Lanthanite-(Nd), Nd2(CO3)3·8H2O.

ABSTRACT: Lanthanite-(Nd), ideally Nd2(CO3)3·8H2O [dineodymium(III) tricarbonate octa-hydrate], is a member of the lanthanite mineral group characterized by the general formula REE2(CO3)3·8H2O, where REE is a 10-coordinated rare earth element. Based on single-crystal X-ray diffraction of a natural sample from Mitsukoshi, Hizen-cho, Karatsu City, Saga Prefecture, Japan, this study presents the first structure determination of lanthanite-(Nd). Its structure is very similar to that of other members of the lanthanite group. It is composed of infinite sheets made up of corner- and edge-sharing of two NdO10-polyhedra (both with site symmetry ..2) and two carbonate triangles (site symmetries ..2 and 1) parallel to the ab plane, and stacked perpendicular to c. These layers are linked to one another only through hydrogen bonding involving the water mol-ecules.

Project description:The crystal structure of the title compound comprises a tetracarbonatozincate(II) anion, [Zn(CO3)4]6−, with point-group symmetry 2 where the ZnII atom is surrounded in a distorted tetrahedral manner by four monodentate carbonate groups. The crystal structure of K6[Zn(CO3)4], hexapotassium tetracarbonatozincate(II), comprises four unique potassium cations (two located on a general position, and two on the twofold rotation axis of the space group C2/c) and a [Zn(CO3)4]6− anion. The ZnII atom of the latter is located on the twofold rotation axis and is surrounded in a slightly distorted tetrahedral manner by two pairs of monodentately binding carbonate groups, with Zn—O distances of 1.9554 (18) and 1.9839 (18) Å. Both carbonate groups exhibit a slight deviation from planarity, with the C atom being shifted by 0.008 (2) and 0.006 (3) Å, respectively, from the plane of the three O atoms. The coordination numbers of the potassium cations range from 6 to 8, using a threshold of 3.0 Å for K—O bonding interactions being significant. In the crystal structure, [KOx] polyhedra and [Zn(CO3)4]6− groups share O atoms to build up the framework structure.

Transcriptome analysis from Nd2-mutant and control fibroblasts

Project description:Background: Mutations of mitochondrial (mt)DNA cause a variety of human diseases and are also implicated in ageing processes. Results: Primary fibroblasts of the conplastic mouse strain C57BL/6J-mtALR/LTJ with a mutation at position nt4738 resulting in a single nucleotide exchange (leucine to methionine) in the mitochondrial NADH dehydrogenase subunit 2 (Nd2) gene of the respiratory chain show higher enzyme activity and ATP production and lower ROS production than control fibroblasts. Furthermore, Nd2-mutant fibroblasts show a higher proliferation rate and a reduction of senescence markers. Transcriptome analysis reveals members of the p38MAPK pathway as being significantly downregulated in Nd2-mutant mice as compared with controls. In line with this, inhibition of p38MAPK with SB203580 enhanced the proliferation and reduced cytokine secretion in senescent Nd2-mutant fibroblasts. Conclusion: Taken together, we report Nd2 as a new mitochondrial gene with age-protective function in mice, and possibly in humans, which interferes with age-related signaling pathways.

Strong localization of oxidized Co3+ state in cobalt-hexacyanoferrate.

Project description:Secondary batteries are important energy storage devices for a mobile equipment, an electric car, and a large-scale energy storage. Nevertheless, variation of the local electronic state of the battery materials in the charge (or oxidization) process are still unclear. Here, we investigated the local electronic state of cobalt-hexacyanoferrate (Na x Co[Fe(CN)6]0.9), by means of resonant inelastic X-ray scattering (RIXS) with high energy resolution (~100?meV). The L-edge RIXS is one of the most powerful spectroscopic technique with element- and valence-selectivity. We found that the local electronic state around Co2+ in the partially-charged Na1.1Co2+0.5Co3+0.5[Fe2+(CN)6]0.9 film (x?=?1.1) is the same as that of the discharged Na1.6Co2+[Fe2+(CN)6]0.9 film (x?=?1.6) within the energy resolution, indicating that the local electronic state around Co2+ is invariant against the partial oxidization. In addition, the local electronic state around the oxidized Co3+ is essentially the same as that of the fully-charged film Co3+[Fe2+(CN)6]0.3[Fe3+(CN)6]0.6 (x?=?0.0) film. Such a strong localization of the oxidized Co3+ state is advantageous for the reversibility of the redox process, since the localization reduces extra reaction within the materials and resultant deterioration.

Mitochondrial miR-762 regulates apoptosis and myocardial infarction by impairing ND2.

