Project description:Common strategies for conversion of lignocellulosic biomass to chemical products center on deconstructing biomass polymers into fermentable sugars. Here, we demonstrate an alternative strategy, a growth-coupled, high-yield bioconversion, by feeding cells a non-sugar substrate, by-passing central metabolism, and linking a key metabolic step to generation of acetyl-CoA that is required for biomass and energy generation. Specifically, we converted levulinic acid (LA), an established degradation product of lignocellulosic biomass, to butanone (a.k.a. methyl-ethyl ketone - MEK), a widely used industrial solvent. Our strategy combines a catabolic pathway from Pseudomonas putida that enables conversion of LA to 3-ketovaleryl-CoA, a CoA transferase that generates 3-ketovalerate and acetyl-CoA, and a decarboxylase that generates 2-butanone. By removing the ability of E. coli to consume LA and supplying excess acetate as a carbon source, we built a strain of E. coli that could convert LA to butanone at high yields, but at the cost of significant acetate consumption. Using flux balance analysis as a guide, we built a strain of E. coli that linked acetate assimilation to production of butanone. This strain was capable of complete bioconversion of LA to butanone with a reduced acetate requirement and increased specific productivity. To demonstrate the bioconversion on real world feedstocks, we produced LA from furfuryl alcohol, a compound readily obtained from biomass. These LA feedstocks were found to contain inhibitors that prevented cell growth and bioconversion of LA to butanone. We used a combination of column chromatography and activated carbon to remove the toxic compounds from the feedstock, resulting in LA that could be completely converted to butanone. This work motivates continued collaboration between chemical and biological catalysis researchers to explore alternative conversion pathways and the technical hurdles that prevent their rapid deployment.

Project description:Microorganisms can catabolize a wide range of organic compounds and therefore have the potential to perform many industrially relevant bioconversions. One barrier to realizing the potential of biorefining strategies lies in our incomplete knowledge of metabolic pathways, including those that can be used to assimilate naturally abundant or easily generated feedstocks. For instance, levulinic acid (LA) is a carbon source that is readily obtainable as a dehydration product of lignocellulosic biomass and can serve as the sole carbon source for some bacteria. Yet, the genetics and structure of LA catabolism have remained unknown. Here, we report the identification and characterization of a seven-gene operon that enables LA catabolism in Pseudomonas putida KT2440. When the pathway was reconstituted with purified proteins, we observed the formation of four acyl-CoA intermediates, including a unique 4-phosphovaleryl-CoA and the previously observed 3-hydroxyvaleryl-CoA product. Using adaptive evolution, we obtained a mutant of Escherichia coli LS5218 with functional deletions of fadE and atoC that was capable of robust growth on LA when it expressed the five enzymes from the P. putida operon. This discovery will enable more efficient use of biomass hydrolysates and metabolic engineering to develop bioconversions using LA as a feedstock.

Project description:In this work, acid-catalyzed conversion of cellulose into levulinic acid in a biphasic solvent system was developed. Compared to a series of catalysts investigated in this study, the Amberlyst-15 as a more efficient acid catalyst was used in the hydrolysis of cellulose and further dehydration of derived intermediates into levulinic acid. Besides, the mechanism of biphasic solvent system in the conversion of cellulose was studied in detail, and the results showed biphasic solvent system can promote the conversion of cellulose and suppress the polymerization of the by-products (such as lactic acid).The reaction conditions, such as temperature, time, and catalyst loading were changed to investigate the effect on the yield of levulinic acid. The results indicated that an appealing LA yield of 59.24% was achieved at 200°C and 180 min with a 2:1 ratio of Amberlyst-15 catalyst and cellulose in GVL/H2O under N2 pressure. The influence of different amounts of NaCl addition to this reaction was also investigated. This study provides an economical and environmental-friendly method for the acid-catalyzed conversion of cellulose and high yield of the value-added chemical.

Project description:5-Hydroxymethylfurfural (HMF) is a versatile intermediate in biomass conversion pathways. However, the notoriously unstable nature of HMF imposes challenges to design selective routes to chemicals such as furan-2,5-dicarboxylic acid (FDCA). Here, a new strategy for obtaining furans is presented, bypassing the formation of the unstable HMF. Instead of starting with glucose/fructose and thus forming HMF as an intermediate, the new route starts from uronic acids, which are abundantly present in many agro residues such as sugar beet pulp, potato pulp, and citrus peels. Conversion of uronic acids, via ketoaldonic acids, to the intermediate formylfuroic acid (FFA) esters, and subsequently to FDCA esters, proceeds without formation of levulinic acid or insoluble humins. This new route provides an attractive strategy to valorize agricultural waste streams and a route to furanic building blocks without the co-production of levulinic acid or humins.

Project description:We report here the synthesis of poly(4-ketovalerolactone) (PKVL) via ring-opening transesterification polymerization (ROTEP) of the monomer 4-ketovalerolactone (KVL, two steps from levulinic acid). The polymerization of KVL proceeds to high equilibrium monomer conversion (up to 96% in the melt) to give the semicrystalline polyketoester PKVL with low dispersity. PKVL displays glass transition temperatures of 7 °C and two melting temperatures at 132 and 148 °C. This polyester can be chemically recycled through hydrolytic degradation. Under aqueous neutral or acidic conditions, the dominating pathway for polyester hydrolysis is through backbiting from the chain end. Under basic conditions, mid-chain cleavage, accelerated by the ketone carbonyl group in the backbone, promotes the hydrolysis of nearby backbone ester bonds. The final hydrolysis product is 5-hydroxylevulinic acid, the ring opened hydrolysis product of KVL. PKVL was also observed to degrade under the action of a Brønsted acid to a bis-spirocyclic dilactone natural product altaicadispirolactone, which is a dimer of KVL. This constitutes a rare example of a one-step synthesis of a secondary metabolite of non-trivial structure in which a polymer was the starting material and the sole source of matter. Analogous ROTEP of the isomeric 4-membered lactone 4-acetyl-β-propiolactone (APL) was also explored, although this chemistry was not as well-behaved as the KVL to PKVL polymerization.

