Raman spectroscopy as a tool for monitoring mesoscale continuous-flow organic synthesis: Equipment interface and assessment in four medicinally-relevant reactions.

ABSTRACT: An apparatus is reported for real-time Raman monitoring of reactions performed using continuous-flow processing. Its capability is assessed by studying four reactions, all involving formation of products bearing ?,?-unsaturated carbonyl moieties; synthesis of 3-acetylcoumarin, Knoevenagel and Claisen-Schmidt condensations, and a Biginelli reaction. In each case it is possible to monitor the reactions and also in one case, by means of a calibration curve, determine product conversion from Raman spectral data as corroborated by data obtained using NMR spectroscopy.

Project description:This article describes the development and manufacturing of lab equipment, which is needed for the use in flow chemistry. We developed a rack of four syringe pumps controlled by one Arduino computer, which can be manufactured with a commonly available 3D printer and readily available parts. Also, we printed various flow reactor cells, which are fully customizable for each individual reaction. With this equipment we performed some multistep glycosylation reactions, where multiple 3D-printed flow reactors were used in series.

C-N Bond Formation by Consecutive Continuous-Flow Reductions towards A Medicinally Relevant Piperazine Derivative.

Project description:A new, continuous-flow consecutive reduction method was developed for the C-N bond formation in the synthesis of the key intermediate of the antipsychotic drug cariprazine. The two-step procedure consists of a DIBAL-H mediated selective ester reduction conducted in a novel, miniature alternating diameter reactor, followed by reductive amination using catalytic hydrogenation on 5% Pt/C. The connection of the optimized modules was accomplished using an at-line extraction to prevent precipitation of the aluminum salt byproducts.

Transition across a sharp interface: Data from Raman and Brillouin imaging spectroscopy.

Project description:Brillouin and Raman imaging are powerful techniques for the investigation of complex materials and they are widely used in material science and biophysics [1], [2], [3], [4], [5], [6], [7]. When dealing with microstructures, the results interpretation requires an accurate understanding of the interaction processes in presence of acoustic and chemical boundaries between different materials [8], [9], [10], [11], [12], [13], [14], [15]. The data here reported are obtained while scanning with sub-micron resolution the sharp interfaces between vitreous-SiO2/Water and Polyethylene (PET)/Glycerol. Molecular and acoustic vibrations were observed by means of a recently developed micro-spectrometer, which acquires simultaneously Raman and Brillouin spectra on the same point with high spatial and spectral resolution [3]. Two external optic configurations were adopted in order to evidence the dependency of the measurements on the optical scattering volume. The evolution of the detected phonon modes, propagating and not propagating, is obtained by a direct observation of the raw data for the two interfaces, which present different acoustic mismatch. These experimental records can be exploited by researchers employing Raman and Brillouin imaging to discuss the resolution limit of the techniques and to compare the effect of different experimental set-ups. Moreover, thanks to their high spectral resolution they can be useful to researchers working on acoustic phonon transport at interfaces to model the dependency of transmission of long wavelength phonons on the acoustic mismatch.

Revealing unconventional host-guest complexation at nanostructured interface by surface-enhanced Raman spectroscopy.

Project description:Interfacial host-guest complexation offers a versatile way to functionalize nanomaterials. However, the complicated interfacial environment and trace amounts of components present at the interface make the study of interfacial complexation very difficult. Herein, taking the advantages of near-single-molecule level sensitivity and molecular fingerprint of surface-enhanced Raman spectroscopy (SERS), we reveal that a cooperative effect between cucurbit[7]uril (CB[7]) and methyl viologen (MV2+2I-) in aggregating Au NPs originates from the cooperative adsorption of halide counter anions I-, MV2+, and CB[7] on Au NPs surface. Moreover, similar SERS peak shifts in the control experiments using CB[n]s but with smaller cavity sizes suggested the occurrence of the same guest complexations among CB[5], CB[6], and CB[7] with MV2+. Hence, an unconventional exclusive complexation model is proposed between CB[7] and MV2+ on the surface of Au NPs, distinct from the well-known 1:1 inclusion complexation model in aqueous solutions. In summary, new insights into the fundamental understanding of host-guest interactions at nanostructured interfaces were obtained by SERS, which might be useful for applications related to host-guest chemistry in engineered nanomaterials.

Real-Time Monitoring of Enzyme-Catalysed Reactions using Deep UV Resonance Raman Spectroscopy.

Project description:For enzyme-catalysed biotransformations, continuous in situ detection methods minimise the need for sample manipulation, ultimately leading to more accurate real-time kinetic determinations of substrate(s) and product(s). We have established for the first time an on-line, real-time quantitative approach to monitor simultaneously multiple biotransformations based on UV resonance Raman (UVRR) spectroscopy. To exemplify the generality and versatility of this approach, multiple substrates and enzyme systems were used involving nitrile hydratase (NHase) and xanthine oxidase (XO), both of which are of industrial and biological significance, and incorporate multistep enzymatic conversions. Multivariate data analysis of the UVRR spectra, involving multivariate curve resolution-alternating least squares (MCR-ALS), was employed to effect absolute quantification of substrate(s) and product(s); repeated benchmarking of UVRR combined with MCR-ALS by HPLC confirmed excellent reproducibility.

In?Situ Shell-Isolated Nanoparticle-Enhanced Raman Spectroscopy of Nickel-Catalyzed Hydrogenation Reactions.