Project description:Mitochondrial dysfunction plays a major role in the pathogenesis of cardiovascular diseases. MicroRNAs (miRNAs) are small RNAs that act as negative regulators of gene expression, but how miRNAs affect mitochondrial function in the heart is unclear. Using a miRNA microarray assay, we found that miR-762 predominantly translocated in the mitochondria and was significantly upregulated upon anoxia/reoxygenation (A/R) treatment. Knockdown of endogenous miR-762 significantly attenuated the decrease in intracellular ATP levels, the increase in ROS levels, the decrease in mitochondrial complex I enzyme activity and the increase in apoptotic cell death in cardiomyocytes, which was induced by A/R treatment. In addition, knockdown of miR-762 ameliorated myocardial ischemia/reperfusion (I/R) injury in mice. Mechanistically, we showed that enforced expression of miR-762 dramatically decreased the protein levels of endogenous NADH dehydrogenase subunit 2 (ND2) but had no effect on the transcript levels of ND2. The luciferase reporter assay showed that miR-762 bound to the coding sequence of ND2. In addition, knockdown of endogenous ND2 significantly decreased intracellular ATP levels, increased ROS levels, reduced mitochondrial complex I enzyme activity and increased apoptotic cell death in cardiomyocytes, which was induced by A/R treatment. Furthermore, we found that the inhibitory effect of miR-762 downregulation was attenuated by ND2 knockdown. Thus, our findings suggest that miR-762 participates in the regulation of mitochondrial function and cardiomyocyte apoptosis by ND2, a core assembly subunit of mitochondrial complex I. Our results revealed that mitochondrial miR-762, as a new player in mitochondrial dysfunction, may provide a new therapeutic target for myocardial infarction.

MITRAC15/COA1 promotes mitochondrial translation in a ND2 ribosome-nascent chain complex.

Project description:The mitochondrial genome encodes for thirteen core subunits of the oxidative phosphorylation system. These proteins assemble with imported proteins in a modular manner into stoichiometric enzyme complexes. Assembly factors assist in these biogenesis processes by providing co-factors or stabilizing transient assembly stages. However, how expression of the mitochondrial-encoded subunits is regulated to match the availability of nuclear-encoded subunits is still unresolved. Here, we address the function of MITRAC15/COA1, a protein that participates in complex I biogenesis and complex IV biogenesis. Our analyses of a MITRAC15 knockout mutant reveal that MITRAC15 is required for translation of the mitochondrial-encoded complex I subunit ND2. We find that MITRAC15 is a constituent of a ribosome-nascent chain complex during ND2 translation. Chemical crosslinking analyses demonstrate that binding of the ND2-specific assembly factor ACAD9 to the ND2 polypeptide occurs at the C-terminus and thus downstream of MITRAC15. Our analyses demonstrate that expression of the founder subunit ND2 of complex I undergoes regulation. Moreover, a ribosome-nascent chain complex with MITRAC15 is at the heart of this process.

An evolutionary switch in ND2 enables Src kinase regulation of NMDA receptors.

Project description:The non-receptor tyrosine kinase Src is a key signalling hub for upregulating the function of N-methyl D-aspartate receptors (NMDARs). Src is anchored within the NMDAR complex via NADH dehydrogenase subunit 2 (ND2), a mitochondrially encoded adaptor protein. The interacting regions between Src and ND2 have been broadly identified, but the interaction between ND2 and the NMDAR has remained elusive. Here we generate a homology model of ND2 and dock it onto the NMDAR via the transmembrane domain of GluN1. This interaction is enabled by the evolutionary loss of three helices in bilaterian ND2 proteins compared to their ancestral homologues. We experimentally validate our model and demonstrate that blocking this interaction with an ND2 fragment identified in our experimental studies prevents Src-mediated upregulation of NMDAR currents in neurons. Our findings establish the mode of interaction between an NMDAR accessory protein with one of the core subunits of the receptor.

Nd(2)(WO(4))(3).

Project description:The title compound, dineodymium(III) tris-[tungstate(VI)], is a member of the Eu(2)(WO(4))(3) structure family and crystallizes isotypically with other rare earth tungstates and molybdates of this formula type. The structure is a derivative of the scheelite (CaWO(4)) structure and can be considered as an ordered defect variant with a threefold scheelite supercell and one rare earth (RE) site unoccupied. The Nd(3+) cations are coordinated by eight O atoms in form of a distorted bicapped trigonal prism. The two unique W cations are tetra-hedrally surrounded by O atoms. One WO(4) tetra-hedron has 2 symmetry and is relatively undistorted whereas the other tetra-hedron differs considerably from an ideal geometry. This is caused by an additional remote O atom at a distance of 2.149?(4)?Å. The resulting WO(4 + 1) polyhedra form W(2)O(8) dimers through edge-sharing. Together with the WO(4) and NdO(8) units, the three-dimensional set-up is accomplished.
