Complete Genome Sequence of an H12N8 Avian Influenza Virus Isolated from Wild Bird Feces in Hunan East Dongting Lake National Nature Reserve.

ABSTRACT: An H12N8 subtype avian influenza virus (AIV) was isolated from a wild bird in China in 2011. It is the first report of isolation of the H12N8 subtype AIV in Asia. Phylogenetic analysis results suggested it is a reassortant, and all eight gene segments belong to the Eurasian gene pool.

Phylogenetic analysis showed that it was a novel recombinant AIV between domestic ducks and wild waterfowl. Investigation of this virus is helpful for our understanding of the ecology of AIV in this region.

Phylogenetic and antigenic characterization of reassortant H9N2 avian influenza viruses isolated from wild waterfowl in the East Dongting Lake wetland in 2011-2012.

Project description:BackgroundWild waterfowl are recognized as the natural reservoir for influenza A viruses. Two distinct lineages, the American and Eurasian lineages, have been identified in wild birds. Gene flow between the two lineages is limited. The H9N2 virus has become prevalent in poultry throughout Eurasia, and mainly circulates in wild ducks and shorebirds in North America.MethodsIn this study, 22 H9N2 avian influenza viruses were isolated from wild waterfowl feces in East Dongting Lake Nature Reserve in November 2011 and March 2012. The phylogenetic, molecular, and antigenic characteristics of these viruses were analyzed based on analyses of the whole genome sequence of each isolate.ResultsPhylogenetic analyses indicated that these H9N2 viruses were generated by reassortment events. The HA, NA, PA, and NS genes were derived from the American gene pool, and the other four genes were derived from the Eurasian gene pool. Antigenic analyses indicated that these viruses were significantly different from the Eurasian lineage viruses.ConclusionsThis study presents the isolation of novel intercontinental recombinant H9N2 viruses from wild waterfowl in the East Dongting Lake wetland. The novel genotype H9N2 virus has not been detected in poultry in the region yet, and may be transmitted to naïve birds in poultry farms. Therefore, our results highlight the need for ongoing surveillance of wild birds and poultry in this region.

Investigation of avian influenza infections in wild birds, poultry and humans in Eastern Dongting Lake, China.

Project description:We investigated avian influenza infections in wild birds, poultry, and humans at Eastern Dongting Lake, China. We analyzed 6,621 environmental samples, including fresh fecal and water samples, from wild birds and domestic ducks that were collected from the Eastern Dongting Lake area from November 2011 to April 2012. We also conducted two cross-sectional serological studies in November 2011 and April 2012, with 1,050 serum samples collected from people exposed to wild birds and/or domestic ducks. Environmental samples were tested for the presence of avian influenza virus (AIV) using quantitative PCR assays and virus isolation techniques. Hemagglutination inhibition assays were used to detect antibodies against AIV H5N1, and microneutralization assays were used to confirm these results. Among the environmental samples from wild birds and domestic ducks, AIV prevalence was 5.19 and 5.32%, respectively. We isolated 39 and 5 AIVs from the fecal samples of wild birds and domestic ducks, respectively. Our analysis indicated 12 subtypes of AIV were present, suggesting that wild birds in the Eastern Dongting Lake area carried a diverse array of AIVs with low pathogenicity. We were unable to detect any antibodies against AIV H5N1 in humans, suggesting that human infection with H5N1 was rare in this region.

Surveillance of Schistosoma japonicum infection in domestic ruminants in the Dongting Lake region, Hunan province, China.