Project description:These days, there is a need to develop novel and emerging processing pathways that permit production of value-added substances and fuels considering sustainability aspects. In this sense, levulinic acid (LA) is one of the most promising biorefinery products. This paper presents environmental and safety assessments of LA production via acid-catalyzed dehydration (ACD) of biomass. The process was modeled by using Aspen Plus process simulation software based on a capacity of 132 000 tons per annum of banana rachis (main raw material). Likewise, environmental and safety assessments were developed. Parameters such as heats of reaction, explosivity, toxicity of substances, and operational conditions along with extended mass and energy balances were used to perform safety and environmental analyses. In this regard, the modeled topology showed an inherent safety index (ISI) score of 24 with an equal contribution of 12 points for both chemical inherent safety index (CIS) and process inherent safety index (PIS). ACD showed a good safety performance, with moderate concerns related to the handling of formic acid. Moreover, the waste reduction algorithm (WAR) was used to assess environmental performance and estimate potential environmental impacts (PEIs) of the simulated topology. It was performed considering four case studies to determine the influence of mass streams (case 1), product streams (case 2), energy streams (case 3), and simultaneous products and energy contribution (case 4). This analysis showed that for this process, the total inletting flow of impacts that enter was less than the amount of these that leave the system according to a generation rate of the PEI for case 1 (-1.89 × 102 PEI/h) and case 3 (-1.83 × 102 PEI/h). From the environmental viewpoint, the major concern is associated with the photochemical oxidation potential category because of the handling of volatile organic compounds through the process.

Project description:Glucosamine is a monomer of chitosan, which is a biopolymer derived from the exoskeletons of crustaceans. This work investigated the conversion of glucosamine into the platform chemical levulinic acid (LA), in a sulfamic acid-catalyzed hydrothermal reaction. The optimized results of LA production showed that the conditions of 200 °C, 125 g L-1 glucosamine, 0.3 M sulfamic acid, and 15 min produced a 33.76 ± 0.19 mol% LA yield. The same conditions produced only 0.14 mol% 5-HMF yield. These results show that glucosamine is a potential bioresource to produce platform chemicals. Also, in the field of biofuels and chemical synthesis processes, the catalytic system using sulfamic acid is significant.

Project description:Methylorubrum extorquens AM1 is an efficient platform strain possessing biotechnological potential in formate- and methanol-based single carbon (C1) bioeconomy. Constitutive expression or costly chemical-inducible expression systems are not always desirable. Here, several glucose-, xylose-, and levulinic acid (LA)-inducible promoter systems were assessed for the induction of green fluorescent protein (GFP) as a reporter protein. Among them, the LA-inducible gene expression system (HpdR/P hpdH ) showed a strong expression of GFP (51-fold) compared to the control. The system was induced even at a low concentration of LA (0.1 mM). The fluorescence intensity increased with increasing concentrations of LA up to 20 mM. The system was tunable and tightly controlled with meager basal expression. The maximum GFP yield obtained using the system was 42 mg/g biomass, representing 10% of the total protein content. The efficiency of the proposed system was nearly equivalent (90%-100%) to that of the widely used strong promoters such as P mxaF and P L/O4 . The HpdR/P hpdH system worked equally efficiently in five different strains of M. extorquens. LA is a low-cost, renewable, and sustainable platform chemical that can be used to generate a wide range of products. Hence, the reported system in potent strains of M. extorquens is highly beneficial in the C1-biorefinery industry to produce value-added products and bulk chemicals.

Project description:Levulinic acid (LA) can be cost-effectively produced from a vast array of renewable carbohydrate-containing biomaterials. LA could facilitate the commercialization of the polymer poly(hydroxybutyrate-co-hydroxyvalerate) (PHBV) and PHBV-based products as carbon substrates. Therefore, this paper focused on the production of PHBV by Ralstonia eutropha with LA for hydroxyvalerate (HV) production, which plays an important role in enhancing the thermal properties of PHBV. Accordingly, the HV content of PHBV varied from 0-40.9% at different concentrations of LA. Stimulation of cell growth and PHBV accumulation were observed when 2-6 g L(-1) LA was supplied to the culture. The optimal nitrogen sources were determined to be 0.5 g L(-1) ammonium chloride and 2 g L(-1) casein peptone. It was determined that the optimal pH for cell growth and PHBV accumulation was 7.0. When the cultivation was performed in large scale (2 L fermenter) with a low DO concentration of 30% and a pH of 7.0, a high maximum dry cell weight of 15.53 g L(-1) with a PHBV concentration of 12.61 g L(-1) (53.9% HV), up to 81.2% of the dry cell weight, was obtained. The melting point of PHBV found to be decreased as the fraction of HV present in the polymer increased, which resulted in an improvement in the ductility and flexibility of the polymer. The results of this study will improve the understanding of the PHBV accumulation and production by R. eutropha and will be valuable for the industrial production of biosynthesized polymers.