Project description:Synthesis methods to prepare lower transition metal catalysts and specifically Ni for Shell-Isolated Nanoparticle-Enhanced Raman Spectroscopy (SHINERS) are explored. Impregnation, colloidal deposition, and spark ablation have been investigated as suitable synthesis routes to prepare SHINERS-active Ni/Au@SiO2 catalyst/Shell-Isolated Nanoparticles (SHINs). Ni precursors are confirmed to be notoriously difficult to reduce and the temperatures required are generally harsh enough to destroy SHINs, rendering SHINERS experiments on Ni infeasible using this approach. For colloidally synthesized Ni nanoparticles deposited on Au@SiO2 SHINs, stabilizing ligands first need to be removed before application is possible in catalysis. The required procedure results in transformation of the metallic Ni core to a fully oxidized metal nanoparticle, again too challenging to reduce at temperatures still compatible with SHINs. Finally, by use of spark ablation we were able to prepare metallic Ni catalysts directly on Au@SiO2 SHINs deposited on a Si wafer. These Ni/Au@SiO2 catalyst/SHINs were subsequently successfully probed with several molecules (i.?e. CO and acetylene) of interest for heterogeneous catalysis, and we show that they could be used to study the in?situ hydrogenation of acetylene. We observe the interaction of acetylene with the Ni surface. This study further illustrates the true potential of SHINERS by opening the door to studying industrially relevant reactions under in?situ or operando reaction conditions.

Monitoring Solid-Phase Reactions in Self-Assembled Monolayers by Surface-Enhanced Raman Spectroscopy.

Project description:Nanopatterned surfaces enhance incident electromagnetic radiation and thereby enable the detection and characterization of self-assembled monolayers (SAMs), for instance in surface-enhanced Raman spectroscopy (SERS). Herein, Au nanohole arrays, developed and characterized as SERS substrates, are exemplarily used for monitoring a solid-phase deprotection and a subsequent copper(I)-catalyzed azide-alkyne cycloaddition "click" reaction, performed directly on the corresponding SAMs. The SERS substrate was found to be highly reliable in terms of signal reproducibility and chemical stability. Furthermore, the intermediates and the product of the solid-phase synthesis were identified by SERS. The spectra of the immobilized compounds showed minor differences compared to spectra of the microcrystalline solids. With its uniform SERS signals and the high chemical stability, the platform paves the way for monitoring molecular manipulations in surface functionalization applications.

In-situ plasmon-driven chemical reactions revealed by high vacuum tip-enhanced Raman spectroscopy.

Project description:With strong surface plasmons excited at the metallic tip, tip-enhanced Raman spectroscopy (TERS) has both high spectroscopic sensitivity and high spatial resolution, and is becoming an essential tool for chemical analysis. It is a great challenge to combine TERS with a high vacuum system due to the poor optical collection efficiency. We used our innovatively designed home-built high vacuum TERS (HV-TERS) to investigate the plasmon-driven in-situ chemical reaction of 4-nitrobenzenethiol dimerizing to dimercaptoazobenzene. The chemical reactions can be controlled by the plasmon intensity, which in turn can be controlled by the incident laser intensity, tunneling current and bias voltage. The temperature of such a chemical reaction can also be obtained by the clearly observed Stokes and Anti-Stokes HV-TERS peaks. Our findings offer a new way to design a highly efficient HV-TERS system and its applications to chemical catalysis and synthesis of molecules, and significantly extend the studies of chemical reactions.

Continuous flow-resonance Raman spectroscopy of an intermediate redox state of cytochrome C.

Project description:An intermediate redox state of cytochrome c at alkaline pH, generated upon rapid reduction by sodium dithionite, has been observed by resonance Raman (RR) spectroscopy in combination with the continuous flow technique. The RR spectrum of the intermediate state is reported for excitation both in the (alpha, beta) and the Soret optical absorption band. The spectra of the intermediate state are more like those of the stable reduced form than those of the stable oxidized form. For excitation of 514.5 nm, the most prominent indication of an intermediate state is the wave-number shift of one RR band from 1,562 cm-1 in the stable oxidized state through 1,535 cm-1 in the intermediate state to 1,544 cm-1 in the stable reduced state. For excitation at 413.1 nm, a band, present at 1,542 cm-1 in the stable reduced state but not present in the stable oxidized state, is absent in the intermediate state. We interpret the intermediate species as the state where the heme iron is reduced but the protein remains in the conformation of the oxidized state, with methionine-80 displaced as sixth ligand to the heme iron, before relaxing to the conformation of the stable reduced state, with methionine-80 returned as sixth ligand.

Raman spectroscopy and colorectal cancer

Project description:Background and study aims Bowel cancer is one of the commonest cancers worldwide. Earlier detection causes better outcomes for patients and longer survival. Symptoms of bowel cancer are non-specific and are shared by harmless bowel disorders. It is a challenge for doctors in general practice to diagnose bowel cancer and many symptomatic patients are sent to hospital for tests to rule it out. This is normally a colonoscopy, which is expensive, uncomfortable and can be harmful. The current approach to diagnosis causes great anxiety in patients waiting for these tests and is not a prudent approach to diagnosis. To allow earlier diagnosis by GPs we have studied whether a newly designed blood test taken in primary care is accurate and effective in patients with bowel symptoms that could be linked with cancer. This would benefit a large number of patients who are concerned about their bowel symptoms without the need for referral to hospital. It could also lead to diagnosis of bowel cancer at an earlier stage improving survival in the longer term. The aim of this study is to use a newly developed blood test for bowel cancer in primary care to achieve an earlier diagnosis. This would allow more timely appropriate treatment both for patients diagnosed with bowel cancer and those who are cancer free. Who can participate? Adults aged 50 years and older who have symptoms of bowel cancer. What does the study involve? Participants are randomly allocated to one of two groups. Those in the first group have their Raman spectroscopy blood test done immediately and those in the second grou have their sample tested after they’ve had a diagnosis. The amount of participants who are referred on the USC pathway from each group are assessed after 12 months.
