Serial dilution via surface energy trap-assisted magnetic droplet manipulation.

ABSTRACT: This paper demonstrates a facile method of generating precise serial dilutions in the form of droplets on an open surface platform. The method relies on the use of surface energy traps (SETs), etched areas of high surface energy on a Teflon coated glass substrate, to assist in the magnetic manipulation of droplets to meter and dispense liquid of defined volumes for the preparation of serial dilutions. The volume of the dispensed liquid can be precisely controlled by the size of the SETs, facilitating generation of concentration profiles of high linearity. We have applied this approach to the generation of serial dilutions of antibiotics for anti-microbial susceptibility testing (AST).

Magnetic-Responsive Bendable Nozzles for Open Surface Droplet Manipulation.

Project description:The handling of droplets in a controlled manner is essential to numerous technological and scientific applications. In this work, we present a new open-surface platform for droplet manipulation based on an array of bendable nozzles that are dynamically controlled by a magnetic field. The actuation of these nozzles is possible thanks to the magnetically responsive elastomeric composite which forms the tips of the nozzles; this is fabricated with Fe3O4 microparticles embedded in a polydimethylsiloxane matrix. The transport, mixing, and splitting of droplets can be controlled by bringing together and separating the tips of these nozzles under the action of a magnet. Additionally, the characteristic configuration for droplet mixing in this platform harnesses the kinetic energy from the feeding streams; this provided a remarkable reduction of 80% in the mixing time between drops of liquids about eight times more viscous than water, i.e., 6.5 mPa/s, when compared against the mixing between sessile drops of the same fluids.

Programmable droplet manipulation by a magnetic-actuated robot.

Project description:Droplet manipulations are fundamental to numerous applications, such as water collection, medical diagnostics, and drug delivery. Structure-based liquid operations have been widely used both in nature and in artificial materials. However, current strategies depend mainly on fixed structures to realize unidirectional water movement, while multiple manipulation of droplets is still challenging. Here, we propose a magnetic-actuated robot with adjustable structures to achieve programmable multiple manipulations of droplets. The adjustable structure redistributes the resisting forces from the front and rear ends of the droplets, which determine the droplet behaviors. We can transport, split, release, and rotate the droplets using the robot. This robot is universally applicable for manipulation of various fluids in rough environments. These findings offer an efficient strategy for automated manipulation of droplets.

Multifunctional Magnetocontrollable Superwettable-Microcilia Surface for Directional Droplet Manipulation.

Project description:In nature, fluid manipulations are ubiquitous in organisms, and they are crucial for many of their vital activities. Therefore, this process has also attracted widescale research attention. However, despite significant advances in fluid transportation research over the past few decades, it is still hugely challenging to achieve efficient and nondestructive droplet transportation owing to contamination effects and controllability problems in liquid transportation applications. To this end, inspired by the motile microcilia of micro-organisms, the superhydrophobicity of lotus leaves, the underwater superoleophobicity of filefish skin, and pigeons' migration behavior, a novel manipulation strategy is developed for droplets motion. Specifically, herein, a superwettable magnetic microcilia array surface with a structure that is switchable by an external magnetic field is constructed for droplet manipulation. It is found that under external magnetic fields, the superhydrophobic magnetic microcilia array surface can continuously and directionally manipulate the water droplets in air and that the underwater superoleophobic magnetic microcilia array surface can control the oil droplets underwater. This work demonstrates that the nondestructive droplet transportation mechanism can be used for liquid transportation, droplet reactions, and micropipeline transmission, thus opening up an avenue for practical applications of droplet manipulation using intelligent microstructure surfaces.

Magnetic Janus origami robot for cross-scale droplet omni-manipulation.

Project description:The versatile manipulation of cross-scale droplets is essential in many fields. Magnetic excitation is widely used for droplet manipulation due to its distinguishing merits. However, facile magnetic actuation strategies are still lacked to realize versatile multiscale droplet manipulation. Here, a type of magnetically actuated Janus origami robot is readily fabricated for versatile cross-scale droplet manipulation including three-dimensional transport, merging, splitting, dispensing and release of daughter droplets, stirring and remote heating. The robot allows untethered droplet manipulation from ~3.2 nL to ~51.14 μL. It enables splitting of droplet, precise dispensing (minimum of ~3.2 nL) and release (minimum of ~30.2 nL) of daughter droplets. The combination of magnetically controlled rotation and photothermal properties further endows the robot with the ability to stir and heat droplets remotely. Finally, the application of the robot in polymerase chain reaction (PCR) is explored. The extraction and purification of nucleic acids can be successfully achieved.

