Design plans for an inexpensive tail flick analgesia meter.

ABSTRACT: While the pedagogical benefits of incorporating inquiry driven labs into an undergraduate curriculum are well established, often the prohibitive costs of providing equipment for such labs limits the types of experiences that can be offered. For example, the lab portion of Advanced Neuroscience at Centenary College of Louisiana consists of a semester-long research project developed by the students. Frequently, these junior- and senior-level students generate interesting research questions that must be culled or scaled back simply due to a lack of appropriate equipment. In the most recent iteration of the class, the students wanted to examine analgesia using the tail flick test, a measure of spinal nociception. In this test a rodent subject is restrained; its tail is exposed to a heat source; and the latency to flick its tail away from the noxious stimuli is recorded. As commercial devices were far beyond the lab budget, we sought to develop an inexpensive tail flick analgesia meter that was easy to use and generated reliable data. The prototype device was tested by students in the above-mentioned class and was found to consistently produce reliable data in agreement with the literature. Here we present plans for a tail flick analgesia meter that can be constructed for $50-75, roughly 100 times cheaper than commercial devices.

Project description:Plant growth chambers produce controlled environments, which are crucial in making reproducible observations in experimental plant biology research. Commercial plant growth chambers can provide precise controls of environmental parameters, such as temperature, humidity, and light cycle, and the capability via complex programming to regulate these environmental parameters. But they are expensive. The high cost of maintaining a controlled growth environment is often a limiting factor when determining experiment size and feasibility. To overcome the limitation of commercial growth chambers, we designed and constructed an inexpensive plant growth chamber with consumer products for a material cost of $2,300. For a comparable growth space, a commercial plant growth chamber could cost $40,000 or more. Our plant growth chamber had outside dimensions of 1.5 m (W) x 1.8 m (D) x 2 m (H), providing a total growth area of 4.5 m2 with 40-cm high clearance. The dimensions of the growth area and height can be flexibly changed. Fluorescent lights with large reflectors provided a relatively spatially uniform photosynthetically active radiation intensity of 140-250 ?moles/m2/sec. A portable air conditioner provided an ample cooling capacity, and a cooling water mister acted as a powerful humidifier. Temperature, relative humidity, and light cycle inside the chamber were controlled via a z-wave home automation system, which allowed the environmental parameters to be monitored and programmed through the internet. In our setting, the temperature was tightly controlled: 22.2°C±0.8°C. The one-hour average relative humidity was maintained at 75%±7% with short spikes up to ±15%. Using the interaction between Arabidopsis and one of its bacterial pathogens as a test experimental system, we demonstrate that experimental results produced in our chamber were highly comparable to those obtained in a commercial growth chamber. In summary, our design of an inexpensive plant growth chamber will tremendously increase research opportunities in experimental plant biology.

Project description:BackgroundThe objective of this study was to compare the changes in the electroencephalogram (EEG) in response to noxious stimuli with tail flick and hot plate responses of rats administered opiorphin.MethodsFemale Sprague -Dawley rats (n = 8 per group) randomly received intravenous (IV) injection of morphine (1 mg/kg,) or opiorphin (2 mg/kg,) or saline (0.5 ml,) in each of the three testing methods (EEG, tail flick and hot plate). Each type of test (n = 24 per test) was conducted in different population of rats on separate occasions. The tail flick and hot plate latencies were recorded until 5 min after test drug administration to conscious rats. The EEG was recorded in anaesthetised rats subjected to noxious thermal and electrical stimuli after test drug administration. At the end of 5 min in each of the testing methods rats were administered naloxone subcutaneously (SC) (1 mg/kg) and the test procedure was repeated.ResultsThere was no significant increase in the median frequency and spectral edge frequency (F50 & F95) of EEG, indicators of nociception, of morphine and opiorphin groups after noxious stimulation. Noxious stimuli caused a significant increase in both F50 and F95 of the saline group. An injection of naloxone significantly increased the F50, thus blocking the action of both opiorphin and morphine. There was a significant increase in the tail flick latency after administration of opiorphin and morphine as compared to the baseline values. Rats of morphine group spent significantly longer on the hot plate when compared to those of the opiorphin and saline groups. There was no significant difference in the hot plate latencies of opiorphin and saline groups.ConclusionThe results of this study suggest that the analgesic effect of opiorphin occurs at the spinal level and it is not as effective as morphine at supraspinal level. It may be due to rapid degradation of opiorphin or limited ability of opiorphin to cross the blood brain barrier or a higher dose of opiorphin is required for its action in the brain. Pharmacokinetic/pharmacodynamics studies along with in vivo penetration of opiorphin in the cerebrospinal fluid are required for further evaluation of opiorphin analgesia.

Project description:BackgroundPhysical inactivity is a global issue that affects people's health and productivity. With the advancement of mobile technologies, many apps have been developed to facilitate health self-management. However, few studies have examined the effectiveness of these mobile health (mHealth) apps in motivating exercise adherence.ObjectiveThis study aims to demonstrate the enhanced action design research (ADR) process and improve the design of mHealth apps for exercise self-management. Specifically, we investigate whether sending motivational messages improves adherence to exercise plans, whether the motivational effect is affected by personality, the impact of message type and repetition, and the process of involving a field experiment in the design process and learning new design principles from the results.MethodsThis formative research was conducted by proposing an enhanced ADR process. We incorporated a field experiment into the process to iteratively refine and evaluate the design until it converges into a final mHealth app. We used the Apple ResearchKit to develop the mHealth app and promoted it via trainers at their gyms. We targeted users who used the app for at least two months. Participants were randomly assigned to 1 of the 12 groups in a 2×3×2 factorial design and remained blinded to the assigned intervention. The groups were defined based on personality type (thinking or feeling), message type (emotional, logical, or none), and repetition (none or once). Participants with different personality types received tailored and repeated messages. Finally, we used the self-reported completion rate to measure participants' adherence level to exercise plans. By analyzing users' usage patterns, we could verify, correct, and enhance the mHealth app design principles.ResultsIn total, 160 users downloaded the app, and 89 active participants remained during the 2-month period. The results suggest a significant main effect of personality type and repetition and a significant interaction effect between personality type and repetition. The adherence rate of people with feeling personality types was 18.15% higher than that of people with thinking types. Emotional messages were more effective than logical messages in motivating exercise adherence. Although people received repeated messages, they were more likely to adhere to exercise plans. With repeated reminders, the adherence rates of people with thinking personality types were significantly improved by 27.34% (P<.001).ConclusionsThis study contributes to the literature on mHealth apps. By incorporating a field experiment into the ADR process, we demonstrate the benefit of combining design science and field experiments. This study also contributes to the research on mHealth apps. The principles learned from this study can be applied to improve the effectiveness of mHealth apps. The app design can be considered a foundation for the development of more advanced apps for specific diseases, such as diabetes and asthma, in future research.
