Project description:Real-time structural changes in Na-β″-alumina were observed in situ using transmission electron microscopy (TEM) with electron beam irradiation. Na-β″-alumina has been widely investigated as a solid electrolyte material for sodium-sulfur secondary batteries owing to its high ionic conductivity. This high conductivity is known to be due to the Na+ ions on the loosely packed conduction planes of Na-β″-alumina. In the present study, we acquired real-time videos of the generation of spinel blocks caused by the conduction of Na+ ions. In addition, by observing Na extraction during electron beam irradiation, we experimentally confirmed that spinel block generation originates from the Na+ ion conduction, which has been a subject of recent debate.

Project description:Monolayer molybdenum disulfide (MoS2) semiconductors are the new generation of two-dimensional materials that possess several advantages compared to graphene due to their tunable bandgap and high electron mobility. Several approaches have been used to modify their physical properties for optical device applications. Here, we report a facile and non-destructive surface modification method for monolayer MoS2 via electron irradiation at a low, 5 kV accelerating voltage. After electron irradiation, the results of Raman and photoluminescence spectroscopy confirmed that the structure remains unchanged. However, when the modified surface was illuminated with a 532 nm laser for a prolonged period, the PL intensity was quenched as a result of oxygen desorption. Interestingly, the PL intensity can be recovered when left in ambient conditions for 10 h. The analysis of the PL spectrum revealed a decrease of trion, which is consistent with the readsorbed O2 molecules on the surface that deplete electrons and lead to PL recovery. We attribute this effect to the enhancement of the n-type character of monolayer MoS2 after electron irradiation. The sensitive nature of the modified surface to oxygen suggests that this approach may be used as a tool for the fabrication of MoS2 oxygen sensors.

Project description:Crystalline films of pentacene molecules, two to four monolayers in thickness, are grown via in situ sublimation on silicon substrates in the ultrahigh vacuum chamber of a low-energy electron microscope. It is observed that the diffraction pattern of the pentacene layers fades upon irradiation with low-energy electrons. The damage cross section is found to increase by more than an order of magnitude for electron energies from 0 to 10 eV and by another order of magnitude from 10 to 40 eV. Close to 0 eV, damage is virtually nil. Creation of chemically reactive atomic centers after electron attachment or impact ionization is thought to trigger chemical reactions between neighboring molecules that gradually transform the layer into a disordered carbon nanomembrane. Additionally, diminishing spectroscopic features related to the unoccupied band structure of the layers, accompanied by loss of definition in real-space images, and an increase in the background intensity of diffraction images during irradiation point to chemical changes and formation of a disordered layer.

Project description:The electron beam irradiation (EBI) of native lignin has received little attention. Thus, its potential use in lignin-based biorefineries is not fully understood. EBI was applied to selected lignin samples and the structural and chemical changes were analyzed, revealing the suitability, limitations, and potential purpose of EBI in wood biorefineries. Isolated milled wood, kraft, and sulfite lignin from beech and eucalyptus were subjected to up to 200 kGy of irradiation. The analysis included gel permeation chromatography for molar masses, heteronuclear single quantum coherence (HSQC)- and 31P NMR and headspace gas chromatography-mass spectrometry for functional groups, and thermogravimetric analysis for thermal stability. Most samples resisted irradiation. Subtle changes occurred in the molecular weight distribution and thermal stability of milled wood lignin. EBI was found to be a suitable pretreatment method for woody biomass if the avoidance of lignin condensation and chemical modification is a high priority.

Project description:ZnO nanorods (NRs) with an average length and diameter of 186 and 20 nm, respectively, were prepared through a mild solvothermal route and used as photocatalysts either as dispersed powder or immobilized on glass slides. The ZnO NRs were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), and X-ray diffraction (XRD). Dispersed ZnO NRs and, to a lesser extent, immobilized ZnO NRs were demonstrated to exhibit high photocatalytic activity under simulated sunlight of low intensity (5.5 mW/cm²) both for the degradation of the Orange II dye and for Escherichia coli bacterial decontamination (2.5-fold survival decrease after 180 min irradiation for immobilized NRs). SEM, atomic force microscopy (AFM), fluorescence spectroscopy, and epifluorescence microscopy demonstrate that cell surface damages are responsible of bacterial inactivation. The immobilized ZnO NRs could be reused up to five times for bacterial decontamination at comparable efficiency and therefore have great potential for real environmental applications.

Project description:Impact statementPericardium-based tissue transplantation is a lifesaving treatment. Commercial glutaraldehyde-treated pericardial tissue exhibits cytotoxicity, which is associated with the accelerated graft failure. Replacement of glutaraldehyde has been suggested to overcome those drawbacks. In this study, we report a toxin-free method that combines tissue stabilization with a terminal sterilization. Our data indicate that the SULEEI procedure, which is part of an issued patent, may be a promising first step toward glutaraldehyde-free pericardium-based tissue transplants. Thus, our results may contribute to improving cardiovascular treatment strategies.