Microfluidic serial dilution circuit.

Project description:In vitro evolution of RNA molecules requires a method for executing many consecutive serial dilutions. To solve this problem, a microfluidic circuit has been fabricated in a three-layer glass-PDMS-glass device. The 400-nL serial dilution circuit contains five integrated membrane valves: three two-way valves arranged in a loop to drive cyclic mixing of the diluent and carryover, and two bus valves to control fluidic access to the circuit through input and output channels. By varying the valve placement in the circuit, carryover fractions from 0.04 to 0.2 were obtained. Each dilution process, which is composed of a diluent flush cycle followed by a mixing cycle, is carried out with no pipeting, and a sample volume of 400 nL is sufficient for conducting an arbitrary number of serial dilutions. Mixing is precisely controlled by changing the cyclic pumping rate, with a minimum mixing time of 22 s. This microfluidic circuit is generally applicable for integrating automated serial dilution and sample preparation in almost any microfluidic architecture.

Switchable Wettability and Adhesion of Micro/Nanostructured Elastomer Surface via Electric Field for Dynamic Liquid Droplet Manipulation.

Project description:Dynamic control of liquid wetting behavior on smart surfaces has attracted considerable concern owing to their important applications in directional motion, confined wetting and selective separation. Despite much progress in this regard, there still remains challenges in dynamic liquid droplet manipulation with fast response, no loss and anti-contamination. Herein, a strategy to achieve dynamic droplet manipulation and transportation on the electric field adaptive superhydrophobic elastomer surface is demonstrated. The superhydrophobic elastomer surface is fabricated by combining the micro/nanostructured clusters of hydrophobic TiO2 nanoparticles with the elastomer film, on which the micro/nanostructure can be dynamically and reversibly tuned by electric field due to the electric field adaptive deformation of elastomer film. Accordingly, fast and reversible transition of wetting state between Cassie state and Wenzel state and tunable adhesion on the surface via electric field induced morphology transformation can be obtained. Moreover, the motion states of the surface droplets can be controlled dynamically and precisely, such as jumping and pinning, catching and releasing, and controllable liquid transfer without loss and contamination. Thus this work would open the avenue for dynamic liquid manipulation and transportation, and gear up the broad application prospects in liquid transfer, selective separation, anti-fog, anti-ice, microfluidics devices, etc.

Paired serial limiting dilution assays.

Project description:Serial limiting dilution (SLD) assays are a widely used tool in many areas of public health research to measure the concentration of target entities. This concentration can be estimated via maximum likelihood. Asymptotic as well as exact inference methods have been proposed for hypothesis testing and confidence interval construction in this one-sample problem. However, in many scientific applications, it may be of interest to compare the concentration of target entities between a pair of samples and construct valid confidence intervals for the difference in concentrations. In this paper, an exact, computationally efficient inferential procedure is proposed for hypothesis testing and confidence interval construction in the two-sample SLD assay problem. The proposed exact method is compared to an approach based on asymptotic approximations in simulation studies. The methods are illustrated using data from the University of North Carolina HIV Cure Center.

Fundamentals of magnet-actuated droplet manipulation on an open hydrophobic surface.

Project description:We systematically investigate droplet movement, coalescence, and splitting on an open hydrophobic surface. These processes are actuated by magnetic beads internalized in an oil-coated aqueous droplet using an external magnet. Results are organized into an 'operating diagram' that describes regions of droplet stable motion, breakage, and release from the magnet. The results are explained theoretically with a simple model that balances magnetic, friction, and capillary-induced drag forces and includes the effects of particle type, droplet size, surrounding oil layer, surface tension, and viscosity. Finally, we discuss the implications of the results for the design of magnet-actuated droplet systems for applications such as nucleic acid purification, immunoassay and drug delivery.