Project description:BACKGROUND: Schistosomiasis japonica is prevalent in Asian countries and it remains a major public health problem in China. The major endemic foci are the marsh and lake regions of southern China, particularly the Dongting Lake region bordering Hunan and Hubei provinces, and the Poyang Lake region in Jiangxi province. Domestic ruminants, especially bovines, have long been considered to play a major role in the transmission of Schistosoma japonicum to humans. METHODS AND FINDINGS: A miracidial hatching technique was used to investigate the prevalence of S. japonicum infections in domestic ruminants and field feces collected from two towns located to the south and east of Dongting Lake, Hunan province, between 2005 and 2010. The overall prevalence of infection was not significantly reduced from 4.93% in 2005 to 3.64% in 2008, after which it was maintained at this level. Bovines comprised 23.5-58.2% of the total infected ruminants, while goats comprised 41.8-76.5%. Infection rates in cattle and goats were significantly higher than those found in buffalo in most study years. The prevalence in buffalo younger than three years was significantly higher than that in those aged over three years. All the positive field samples of feces were derived from bovines in Nandashan. In Matang Town, 61.22% of the positive field feces were from bovines, while the rest were from goats. The positive rates for field feces were approximately the same in April and November/October. CONCLUSIONS: The present study found that bovines and goats are major sources of S. japonicum infection in the Dongting lake region and there was age-related resistance in buffalo. Both bovines and goats should be treated equally when controlling S. japonicum infections in the Dongting lake region. It is essential to conduct an additional mass treatment in late March or early April, in addition to the original treatment scheme.

Complete genome sequence of an H9N2 avian influenza virus isolated from egret in Lake Dongting wetland.

Project description:We isolated a recombinant H9N2 avian influenza virus (AIV) from fresh egret feces in the Ardeidae protection region of the Dongting Lake wetland area in China, and it was designated A/Egret/Hunan/1/2012(H9N2). This is the first report of isolating H9N2 AIV from wild birds in the Dongting Lake wetland. Its eight gene segments are generated by reassortment of gene segments of different AIV subtypes. These results are helpful for understanding the epidemiology and evolution of AIV in wild birds during migration.

Microplastics in Sediment and Surface Water of West Dongting Lake and South Dongting Lake: Abundance, Source and Composition.

Project description:Microplastic pollution was investigated in sediment and surface water in West Dongting Lake and South Dongting Lake for the first time. The abundance of microplastics ranged from 616.67 to 2216.67 items/m³ and 716.67 to 2316.67 items/m³ in the lakeshore surface water of West Dongting Lake and South Dongting Lake, respectively. The highest levels of microplastic pollution were found in the lakes' outlets. In the lake center sites of the West Dongting Lake and South Dongting Lake, the abundance of microplastics ranged from 433.33 to 1500 items/m³ and 366.67 to 1566.67 items/m³, respectively. Meanwhile, the study found that in lakeshore sediment of West Dongting Lake and South Dongting Lake, microplastic concentrations ranged from 320 to 480 items/m³ and 200?1150 items/m³. Polystyrene (PS) and polyethylene terephthalate (PET) were most common in the surface water and sediment samples, respectively. In addition, we suggest that the effects of polymer types in microplastics should be taken into account when considering abundance. This study can provide valuable points of reference to better understanding microplastic pollution in inland freshwater areas.

Persistence of avian influenza viruses in lake sediment, duck feces, and duck meat.

Project description:The persistence of 3 low-pathogenicity avian influenza viruses (LPAIV) (H4N6, H5N1, and H6N8) and one human influenza virus (H1N1) as well as Newcastle disease virus (NDV) and enteric cytopathogenic bovine orphan (ECBO) virus was investigated in lake sediment, duck feces, and duck meat at 30, 20, 10, and 0°C using a germ carrier technique. Virus-loaded germ carriers were incubated in each substrate, and residual infectivity of the eluted virus was quantified on cell culture after regular intervals for a maximum of 24 weeks. Data were analyzed by a linear regression model to calculate T(90) values (time required for 90% loss of virus infectivity) and estimated persistence of the viruses. In general, the persistence of all of the viruses was highest in lake sediment, followed by feces, and was the lowest in duck meat at all temperatures. For the avian influenza virus subtypes, T(90) values in sediment ranged from 5 to 11, 13 to 18, 43 to 54, and 66 to 394 days at 30, 20, 10, and 0°C, respectively, which were 2 to 5 times higher than the T(90) values of the viruses in the feces and meat. Although the individual viruses vary in tenacity, the survival time of influenza viruses was shorter than that of NDV and ECBO virus in all substrates. The results of this study suggest that lake sediment may act as a long-term source of influenza viruses in the aquatic habitat, while the viruses may remain infectious for extended periods of time in duck feces and meat at low temperatures, allowing persistence of the viruses in the environment over winter.

