Dichlorodiphenyltrichloroethane (DDT), DDT metabolites and pregnancy outcomes.

ABSTRACT: Organochlorine pesticides (OCPs) are persistent endocrine disruptors. OCPs cross the placenta; this prenatal exposure has been associated with adverse pregnancy outcomes. We investigated associations between prenatal exposure to OCPs and gestational age and birth weight in 600 infants born between 1960 and 1963. The primary OCP was 1,1,1-trichloro-2,2-bis(p-chlorophenyl)ethane (p,p'-DDT), its primary metabolite, 1,1'-dichloro-2,2'-bis(p-chlorophenyl)ethylene (p,p'-DDE) and the contaminant, 1,1,1-trichloro-2-(p-chlorophenyl)-2-(o-chlorophenyl)-ethane (o,p'-DDT). Regression analysis indicated that for each natural log unit increase in p,p'-DDT, birth weight increased by 274 g (95% CI: 122, 425) when controlling for p,p'-DDE and o,p'-DDT. At a given level of p,p'-DDT exposure, o,p'-DDT and p,p'-DDE were associated with decreased birth weight. p,p'-DDE was negatively associated with length of gestation, controlling for p,p'-DDT and o,p'-DDT. These findings suggest opposing associations between exposure to p,p'-DDT and p,p'-DDE and birth weight. We did not find evidence to support mediation by maternal thyroid hormone status nor that the association differed by sex.

Project description:DDT (dichlorodiphenyltrichloroethane) was used worldwide as an organochlorine insecticide to control agricultural pests and vectors of several insect-borne human diseases. It was banned in most industrialized countries; however, due to its persistence in the environment, DDT residues remain in environmental compartments, becoming long-term sources of exposure. To identify and select fungal species suitable for bioremediation of DDT-contaminated sites, soil samples were collected from DDT-contaminated agricultural soils in Poland, and 38 fungal taxa among 18 genera were isolated. Two of them, Trichoderma hamatum FBL 587 and Rhizopus arrhizus FBL 578, were tested for tolerance in the presence of 1-mg liter-1 DDT concentration by using two indices based on fungal growth rate and biomass production (the tolerance indices Rt:Rc and TI), showing a clear tolerance to DDT. The two selected strains were studied to evaluate catabolic versatility on 95 carbon sources with or without DDT by using the Phenotype MicroArray system and to investigate the induced oxidative stress responses. The two strains were able to use most of the substrates provided, resulting in both high metabolic versatility and ecological functionality in the use of carbon sources, despite the presence of DDT. The activation of specific metabolic responses with species-dependent antioxidant enzymes to cope with the induced chemical stress has been hypothesized, since the presence of DDT promoted a higher formation of reactive oxygen species in fungal cells than the controls. The tested fungi represent attractive potential candidates for bioremediation of DDT-contaminated soil and are worthy of further investigations.IMPORTANCE The spread and environmental accumulation of DDT over the years represent not only a threat to human health and ecological security but also a major challenge because of the complex chemical processes and technologies required for remediation. Saprotrophic fungi, isolated from contaminated sites, hold promise for their bioremediation potential toward toxic organic compounds, since they might provide an environment-friendly solution to contamination. Once we verified the high tolerance of autochthonous fungal strains to high concentrations of DDT, we showed how fungi from different phyla demonstrate a high metabolic versatility in the presence of DDT. The isolates showed the singular ability to keep their functionality, despite the DDT-induced production of reactive oxygen species.

Project description:BackgroundFluted pumpkin (Telfairia occidentalis) and amaranth (Amaranthus spp.) are common leafy vegetables produced and consumed in southwestern Nigeria. These vegetables attract insect pests which are controlled by pesticides that may have adverse impacts on human health.ObjectivesTo determine the levels of dichlorodiphenyltrichloroethane (DDT) and hexachlorocyclohexane (HCH) pesticide residues in the two vegetables and evaluate the potential health risks associated with their consumption.MethodsThe pesticide residue levels were quantitatively and qualitatively determined using a gas chromatograph coupled with electron capture detector. Health risk assessment were performed using estimated average daily intake and hazard indices for two weight categories: children (16.7 kg) and adults (60 kg).ResultsThe results showed that delta-HCH, DDT and methoxychlor were predominantly detected in the two vegetables from both farms and markets. In amaranth, the mean concentration of methoxychlor, DDT and delta-HCH were 4.590 ± 2.774 mg/kg (dry weight (dw)), 0.757 ± 0.457 mg/kg (dw) and 0.577 ± 0.390 mg/kg (dw), respectively, while fluted pumpkin levels were 6.223 ± 2.489 mg/kg dw (methoxychlor), 0.504 ± 0.056 mg/kg dw (delta-HCH) and 0.486 ± 0.123 mg/kg dw (DDT). The levels of HCH and DDT residues were generally above the United Kingdom/European Commission maximum residue limit. The analysis of health risk estimates for non-carcinogenic risk revealed that for both vegetables, the hazard quotient for p, p' DDT and methoxychlor was >1 for both children and adults. The health risk estimates for carcinogenic risk revealed that hazard indices values were >1 for children for both vegetables and alpha-HCH had a hazard index >1 for adults for amaranth. This means the residue exceeds acceptable standard and present potential risk to consumers of these vegetables.ConclusionsThe results obtained from the present study indicate that consumption of amaranth contaminated with alpha-HCH could pose a carcinogenic risk for adult consumers. The consumption of the two vegetables could pose both non-carcinogenic and carcinogenic health risks to children and adults. Therefore, there is need for strict enforcement of regulations on pesticide usage to minimize human health risks.Competing interestsThe authors declare no competing financial interests.
