Merging metabolism and power: development of a novel photobioelectric device driven by photosynthesis and respiration.

ABSTRACT: Generation of renewable energy is one of the grand challenges facing our society. We present a new bio-electric technology driven by chemical gradients generated by photosynthesis and respiration. The system does not require pure cultures nor particular species as it works with the core metabolic principles that define phototrophs and heterotrophs. The biology is interfaced with electrochemistry with an alkaline aluminum oxide cell design. In field trials we show the system is robust and can work with an undefined natural microbial community. Power generated is light and photosynthesis dependent. It achieved a peak power output of 33 watts/m(2) electrode. The design is simple, low cost and works with the biological processes driving the system by removing waste products that can impede growth. This system is a new class of bio-electric device and may have practical implications for algal biofuel production and powering remote sensing devices.

Steeper declines in forest photosynthesis than respiration explain age-driven decreases in forest growth.

Project description:The traditional view of forest dynamics originated by Kira and Shidei [Kira T, Shidei T (1967) Jap J Ecol 17:70-87] and Odum [Odum EP (1969) Science 164(3877):262-270] suggests a decline in net primary productivity (NPP) in aging forests due to stabilized gross primary productivity (GPP) and continuously increased autotrophic respiration (Ra). The validity of these trends in GPP and Ra is, however, very difficult to test because of the lack of long-term ecosystem-scale field observations of both GPP and Ra. Ryan and colleagues [Ryan MG, Binkley D, Fownes JH (1997) Ad Ecol Res 27:213-262] have proposed an alternative hypothesis drawn from site-specific results that aboveground respiration and belowground allocation decreased in aging forests. Here, we analyzed data from a recently assembled global database of carbon fluxes and show that the classical view of the mechanisms underlying the age-driven decline in forest NPP is incorrect and thus support Ryan's alternative hypothesis. Our results substantiate the age-driven decline in NPP, but in contrast to the traditional view, both GPP and Ra decline in aging boreal and temperate forests. We find that the decline in NPP in aging forests is primarily driven by GPP, which decreases more rapidly with increasing age than Ra does, but the ratio of NPP/GPP remains approximately constant within a biome. Our analytical models describing forest succession suggest that dynamic forest ecosystem models that follow the traditional paradigm need to be revisited.

Both respiration and photosynthesis determine the scaling of plankton metabolism in the oligotrophic ocean.

Project description:Despite its importance to ocean-climate interactions, the metabolic state of the oligotrophic ocean has remained controversial for >15 years. Positions in the debate are that it is either hetero- or autotrophic, which suggests either substantial unaccounted for organic matter inputs, or that all available photosynthesis (P) estimations (including (14)C) are biased. Here we show the existence of systematic differences in the metabolic state of the North (heterotrophic) and South (autotrophic) Atlantic oligotrophic gyres, resulting from differences in both P and respiration (R). The oligotrophic ocean is neither auto- nor heterotrophic, but functionally diverse. Our results show that the scaling of plankton metabolism by generalized P:R relationships that has sustained the debate is biased, and indicate that the variability of R, and not only of P, needs to be considered in regional estimations of the ocean's metabolic state.

Merging transformation optics with electron-driven photon sources.

Project description:Relativistic electron beams create optical radiation when interacting with tailored nanostructures. This phenomenon has been so far used to design grating-based and holographic electron-driven photon sources. It has been proposed recently that such sources can be used for hybrid electron- and light-based spectroscopy techniques. However, this demands the design of a thin-film source suitable for electron-microscopy applications. Here, we present a mesoscopic structure composed of an array of nanoscale holes in a gold film which is designed using transformation optics and delivers ultrashort chirped electromagnetic wave packets upon 30-200 keV electron irradiation. The femtosecond photon bunches result from coherent scattering of surface plasmon polaritons with hyperbolic dispersion. They decay by radiation in a broad spectral band which is focused into a 1.5 micrometer beam waist. The focusing ability and broadband nature of this photon source will initiate applications in ultrafast spectral interferometry techniques.

A Novel Photo-Driven Hydrogenation Reaction of an NAD+-Type Complex Toward Artificial Photosynthesis.

Project description:The photocatalytic reduction of carbon dioxide (CO2) to value-added chemicals is an attractive strategy to utilize CO2 as a feedstock for storing renewable energy, such as solar energy, in chemical bonds. Inspired by the biological function of the nicotinamide adenine dinucleotide redox couple (NAD+/NADH), we have been developing transition-metal complexes containing NAD+/NADH-functionalized ligands to create electro- and/or photochemically renewable hydride donors for the conversion of CO2 into value-added chemicals. Our previous findings have provided insights for the development of photocatalytic organic hydride reduction reactions for CO2, however, further examples, as well as investigation, of these photo-driven NAD+/NADH-type hydrogenation and organic hydride transfer reactions are required not only to explore the mechanism in detail but also to develop a highly efficient catalyst for artificial photosynthesis. In this paper, we report the synthesis, characterization, and photo-induced NAD+/NADH conversion properties of a new ruthenium(II) complex, [Ru(bpy)2(Me-pn)](PF6)2 (1), which contains a new NAD+-type ligand, Me-pn (2-methyl-6-(pyridin-2-yl)-1,5-naphthyridine). In addition, we have succeeded in the isolation of the corresponding two-electron reduced ruthenium(II) complex containing the NADH-type ligand Me-pnHH (2-methyl-6-(pyridin-2-yl)-1,4-dihydro-1,5-naphthyridine), i.e., [Ru(bpy)2(Me-pnHH)](PF6)2 (1HH), by the photo-induced hydrogenation reaction of 1. Thus, in this study, a new photo-driven NAD+/NADH-type hydrogenation reaction for possible CO2 reduction using the NAD+/NADH redox function has been constructed.

