucoID should have the potential of accelerating the pace of personalized cancer treatment.

Detecting DNA and RNA and Differentiating Single-Nucleotide Variations via Field-Effect Transistors.

Project description:We detect short oligonucleotides and distinguish between sequences that differ by a single base, using label-free, electronic field-effect transistors (FETs). Our sensing platform utilizes ultrathin-film indium oxide FETs chemically functionalized with single-stranded DNA (ssDNA). The ssDNA-functionalized semiconducting channels in FETs detect fully complementary DNA sequences and differentiate these sequences from those having different types and locations of single base-pair mismatches. Changes in charge associated with surface-bound ssDNA vs double-stranded DNA (dsDNA) alter FET channel conductance to enable detection due to differences in DNA duplex stability. We illustrate the capability of ssDNA-FETs to detect complementary RNA sequences and to distinguish from RNA sequences with single nucleotide variations. The development and implementation of electronic biosensors that rapidly and sensitively detect and differentiate oligonucleotides present new opportunities in the fields of disease diagnostics and precision medicine.

Poly-amido-saccharides: synthesis via anionic polymerization of a β-lactam sugar monomer.

Project description:Enantiopure poly-amido-saccharides (PASs) with a defined molecular weight and narrow dispersity are synthesized using an anionic ring-opening polymerization of a β-lactam sugar monomer. The PASs have a previously unreported main chain structure that is composed of pyranose rings linked through the 1- and 2-positions by an amide with α-stereochemistry. The monomer is synthesized in one-step from benzyl-protected D-glucal and polymerized using mild reaction conditions to give degrees of polymerization ranging from 25 to >120 in high yield. Computational modeling reveals how the monomer's structure and steric bulk affect the thermodynamics and kinetics of polymerization. Protected and deprotected polymers and model compounds are characterized using a variety of methods (NMR, GPC, IR, DLS, etc.). On the basis of circular dichroism, the deprotected polymer possesses a regular secondary structure in aqueous solution, which agrees favorably with the prediction of a helical structure using molecular modeling. Furthermore, we provide evidence suggesting that the polymers bind the lectin concanavalin A at the same site as natural carbohydrates, showing the potential of these polymers to mimic natural polysaccharides. PASs offer the advantages associated with synthetic polymers, such as greater control over structure and derivitization. At the same time, they preserve many of the structural features of natural polysaccharides, such as a stereochemically regular, rigid pyranose backbone, that make natural carbohydrate polymers important materials both for their unique properties and useful applications.

A Polymer-Based Indicator for Detecting Dexamethasone in Herbal Medicine Using Polymethylmethacrylate (PMMA).

Project description:Dexamethasone is a chemical drug that is usually added to herbal medicine because of its effects on pain relief, arthritis, anti-inflammation, etc. Chemical drugs should not be used in herbal medicine because uncontrolled consumption causes some side effects. A polymer-based indicator was developed to detect dexamethasone in herbal medicine samples in a fast and simple way compared to instrumental analysis. The indicator strips were made by mixing polymethylmethacrylate (PMMA) polymer with sulfuric acid (H2SO4) as a reagent. When reacting with dexamethasone, H2SO4 can cause the color to change into a specific light pink-purple color. Indicator strips were prepared with a composition of 5% PMMA in ethyl acetate:H2SO4 (9:1) by using the reagent blending method. The indicator strips showed a pink-purple color when they were applied to a positive herb containing dexamethasone. The indicator strips could selectively detect dexamethasone rather than other active substances that are often found in herbal medicine. These indicator strips could also detect dexamethasone with the smallest detection limit of 13.13 ppm, and they had a stability of up to 36 days. Detection was carried out in real samples to show the performance of the indicator strips. The result showed that of nine samples, five were confirmed to contain dexamethasone. These results showed a good agreement with the results of thin-layer chromatography (TLC) and high-performance liquid chromatography (HPLC). According to the result, these indicator strips provide a simple and applicable method for on-site analysis to detect dexamethasone in samples.

