Full Genome Sequence of a Recombinant H1N2 Avian Influenza Virus Isolated from Wild Waterfowl in the East Dongting Lake Wetland.

ABSTRACT: Here, we report the full genome sequence of an H1N2 avian influenza virus (AIV) isolated from wild waterfowl in Dongting Lake. Phylogenetic analysis showed that it was a novel recombinant AIV between domestic ducks and wild waterfowl. Investigation of this virus is helpful for our understanding of the ecology of AIV in this region.

Phylogenetic and antigenic characterization of reassortant H9N2 avian influenza viruses isolated from wild waterfowl in the East Dongting Lake wetland in 2011-2012.

Project description:BackgroundWild waterfowl are recognized as the natural reservoir for influenza A viruses. Two distinct lineages, the American and Eurasian lineages, have been identified in wild birds. Gene flow between the two lineages is limited. The H9N2 virus has become prevalent in poultry throughout Eurasia, and mainly circulates in wild ducks and shorebirds in North America.MethodsIn this study, 22 H9N2 avian influenza viruses were isolated from wild waterfowl feces in East Dongting Lake Nature Reserve in November 2011 and March 2012. The phylogenetic, molecular, and antigenic characteristics of these viruses were analyzed based on analyses of the whole genome sequence of each isolate.ResultsPhylogenetic analyses indicated that these H9N2 viruses were generated by reassortment events. The HA, NA, PA, and NS genes were derived from the American gene pool, and the other four genes were derived from the Eurasian gene pool. Antigenic analyses indicated that these viruses were significantly different from the Eurasian lineage viruses.ConclusionsThis study presents the isolation of novel intercontinental recombinant H9N2 viruses from wild waterfowl in the East Dongting Lake wetland. The novel genotype H9N2 virus has not been detected in poultry in the region yet, and may be transmitted to naïve birds in poultry farms. Therefore, our results highlight the need for ongoing surveillance of wild birds and poultry in this region.

Complete genome sequence of an H9N2 avian influenza virus isolated from egret in Lake Dongting wetland.

Project description:We isolated a recombinant H9N2 avian influenza virus (AIV) from fresh egret feces in the Ardeidae protection region of the Dongting Lake wetland area in China, and it was designated A/Egret/Hunan/1/2012(H9N2). This is the first report of isolating H9N2 AIV from wild birds in the Dongting Lake wetland. Its eight gene segments are generated by reassortment of gene segments of different AIV subtypes. These results are helpful for understanding the epidemiology and evolution of AIV in wild birds during migration.

Characterization of an H10N8 influenza virus isolated from Dongting lake wetland.

Project description:BACKGROUND: Wild birds, especially those in wetlands and aquatic environments, are considered to be natural reservoirs of avian influenza viruses. It is accepted that water is an important component in the transmission cycle of avian influenza virus. Monitoring the water at aggregation and breeding sites of migratory waterfowl, mainly wetland, is very important for early detection of avian influenza virus. The epidemiology investigation of avian influenza virus was performed in Dongting lake wetland which is an international important wetland. RESULTS: An H10N8 influenza virus was isolated from Dongting Lake wetland in 2007. Phylogenetic analysis indicated that the virus was generated by multiple gene segment reassortment. The isolate was lowly pathogenic for chickens. However, it replicated efficiently in the mouse lung without prior adaptation, and the virulence to mice increased rapidly during adaptation in mouse lung. Sequence analysis of the genome of viruses from different passages showed that multiple amino acid changes were involved in the adaptation of the isolates to mice. CONCLUSIONS: The water might be an important component in the transmission cycle of avian influenza virus, and other subtypes of avian influenza viruses (other than H5, H7 and H9) might evolve to pose a potential threat to mammals and even humans.

Investigation of avian influenza infections in wild birds, poultry and humans in Eastern Dongting Lake, China.

Project description:We investigated avian influenza infections in wild birds, poultry, and humans at Eastern Dongting Lake, China. We analyzed 6,621 environmental samples, including fresh fecal and water samples, from wild birds and domestic ducks that were collected from the Eastern Dongting Lake area from November 2011 to April 2012. We also conducted two cross-sectional serological studies in November 2011 and April 2012, with 1,050 serum samples collected from people exposed to wild birds and/or domestic ducks. Environmental samples were tested for the presence of avian influenza virus (AIV) using quantitative PCR assays and virus isolation techniques. Hemagglutination inhibition assays were used to detect antibodies against AIV H5N1, and microneutralization assays were used to confirm these results. Among the environmental samples from wild birds and domestic ducks, AIV prevalence was 5.19 and 5.32%, respectively. We isolated 39 and 5 AIVs from the fecal samples of wild birds and domestic ducks, respectively. Our analysis indicated 12 subtypes of AIV were present, suggesting that wild birds in the Eastern Dongting Lake area carried a diverse array of AIVs with low pathogenicity. We were unable to detect any antibodies against AIV H5N1 in humans, suggesting that human infection with H5N1 was rare in this region.

Effects of hydrological regime on development of Carex wet meadows in East Dongting Lake, a Ramsar Wetland for wintering waterbirds.