Occurrence dataset of birds in Sihong Hongze Lake Wetlands National Nature Reserve in China.

Project description:BackgroundHongze Lake is China's fourth largest freshwater lake and is also an important habitat for hundreds of thousands of migratory birds on the East Asian-Australian Flyway (EAAF). Sihong Hongze Lake Wetlands National Nature Reserve is located on the northwest of Hongze Lake, Sihong County, Jiangsu Province. The Reserve is a protected large area of natural lake wetlands, marsh wetlands and riverine wetlands and used as a stopover and wintering habitats for migratory birds. Previous studies have conducted bird diversity and temporal-spatial variation in this Reserve, but only for species of Anseriformes. There is still a lack of a comprehensive dataset on the number of bird species and individuals in this Reserve throughout the year. Our study was conducted from July 2020 to June 2021 to observe bird species composition and individual numbers at Sihong Hongze Lake Wetlands National Nature Reserve and provides an occurrence dataset with detailed species and geographic information.New informationThis occurrence dataset is the first public record of birds in Sihong Hongze Lake Wetlands National Nature Reserve for a whole year, which includes the taxonomic information, location information, number, investigation date and endangered level for each species. All data have been published on GBIF.

Complex reassortment of multiple subtypes of avian influenza viruses in domestic ducks at the Dongting Lake Region of China.

Project description:To gain insight into the ecology of avian influenza viruses (AIV), we conducted active influenza virus surveillance in domestic ducks on farms located on the flyway of migratory birds in the Dongting Lake region of Hunan Province, China, from winter 2011 until spring 2012. Specimens comprising 3,030 duck swab samples and 1,010 environmental samples were collected from 101 duck farms. We isolated AIV of various HA subtypes, including H3, H4, H5, H6, H9, H10, H11, and H12. We sequenced the entire coding sequences of the genomes of 28 representative isolates constituting 13 specific subtypes. When the phylogenetic relationships among these isolates were examined, we observed that extensive reassortment events had occurred. Among the 28 Dongting Lake viruses, 21 genotypes involving the six internal genes were identified. Furthermore, we identified viruses or viral genes introduced from other countries, viral gene segments of unknown origin, and a novel HA/NA combination. Our findings emphasize the importance of farmed domestic ducks in the Dongting Lake region to the genesis and evolution of AIV and highlight the need for continued surveillance of domestic ducks in this region.

Effects of hydrological regime on development of Carex wet meadows in East Dongting Lake, a Ramsar Wetland for wintering waterbirds.

Project description:Wet meadows are one of the most important ecological components in floodplain, and are among the most dynamic ecosystems. Understanding the development of wet meadows and contributing environmental factors can provide better support for wetland management. Carex meadows in East Dongting Lake National Nature Reserve (EDLNNR) provide vital wintering ground for thousands of migratory waterbirds, and their ecological functions are under threated due to hydrological alternation. We measured wet meadow expansion in EDLNNR from 1989 to 2014, and explored its responses to hydrological and climatic factors within the generalised additive models (GAM) framework. We found an overall expansion of wet meadows over the study period. However, in contrast to many previous studies, our results showed that water level fluctuations at the hydrologic indicator site had only limited impacts on their development. Instead, sampling year, timing of water level recession, and local rainfall exerted significant effects. The effects of sampling year reflected the changes in sedimentation within Dongting Lake; and effects of timing of water withdrawal might be explained by the life history of the dominant sedge species. Our study suggested that the impacts of large scale hydrological alternation on vegetation may operate indirectly through its effects on sediment balance.