Partitioning net carbon dioxide fluxes into photosynthesis and respiration using neural networks.

Project description:The eddy covariance (EC) technique is used to measure the net ecosystem exchange (NEE) of CO2 between ecosystems and the atmosphere, offering a unique opportunity to study ecosystem responses to climate change. NEE is the difference between the total CO2 release due to all respiration processes (RECO), and the gross carbon uptake by photosynthesis (GPP). These two gross CO2 fluxes are derived from EC measurements by applying partitioning methods that rely on physiologically based functional relationships with a limited number of environmental drivers. However, the partitioning methods applied in the global FLUXNET network of EC observations do not account for the multiple co-acting factors that modulate GPP and RECO flux dynamics. To overcome this limitation, we developed a hybrid data-driven approach based on combined neural networks (NNC-part ). NNC-part incorporates process knowledge by introducing a photosynthetic response based on the light-use efficiency (LUE) concept, and uses a comprehensive dataset of soil and micrometeorological variables as fluxes drivers. We applied the method to 36 sites from the FLUXNET2015 dataset and found a high consistency in the results with those derived from other standard partitioning methods for both GPP (R2 > .94) and RECO (R2 > .8). High consistency was also found for (a) the diurnal and seasonal patterns of fluxes and (b) the ecosystem functional responses. NNC-part performed more realistic than the traditional methods for predicting additional patterns of gross CO2 fluxes, such as: (a) the GPP response to VPD, (b) direct effects of air temperature on GPP dynamics, (c) hysteresis in the diel cycle of gross CO2 fluxes, (d) the sensitivity of LUE to the diffuse to direct radiation ratio, and (e) the post rain respiration pulse after a long dry period. In conclusion, NNC-part is a valid data-driven approach to provide GPP and RECO estimates and complementary to the existing partitioning methods.

Scaling up calcification, respiration, and photosynthesis rates of six prominent coral taxa.

Project description:Coral reefs provide a range of important services to humanity, which are underpinned by community-level ecological processes such as coral calcification. Estimating these processes relies on our knowledge of individual physiological rates and species-specific abundances in the field. For colonial animals such as reef-building corals, abundance is frequently expressed as the relative surface cover of coral colonies, a metric that does not account for demographic parameters such as coral size. This may be problematic because many physiological rates are directly related to organism size, and failure to account for linear scaling patterns may skew estimates of ecosystem functioning. In the present study, we characterize the scaling of three physiological rates - calcification, respiration, and photosynthesis - considering the colony size for six prominent, reef-building coral taxa in Mo'orea, French Polynesia. After a seven-day acclimation period in the laboratory, we quantified coral physiological rates for three hours during daylight (i.e., calcification and gross photosynthesis) and one hour during night light conditions (i.e., dark respiration). Our results indicate that area-specific calcification rates are higher for smaller colonies across all taxa. However, photosynthesis and respiration rates remain constant over the colony-size gradient. Furthermore, we revealed a correlation between the demographic dynamics of coral genera and the ratio between net primary production and calcification rates. Therefore, intraspecific scaling of reef-building coral physiology not only improves our understanding of community-level coral reef functioning but it may also explain species-specific responses to disturbances.

Genetics-driven discovery of novel regulators of lipid metabolism.

Project description:Purpose of reviewResidual cardiovascular disease risk and increasing metabolic syndrome risk underscores a need for novel therapeutics targeting lipid metabolism in humans. Unbiased human genetic screens have proven powerful in identifying novel genomic loci, and this review discusses recent developments in such discovery.Recent findingsRecent human genome-wide association studies have been completed in incredibly large, detailed cohorts, allowing for the identification of more than 300 genomic loci that participate in the regulation of plasma lipid metabolism. However, the discovery of these loci has greatly outpaced the elucidation of the underlying functional mechanisms. The identification of novel roles for long noncoding RNAs, such as CHROME, LeXis, and MeXis, in lipid metabolism suggests that noncoding RNAs should be included in the functional translation of GWAS loci.SummaryUnbiased genetic studies appear to have unearthed a great deal of novel biology with respect to lipid metabolism, yet translation of these findings into actionable mechanisms has been slow. Increased focus on the translation, rather than the discovery, of these loci, with new attention paid to lncRNAs, can help spur the development of novel therapeutics targeting lipid metabolism.

Reciprocal regulation of photosynthesis and mitochondrial respiration by TOR kinase in Chlamydomonas reinhardtii.

Project description:While the role of TOR kinase in the chloroplast biogenesis and transcriptional regulation of photosynthesis is well documented in Arabidopsis, the functional relevance of this metabolic sensor kinase in chloroplast-mitochondria cross talk is unknown. Using Chlamydomonas reinhardtii as the model system, we demonstrate the role of TOR kinase in the regulation of chloroplast and mitochondrial functions: We show that TOR kinase inhibition impairs the maintenance of high ETR associated with PSII and low NPQ and inhibits efficient state transitions between PSII and PSI. While compromised photosynthetic functions are observed in TOR kinase inhibited cells, same conditions lead to augmentation in mitochondrial basal respiration rate by twofold and concomitantly a rise in ATP production. Interestingly, such upregulated mitochondrial functions in TOR-inhibited cells are mediated by fragmented mitochondria via upregulating COXIIb and downregulating Hxk1 and AOX1 protein levels. We propose that TOR kinase may act as a sensor that counter-regulates chloroplast versus mitochondrial functions in a normal C. reinhardtii cell.