Project description:The properties of layered materials are significantly dependent on their lattice orientations. Thus, the growth of graphene nanowalls (GNWs) on Cu through PECVD has been increasingly studied, yet the underlying mechanisms remain unclear. In this study, we examined the GNWs/Cu interface and investigated the evolution of their microstructure using advanced Scanning transmission electron microscopy and Electron Energy Loss Spectroscopy (STEM-EELS). GNWs interface and initial root layers of comprise graphitic carbon with horizontal basal graphene (BG) planes that conform well to the catalyst surface. In the vertical section, the walls show a mix of graphitic and turbostratic carbon, while the latter becomes more noticeable close to the top edges of the GMWs film. Importantly, we identified growth process began with catalysis at Cu interface forming BG, followed by defect induction and bending at 'coalescence points' of neighboring BG, which act as nucleation sites for vertical growth. We reported that although classical thermal CVD mechanism initially dominates, growth of graphene later deviates a few nanometers from the interface to form GNWs. Nascent walls are no longer subjected to the catalytic action of Cu, and their development is dominated by the stitching of charged carbon species originating in the plasma with basal plane edges.

Project description:NK-92 cells and their CAR-modified derivatives exhibit strong cytotoxic activity against tumors and are promising "off-the-shelf" therapeutics compared to CAR-T cells. In order to ensure safety and prevent the occurrence of secondary tumors, (CAR-)NK-92 cells need to be inactivated before transfusion. With increasing clinical use, there is a growing demand for enhanced safety and an efficient method that stops cell proliferation, but better maintains the cytotoxic effector functions of the cells compared to commonly used gamma irradiation. Recently, low-energy electron irradiation (LEEI) was successfully applied for inactivation of bacteria and viruses for vaccine production, and was described as a method for NK-92 inactivation for the first time. In the present publication, data on extensive characterization of LEEI and its comparison with gamma irradiation for the inactivation of parental NK-92 cells as well as CD123-directed CAR-NK-92 cells are provided. Our results show that both irradiation methods cause a progressive decrease in cell viability and are therefore suitable for inhibition of proliferation. Notably, cytotoxicity of the NK cells was significantly reduced by gamma irradiation, but not by LEEI three days after irradiation, compared to non-irradiated cells. Both gamma irradiation as well as LEEI led to substantial DNA-damage and an accumulation of irradiated cells in the G2/M phase. In addition, transcriptomic analysis of cells irradiated with both methods revealed approximately 12-fold more differentially expressed genes after gamma irradiation compared to LEEI. Analysis of surface molecules revealed an irradiation-induced decrease in CD56 expression but no changes in the levels of the activating receptors NKp46, NKG2D, and NKp30. Conclusions: The present data show that LEEI efficiently inactivates (CAR )NK-92 cells and sustains their cytotoxic capacity better than gamma irradiation. Taking into account additional logistic advantages of LEEI proves a potent alternative in the manufacturing of (CAR-)NK-92 cells for clinical application.

Project description:To probe the migration of free radicals (FRs), the reduction behaviours of hexavalent chromium (Cr(VI)) in water and ice by high-energy electron beam (HEEB) irradiation were investigated. Interestingly, the reductive efficiency (RE) of Cr(VI) in water was appreciably higher than that in ice. Thus, it was proposed that the migration ability of FRs in water is distinctly higher than that in ice, likely because the migration performance of FRs is closely related to the intermolecular distance of water molecules. Furthermore, the RE of Cr(VI) in ice decreased gradually with the distance from the irradiated area, indicating that FRs could migrate in ice and that the migration performance was closely related to the RE. Additionally, FRs (hydrated electrons ([Formula: see text]) and hydrogen radicals (·H)) generated during the irradiation process played a key role in the reduction of Cr(VI). Hydroxyl radicals (·OH) and H2O2 were the dominant negative factors for the reduction because of their oxidizing effects, but these factors could be eliminated by the removal of ·OH. This work reveals the migration performance of FRs in different media for the first time. This result may be useful for basic and applied studies in fields of environmental science related to FRs.

Project description:A simple donor-acceptor linked dyad, 9-mesityl-10-methylacridinium ion (Acr(+)-Mes) was incorporated into nanosized mesoporous silica-alumina to form a composite, which in acetonitrile is highly dispersed. In this medium, upon visible light irradiation, the formation of an extremely long-lived electron-transfer state (Acr(•)-Mes(•+)) was confirmed by EPR and laser flash photolysis spectroscopic methods. The composite of Acr(+)-Mes-incorporated mesoporous silica-alumina with an added copper complex [(tmpa)Cu(II)] (ClO(4)(-))2 (tmpa = tris(2-pyridylmethyl)amine) acts as an efficient and robust photocatalyst for the selective oxygenation of p-xylene by molecular oxygen to produce p-tolualdehyde and hydrogen peroxide. Thus, incorporation of Acr(+)-Mes into nanosized mesoporous silica-alumina combined with an O(2)-reduction catalyst ([(tmpa)Cu(II)](2+)) provides a promising method in the development of efficient and robust organic photocatalysts for substrate oxygenation by dioxygen, the ultimate environmentally benign oxidant.
