The role of medial prefrontal cortex in memory and decision making.

ABSTRACT: Some have claimed that the medial prefrontal cortex (mPFC) mediates decision making. Others suggest mPFC is selectively involved in the retrieval of remote long-term memory. Yet others suggests mPFC supports memory and consolidation on time scales ranging from seconds to days. How can all these roles be reconciled? We propose that the function of the mPFC is to learn associations between context, locations, events, and corresponding adaptive responses, particularly emotional responses. Thus, the ubiquitous involvement of mPFC in both memory and decision making may be due to the fact that almost all such tasks entail the ability to recall the best action or emotional response to specific events in a particular place and time. An interaction between multiple memory systems may explain the changing importance of mPFC to different types of memories over time. In particular, mPFC likely relies on the hippocampus to support rapid learning and memory consolidation.

Disrupting the medial prefrontal cortex alters hippocampal sequences during deliberative decision making.

Project description:Current theories of deliberative decision making suggest that deliberative decisions arise from imagined simulations that require interactions between the prefrontal cortex and hippocampus. In rodent navigation experiments, hippocampal theta sequences advance from the location of the rat ahead to the subsequent goal. To examine the role of the medial prefrontal cortex (mPFC) on the hippocampus, we disrupted the mPFC with DREADDs (designer receptors exclusively activated by designer drugs). Using the Restaurant Row foraging task, we found that mPFC disruption resulted in decreased vicarious trial and error behavior, reduced the number of theta sequences, and impaired theta sequences in hippocampus. mPFC disruption led to larger changes in the initiation of the hippocampal theta sequences that represent the current location of the rat rather than to the later portions that represent the future outcomes. These data suggest that the mPFC likely provides an important component to the initiation of deliberative sequences and provides support for an episodic-future thinking, working memory interpretation of deliberation. NEW & NOTEWORTHY The medial prefrontal cortex (mPFC) and hippocampus interact during deliberative decision making. Disruption of the mPFC impaired hippocampal processes, including the local and nonlocal representations of space along each theta cycle and the initiation of hippocampal theta sequences, while sparing place cell firing characteristics and phase precession. mPFC disruption reduced the deliberative behavioral process vicarious trial and error and improved economic behaviors on this task.

Distinct roles of rodent orbitofrontal and medial prefrontal cortex in decision making.

Project description:We investigated how different subregions of rodent prefrontal cortex contribute to value-based decision making, by comparing neural signals related to animal's choice, its outcome, and action value in orbitofrontal cortex (OFC) and medial prefrontal cortex (mPFC) of rats performing a dynamic two-armed bandit task. Neural signals for upcoming action selection arose in the mPFC, including the anterior cingulate cortex, only immediately before the behavioral manifestation of animal's choice, suggesting that rodent prefrontal cortex is not involved in advanced action planning. Both OFC and mPFC conveyed signals related to the animal's past choices and their outcomes over multiple trials, but neural signals for chosen value and reward prediction error were more prevalent in the OFC. Our results suggest that rodent OFC and mPFC serve distinct roles in value-based decision making and that the OFC plays a prominent role in updating the values of outcomes expected from chosen actions.

Role of the medial prefrontal cortex in the effects of rapid acting antidepressants on decision-making biases in rodents.

Project description:Major depressive disorder is a significant and costly cause of global disability. Until the discovery of the rapid acting antidepressant (RAAD) effects of ketamine, treatments were limited to drugs that have delayed clinical benefits. The mechanism of action of ketamine is currently unclear but one hypothesis is that it may involve neuropsychological effects mediated through modulation of affective biases (where cognitive processes such as learning and memory and decision-making are modified by emotional state). Previous work has shown that affective biases in a rodent decision-making task are differentially altered by ketamine, compared to conventional, delayed onset antidepressants. This study sought to further investigate these effects by comparing ketamine with other NMDA antagonists using this decision-making task. We also investigated the subtype selective GluN2B antagonist, CP-101,606 and muscarinic antagonist scopolamine which have both been shown to have RAAD effects. Both CP-101,606 and scopolamine induced similar positive biases in decision-making to ketamine, but the same effects were not seen with other NMDA antagonists. Using targeted medial prefrontal cortex (mPFC) infusions, these effects were localised to the mPFC. In contrast, the GABAA agonist, muscimol, induced general disruptions to behaviour. These data suggest that ketamine and other RAADs mediate a specific effect on affective bias which involves the mPFC. Non-ketamine NMDA antagonists lacked efficacy and we also found that temporary inactivation of the mPFC did not fully recapitulate the effects of ketamine, suggesting a specific mechanism.

