Conference highlights of the 16th International Conference on Human Retrovirology: HTLV and related retroviruses, 26-30 June 2013, Montreal, Canada.

ABSTRACT: The 16th International Conference on Human Retrovirology: HTLV and Related Retroviruses was held in Montreal, Québec from June 26th to June 30th, 2013 and was therefore hosted by a Canadian city for the first time. The major topic of the meeting was human T-lymphotropic viruses (HTLVs) and was covered through distinct oral and poster presentation sessions: clinical research, animal models, immunology, molecular and cellular biology, human endogenous and emerging exogenous retroviruses and virology. In this review, highlights of the meeting are provided by different experts for each of these research areas.

Host-pathogen interactome mapping for HTLV-1 and -2 retroviruses.

Project description:BackgroundHuman T-cell leukemia virus type 1 (HTLV-1) and type 2 both target T lymphocytes, yet induce radically different phenotypic outcomes. HTLV-1 is a causative agent of Adult T-cell leukemia (ATL), whereas HTLV-2, highly similar to HTLV-1, causes no known overt disease. HTLV gene products are engaged in a dynamic struggle of activating and antagonistic interactions with host cells. Investigations focused on one or a few genes have identified several human factors interacting with HTLV viral proteins. Most of the available interaction data concern the highly investigated HTLV-1 Tax protein. Identifying shared and distinct host-pathogen protein interaction profiles for these two viruses would enlighten how they exploit distinctive or common strategies to subvert cellular pathways toward disease progression.ResultsWe employ a scalable methodology for the systematic mapping and comparison of pathogen-host protein interactions that includes stringent yeast two-hybrid screening and systematic retest, as well as two independent validations through an additional protein interaction detection method and a functional transactivation assay. The final data set contained 166 interactions between 10 viral proteins and 122 human proteins. Among the 166 interactions identified, 87 and 79 involved HTLV-1 and HTLV-2 -encoded proteins, respectively. Targets for HTLV-1 and HTLV-2 proteins implicate a diverse set of cellular processes including the ubiquitin-proteasome system, the apoptosis, different cancer pathways and the Notch signaling pathway.ConclusionsThis study constitutes a first pass, with homogeneous data, at comparative analysis of host targets for HTLV-1 and -2 retroviruses, complements currently existing data for formulation of systems biology models of retroviral induced diseases and presents new insights on biological pathways involved in retroviral infection.

Podium Session 1: Prostate Cancer June 26, 2016 1050–1150

Project description:Introduction and Objectives: The phase 2 TERRAIN trial compared the efficacy and safety of enzalutamide (ENZA) vs. bicalutamide (BIC) in patients (pts) with metastatic castration-resistant prostate cancer who had progressed on luteinizing hormone-releasing hormone agonist/antagonist therapy or after bilateral orchiectomy while maintaining castration therapy during the study. An age effect analysis was pre-specified to investigate the efficacy and safety of ENZA vs. BIC. Results are presented in younger (<75 years) and older (≥75 years) pts in the TERRAIN population. Methods: In this double-blind study in North America and Europe, pts were randomized 1:1 to ENZA 160 mg/day or BIC 50 mg/day. The primary efficacy endpoint was centrally assessed progression-free survival (PFS) and a secondary efficacy endpoint was time to prostate-specific antigen (PSA) progression. Results: 184 pts were randomized to ENZA and 191 pts to BIC. 126 (68.5%) and 119 (62.3%) pts were <75, and 58 (31.5%) and 72 (37.7%) pts were ≥75, in the ENZA and BIC arms, respectively. PFS was significantly improved with ENZA vs. BIC in pts <75 years (median 16.6 vs. 5.8 months; HR 0.38 (95% CI 0.27, 0.52) and pts ≥75 years (median 13.8 vs. 6.4 months; HR 0.59 (95% CI 0.37, 0.92). Median time to PSA progression was similarly significantly improved with ENZA vs. BIC in younger (median 22.1 vs. 8.2 months; HR 0.27 (95% CI 0.18, 0.40) and older pts (median 16.6 vs. 5.8 months; HR 0.33 (95% CI 0.19, 0.57). Adverse events (AEs) with ENZA were more frequent in older pts (98.3%) vs. younger pts (92.8%), but a similar distribution of treatment-related AEs between treatment arms was observed in either age group. Conclusions: ENZA had greater efficacy than BIC regardless of age, with superior PFS and time to PSA progression. ENZA showed safety consistent with its known safety profile in both age subgroups.

HIVR4P 2016, Partnering for Prevention: Conference Summary and Highlights.

Project description:HIV Research for Prevention: AIDS Vaccine, Microbicide, and ARV-based Prevention Science (HIVR4P) was built on a growing consensus that effective HIV prevention requires a combination of approaches and that understanding, analyzing, and debating the cross-cutting issues that impact prevention research are all essential to combat the global HIV/AIDS epidemic. To that end, the biennial HIVR4P conference is dedicated to all biomedical HIV prevention research approaches, including HIV vaccines, microbicides, pre-exposure prophylaxis, and treatment as prevention. The HIVR4P 2016 conference was held in Chicago, Illinois (USA), on October 17-21, and included more than 700 scientific presentations and 21 satellite sessions covering the latest and most promising advances across the HIV prevention research field. The theme "Partnering for Prevention" represented the conference's commitment to breaking down silos between research disciplines as well as between researchers, program developers, care providers, advocates, communities, and funders. Delegates spanning 42 countries attended the conference. One-third of those in attendance were early career investigators, which reflects a firm commitment to emerging researchers and ultimately to the goal of developing a sustainable scientific enterprise well into the future. This article presents a concise summary of highlights from the conference.