Cinnamil- and Quinoxaline-Derivative Indicator Dyes for Detecting Volatile Amines in Fish Spoilage.

Project description:Colorimetric indicators are versatile for applications such as intelligent packaging. By interacting with food, package headspace, and/or the ambient environment, color change in these indicators can be useful for reflecting the actual quality and/or monitoring distribution history (e.g., time and temperature) of food products. In this study, indicator dyes based on cinnamil and quinoxaline derivatives were synthesized using aroma compounds commonly present in food: diacetyl, benzaldehyde, p-tolualdehyde and p-anisaldehyde. The identities of cinnamil and quinoxaline derivatives were confirmed by Fourier transform infrared (FT-IR) spectroscopy, mass spectrometry (MS), 1H nuclear magnetic resonance (NMR) and 13C NMR analyses. Photophysical evaluation showed that the orange-colored cinnamil derivatives in dimethylsulfoxide (DMSO) turned to dark brownish coloration when exposed to strong alkalis. The cinnamil and acid-doped quinoxaline derivatives were sensitive to volatile amines commonly present during the spoilage in seafood. Quinoxaline derivatives doped by strong organic acid were effective as pH indicators for volatile amine detection, with lower detection limits than cinnamil. However, cinnamil exhibited more diverse color profiles than the quinoxaline indicators when exposed to ammonia, trimethylamine, triethylamine, dimethylamine, piperidine and hydrazine. Preliminary tests of acid-doped quinoxaline derivatives on fresh fish demonstrated their potential as freshness indicators in intelligent packaging applications.

Accurate Single-Molecule Indicator of Solvent Effects.

Project description:The study of the microscopic structure of solvents is of significant importance for deciphering the essential solvation in chemical reactions and biological processes. Yet conventional technologies, such as neutron diffraction, have an inherent averaging effect as they analyze a group of molecules. In this study, we report a method to analyze the microstructure and interaction in solvents from a single-molecule perspective. A single-molecule electrical nanocircuit is used to directly analyze the dynamic microscopic structure of solvents. Through a single-molecule model reaction, the heterogeneity or homogeneity of solvents is precisely detected at the molecular level. Both the thermodynamics and the kinetics of the model reaction demonstrate the microscopic heterogeneity of alcohol-water and alcohol-n-hexane solutions and the microscopic homogeneity of alcohol-carbon tetrachloride solutions. In addition, a real-time event spectroscopy has been developed to study the dynamic characteristics of the segregated phase and the internal intermolecular interaction in microheterogeneous solvents. The development of such a unique high-resolution indicator with single-molecule and single-event accuracy provides infinite opportunities to decipher solvent effects in-depth and optimizes chemical reactions and biological processes in solution.

Separation of saccharides using fullerene-bonded silica monolithic columns via π interactions in liquid chromatography.

Project description:We report on a potential method to separate sugars by using the specific interaction between fullerenes and saccharides in liquid chromatography (LC). Aromatic rings with high electron density are believed to interact strongly with saccharides due to CH-π and/or OH-π interactions. In this study, the fullerene-bonded columns were used to separate saccharides by LC under aqueous conditions. As a result, 2-aminobenzamide-labeled glucose homopolymer (Glcs) was effectively separated by both C60 and C70 columns in the range of Glc-1 to Glc-20 and high blood glucose level being retained in greater quantity. Furthermore, similar separations were identified by LC-mass spectrometry with non-labeled glucose homopolymers. Theoretical study based on molecular dynamics and DFT calculation demonstrated that a supramolecular complex of saccharide-fullerene was formed through CH-π and/or OH-π interactions, and that the interactions between saccharide and fullerene increase with the increase units of the saccharide. Additionally, the C60 column retained disaccharides containing maltose, trehalose, and sucrose. In this case, it was assumed that the retention rates were determined by the difference of the dipole moment in each saccharide. These results suggest that the dipole-induced dipole interaction was dominant, and that maltose-with the higher dipole moment-was more strongly retained compared to other disaccharides having lower dipole moment.