Adenosine A2A Receptors in the Rat Prelimbic Medial Prefrontal Cortex Control Delay-Based Cost-Benefit Decision Making.

Project description:Adenosine A2A receptors (A2ARs) were recently described to control synaptic plasticity and network activity in the prefrontal cortex (PFC). We now probed the role of these PFC A2AR by evaluating the behavioral performance (locomotor activity, anxiety-related behavior, cost-benefit decision making and working memory) of rats upon downregulation of A2AR selectively in the prelimbic medial PFC (PLmPFC) via viral small hairpin RNA targeting the A2AR (shA2AR). The most evident alteration observed in shA2AR-treated rats, when compared to sh-control (shCTRL)-treated rats, was a decrease in the choice of the large reward upon an imposed delay of 15 s assessed in a T-maze-based cost-benefit decision-making paradigm, suggestive of impulsive decision making. Spontaneous locomotion in the open field was not altered, suggesting no changes in exploratory behavior. Furthermore, rats treated with shA2AR in the PLmPFC also displayed a tendency for higher anxiety levels in the elevated plus maze (less entries in the open arms), but not in the open field test (time spent in the center was not affected). Finally, working memory performance was not significantly altered, as revealed by the spontaneous alternation in the Y-maze test and the latency to reach the platform in the repeated trial Morris water maze. These findings constitute the first direct demonstration of a role of PFC A2AR in the control of behavior in physiological conditions, showing their major contribution for the control of delay-based cost-benefit decisions.

Dorsolateral and medial prefrontal cortex mediate the influence of incidental priming on economic decision making in obesity.

Project description:Obese individuals discount future rewards to a higher degree than lean individuals, which is generally considered disadvantageous. Moreover, their decisions are altered more easily by decision-irrelevant cues. Here, we investigated neural correlates of this phenomenon using functional MRI. We tested 30 lean and 26 obese human subjects on a primed delay discounting paradigm using gustatory and visual cues of positive, neutral and negative valence to bias their intertemporal preferences. We hypothesised that activation differences in reward-related and behavioural control areas, and changes in connectivity between these areas, would reflect the effect of these cues. Here, obese subjects were more susceptible to priming with negative gustatory cues towards delayed choices as opposed to lean subjects. This was related to lower activity in the left dorsolateral prefrontal cortex during priming. Modulation of functional connectivity between the dlPFC and the ventromedial PFC by the behavioural priming effect correlated negatively with BMI. This might indicate that default goals of obese individuals were different from those of lean participants, as the dlPFC has been suggested to be involved in internal goal pursuit. The present results further our understanding of the role of the PFC in decision-making and might inform future weight-management approaches based on non-invasive brain stimulation.

Dissociable roles for the ventral and dorsal medial prefrontal cortex in cue-guided risk/reward decision making.

Project description:Converging evidence from studies with animals and humans have implicated separate regions of the medial prefrontal cortex (mPFC) corresponding to the anterior cingulate cortex (ACC), in mediating different aspects of reward-related decisions involving uncertainty or risk. However, the dissociable contributions of subregions of the ACC remain unclear, as discrepancies exist between human neuroimaging findings and preclinical rodent studies. To clarify how ventral vs. dorsal regions of the mPFC contribute to risk/reward decision making, the present study assessed the effects of inactivation of different subregions on performance of a "Blackjack task" that measured cue-guided decision making and shares similarities with paradigms used with humans. Male, Long-Evans rats were well-trained to choose between a Small/Certain reward vs a Large/Risky reward delivered with variable probabilities (i.e., good vs. poor-odds, 50% vs. 12.5%). The odds of obtaining the larger reward was signaled by auditory cues at the start of each trial. Inactivation of the ventral, infralimbic region of the mPFC increased risky choice selectively when the odds of winning were poor. By contrast, inactivation of the prelimbic and anterior cingulate regions of the dorsal mPFC led to suboptimal reductions in risky choice on good-odds trials. The effects of prelimbic vs anterior cingulate inactivations were associated with context-dependent alterations in reward vs negative feedback, respectively. These results further clarify the distinct yet complementary manners in which separate ACC regions promote optimal risk/reward decision making and complement neuroimaging findings that activity in human ventral vs dorsal ACC promotes risk aversion or risky choices.

