Loss of arbuscular mycorrhizal fungal diversity in trap cultures during long-term subculturing.

ABSTRACT: Long-term successional dynamics of an inoculum of arbuscular mycorrhizal fungi (AMF) associated with the maize rhizosphere (from traditionally managed agroecosystems in Los Tuxtlas, Veracruz, Mexico), was followed in Bracchiaria comata trap cultures for almost eight years. The results indicate that AMF diversity is lost following long-term subculturing of a single plant host species. Only the dominant species, Claroideoglomus etunicatum, persisted in pot cultures after 13 cycles. The absence of other morphotypes was demonstrated by an 18S rDNA survey, which confirmed that the sequences present solely belonged to C. etunicatum. Members of Diversisporales were the first to decrease in diversity, and the most persistent species belonged to Glomerales.

Project description:Growing evidence showed that efficient acquisition and use of nutrients by crops is controlled by root-associated microbiomes. Efficient management of this system is essential to improving crop yield, while reducing the environmental footprint of crop production. Both endophytic and rhizospheric microorganisms can directly promote crop growth, increasing crop yield per unit of soil nutrients. A variety of plant symbionts, most notably the arbuscular mycorrhizal fungi (AMF), nitrogen-fixing bacteria, and phosphate-potassium-solubilizing microorganisms entered the era of large-scale applications in agriculture, horticulture, and forestry. The purpose of this study is to compile data to give a complete and comprehensive assessment and an update of mycorrhizal-based inoculant uses in agriculture in the past, present, and future. Based on available data, 68 mycorrhizal products from 28 manufacturers across Europe, America, and Asia were examined on varying properties such as physical forms, arbuscular mycorrhizal fungal composition, number of active ingredients, claims of purpose served, mode of application, and recommendation. Results show that 90% of the products studied are in solid formula-powder (65%) and granular (25%), while only 10% occur in liquid formula. We found that 100% of the products are based on the Glomeraceae of which three species dominate among all the products in the order of Rhizophagus irregularis (39%), Funneliformis mosseae (21%), Claroideoglomus etunicatum (16%). Rhizophagus clarus is the least common among all the benchmark products. One third of the products is single species AMF and only 19% include other beneficial microbes. Of the sampled products, 44% contain AMF only while the rest are combined with varying active ingredients. Most of the products (84%) claimed to provide plant nutrient benefits. Soil application dominates agricultural practices of the products and represents 47%. A substantial amount of the inoculants were applied in cereal production. Recommended application doses varied extensively per plant, seed and hectare. AMF inoculant seed coating accounted for 26% of the products' application and has great potential for increased inoculation efficiency over large-scale production due to minimum inoculum use. More applied research should also be conducted on the possible combination of AMF with other beneficial microbes.

Project description:BACKGROUND:Establishing mixed plantations is an effective way to improve soil fertility and increase forest productivity. Arbuscular mycorrhizal (AM) fungi are obligate symbiotic fungi that can promote mineral nutrient absorption and regulate intraspecific and interspecific competition in plants. However, the effects of mixed plantations on the community structure and abundance of AM fungi are still unclear. Illumina MiSeq sequencing was used to investigate the AM fungal community in the roots and soils of pure and mixed plantations (Juglans mandshurica × Larix gmelinii). The objective of this study is to compare the differential responses of the root and rhizosphere soil AM fungal communities of Juglans mandshurica to long-term mixed plantation management. RESULTS:Glomus and Paraglomus were the dominant genera in the root samples, accounting for more than 80% of the sequences. Compared with that in the pure plantation, the relative abundance of Glomus was higher in the mixed plantation. Glomus, Diversispora and Paraglomus accounted for more than 85% of the sequences in the soil samples. The relative abundances of Diversispora and an unidentified genus of Glomeromycetes were higher and lower in the pure plantation, respectively. The Root_P samples (the roots in the pure plantation) had the highest number of unique OTUs (operational taxonomic units), which belonged mainly to an unidentified genus of Glomeromycetes, Paraglomus, Glomus and Acaulospora. The number of unique OTUs detected in the soil was lower than that in the roots. In both the root and soil samples, the forest type did not have a significant effect on AM fungal diversity, but the Sobs value and the Shannon, Chao1 and Ace indices of AM fungi in the roots were significantly higher than those in the soil. CONCLUSIONS:Mixed forest management had little effect on the AM fungal community of Juglans mandshurica roots and significantly changed the community composition of the soil AM fungi, but not the diversity.