Project description:Wet meadows are one of the most important ecological components in floodplain, and are among the most dynamic ecosystems. Understanding the development of wet meadows and contributing environmental factors can provide better support for wetland management. Carex meadows in East Dongting Lake National Nature Reserve (EDLNNR) provide vital wintering ground for thousands of migratory waterbirds, and their ecological functions are under threated due to hydrological alternation. We measured wet meadow expansion in EDLNNR from 1989 to 2014, and explored its responses to hydrological and climatic factors within the generalised additive models (GAM) framework. We found an overall expansion of wet meadows over the study period. However, in contrast to many previous studies, our results showed that water level fluctuations at the hydrologic indicator site had only limited impacts on their development. Instead, sampling year, timing of water level recession, and local rainfall exerted significant effects. The effects of sampling year reflected the changes in sedimentation within Dongting Lake; and effects of timing of water withdrawal might be explained by the life history of the dominant sedge species. Our study suggested that the impacts of large scale hydrological alternation on vegetation may operate indirectly through its effects on sediment balance.

Highly Pathogenic Avian Influenza (H5N1) Landscape Suitability Varies by Wetland Habitats and the Degree of Interface between Wild Waterfowl and Poultry in India.

Project description:Highly pathogenic avian influenza (HPAI) virus, subtype H5N1, constitutes one of the world's most important health and economic concerns given the catastrophic impact of epizootics on the poultry industry, the high mortality attending spillover in humans, and its potential as a source subtype for a future pandemic. Nevertheless, we still lack an adequate understanding of HPAI H5N1 epidemiology and infection ecology. The nature of the wild waterfowl-poultry interface, and the sharing of diverse wetland habitat among these birds, currently underscore important knowledge gaps. India has emerged as a global hotspot for HPAI H5N1, while also providing critical wintering habitat for many species of migratory waterfowl and year-round habitat for several resident waterfowl species. The current study sought to examine the extent to which the wild waterfowl-poultry interface, varied wetland habitat, and climate influence HPAI H5N1 epizootics in poultry in India. Using World Organisation for Animal Health reported outbreaks, this study showed that the wild waterfowl-poultry interface and lacustrine, riparian, and coastal marsh wetland systems were strongly associated with landscape suitability, and these relationships varied by scale. Although increasing poultry density was associated with increasing risk, this was only the case in the absence of wild waterfowl habitat, and only at a local scale. In landscapes increasingly shared between wild waterfowl and poultry, suitability was greater among lower density poultry, again at a local scale only. These findings provide further insight into the occurrence of HPAI H5N1 in India and suggest important landscape targets for blocking the waterfowl-poultry interface to interrupt virus transmission and prevent future outbreaks.

Microplastics in Sediment and Surface Water of West Dongting Lake and South Dongting Lake: Abundance, Source and Composition.

Project description:Microplastic pollution was investigated in sediment and surface water in West Dongting Lake and South Dongting Lake for the first time. The abundance of microplastics ranged from 616.67 to 2216.67 items/m³ and 716.67 to 2316.67 items/m³ in the lakeshore surface water of West Dongting Lake and South Dongting Lake, respectively. The highest levels of microplastic pollution were found in the lakes' outlets. In the lake center sites of the West Dongting Lake and South Dongting Lake, the abundance of microplastics ranged from 433.33 to 1500 items/m³ and 366.67 to 1566.67 items/m³, respectively. Meanwhile, the study found that in lakeshore sediment of West Dongting Lake and South Dongting Lake, microplastic concentrations ranged from 320 to 480 items/m³ and 200?1150 items/m³. Polystyrene (PS) and polyethylene terephthalate (PET) were most common in the surface water and sediment samples, respectively. In addition, we suggest that the effects of polymer types in microplastics should be taken into account when considering abundance. This study can provide valuable points of reference to better understanding microplastic pollution in inland freshwater areas.

Evidence of infection by H5N2 highly pathogenic avian influenza viruses in healthy wild waterfowl.

Project description:The potential existence of a wild bird reservoir for highly pathogenic avian influenza (HPAI) has been recently questioned by the spread and the persisting circulation of H5N1 HPAI viruses, responsible for concurrent outbreaks in migratory and domestic birds over Asia, Europe, and Africa. During a large-scale surveillance programme over Eastern Europe, the Middle East, and Africa, we detected avian influenza viruses of H5N2 subtype with a highly pathogenic (HP) viral genotype in healthy birds of two wild waterfowl species sampled in Nigeria. We monitored the survival and regional movements of one of the infected birds through satellite telemetry, providing a rare evidence of a non-lethal natural infection by an HP viral genotype in wild birds. Phylogenetic analysis of the H5N2 viruses revealed close genetic relationships with H5 viruses of low pathogenicity circulating in Eurasian wild and domestic ducks. In addition, genetic analysis did not reveal known gallinaceous poultry adaptive mutations, suggesting that the emergence of HP strains could have taken place in either wild or domestic ducks or in non-gallinaceous species. The presence of coexisting but genetically distinguishable avian influenza viruses with an HP viral genotype in two cohabiting species of wild waterfowl, with evidence of non-lethal infection at least in one species and without evidence of prior extensive circulation of the virus in domestic poultry, suggest that some strains with a potential high pathogenicity for poultry could be maintained in a community of wild waterfowl.