Distinct encoding of decision confidence in human medial prefrontal cortex.

Project description:Our confidence in a choice and the evidence pertaining to a choice appear to be inseparable. However, an emerging computational consensus holds that the brain should maintain separate estimates of these quantities for adaptive behavioral control. We have devised a psychophysical task to decouple confidence in a perceptual decision from both the reliability of sensory evidence and the relation of such evidence with respect to a choice boundary. Using human fMRI, we found that an area in the medial prefrontal cortex, the perigenual anterior cingulate cortex (pgACC), tracked expected performance, an aggregate signature of decision confidence, whereas neural areas previously proposed to encode decision confidence instead tracked sensory reliability (posterior parietal cortex and ventral striatum) or boundary distance (presupplementary motor area). Supporting that information encoded by pgACC is central to a subjective sense of decision confidence, we show that pgACC activity does not simply covary with expected performance, but is also linked to within-subject and between-subject variation in explicit confidence estimates. Our study is consistent with the proposal that the brain maintains choice-dependent and choice-independent estimates of certainty, and sheds light on why dysfunctional confidence often emerges following prefrontal lesions and/or degeneration.

Hunger or thirst state uncertainty is resolved by outcome evaluation in medial prefrontal cortex to guide decision-making.

Project description:Physiological need states direct decision-making toward re-establishing homeostasis. Using a two-alternative forced choice task for mice that models elements of human decisions, we found that varying hunger and thirst states caused need-inappropriate choices, such as food seeking when thirsty. These results show limits on interoceptive knowledge of hunger and thirst states to guide decision-making. Instead, need states were identified after food and water consumption by outcome evaluation, which depended on the medial prefrontal cortex.

Prefrontal cortex exhibits multidimensional dynamic encoding during decision-making.

Project description:Recent work has suggested that the prefrontal cortex (PFC) plays a key role in context-dependent perceptual decision-making. In this study, we addressed that role using a new method for identifying task-relevant dimensions of neural population activity. Specifically, we show that the PFC has a multidimensional code for context, decisions and both relevant and irrelevant sensory information. Moreover, these representations evolve in time, with an early linear accumulation phase followed by a phase with rotational dynamics. We identify the dimensions of neural activity associated with these phases and show that they do not arise from distinct populations but from a single population with broad tuning characteristics. Finally, we use model-based decoding to show that the transition from linear to rotational dynamics coincides with a plateau in decoding accuracy, revealing that rotational dynamics in the PFC preserve sensory choice information for the duration of the stimulus integration period.

Making sense: Dopamine activates conscious self-monitoring through medial prefrontal cortex.

Project description:When experiences become meaningful to the self, they are linked to synchronous activity in a paralimbic network of self-awareness and dopaminergic activity. This network includes medial prefrontal and medial parietal/posterior cingulate cortices, where transcranial magnetic stimulation may transiently impair self-awareness. Conversely, we hypothesize that dopaminergic stimulation may improve self-awareness and metacognition (i.e., the ability of the brain to consciously monitor its own cognitive processes). Here, we demonstrate improved noetic (conscious) metacognition by oral administration of 100 mg dopamine in minimal self-awareness. In a separate experiment with extended self-awareness dopamine improved the retrieval accuracy of memories of self-judgment (autonoetic, i.e., explicitly self-conscious) metacognition. Concomitantly, magnetoencephalography (MEG) showed increased amplitudes of oscillations (power) preferentially in the medial prefrontal cortex. Given that electromagnetic activity in this region is instrumental in self-awareness, this explains the specific effect of dopamine on explicit self-awareness and autonoetic metacognition.
