Determining elasticity from single polymer dynamics.

ABSTRACT: The ability to determine polymer elasticity and force-extension relations from polymer dynamics in flow has been challenging, mainly due to difficulties in relating equilibrium properties such as free energy to far-from-equilibrium processes. In this work, we determine polymer elasticity from the dynamic properties of polymer chains in fluid flow using recent advances in statistical mechanics. In this way, we obtain the force-extension relation for DNA from single molecule measurements of polymer dynamics in flow without the need for optical tweezers or bead tethers. We further employ simulations to demonstrate the practicality and applicability of this approach to the dynamics of complex fluids. We investigate the effects of flow type on this analysis method, and we develop scaling laws to relate the work relation to bulk polymer viscometric functions. Taken together, our results show that nonequilibrium work relations can play a key role in the analysis of soft material dynamics.

Determining Trap Compliances, Microsphere Size Variations, and Response Linearities in Single DNA Molecule Elasticity Measurements with Optical Tweezers.

Project description:We previously introduced the use of DNA molecules for calibration of biophysical force and displacement measurements with optical tweezers. Force and length scale factors can be determined from measurements of DNA stretching. Trap compliance can be determined by fitting the data to a nonlinear DNA elasticity model, however, noise/drift/offsets in the measurement can affect the reliability of this determination. Here we demonstrate a more robust method that uses a linear approximation for DNA elasticity applied to high force range (25-45 pN) data. We show that this method can be used to assess how small variations in microsphere sizes affect DNA length measurements and demonstrate methods for correcting for these errors. We further show that these measurements can be used to check assumed linearities of system responses. Finally, we demonstrate methods combining microsphere imaging and DNA stretching to check the compliance and positioning of individual traps.

Cytoskeleton-inspired artificial protein design to enhance polymer network elasticity.

Project description:Reducing topological network defects to enhance elasticity in polymeric materials remains a grand challenge. Efforts to control network topology, primarily focused on crosslinking junctions, continue to underperform compared to theoretical estimations from idealized networks using affine and phantom network theories. Here, artificial protein technology was adapted for the design of polymer-network hydrogels with precisely defined coil-like and rod-like strands to observe the impact of strand rigidity on the mechanical properties of polymeric materials. Cytoskeleton-inspired polymer-network hydrogels incorporated with rod-like protein strands nearly tripled the gel shear elastic modulus and relaxation time compared to coil-like protein strands, indicating an enhanced effective crosslinking density. Furthermore, asymmetric rod-coil protein designs in network strands with an optimal rod:coil ratio improved the hydrogel relaxation time, enhancing the stability of physical macromolecular associations by modulating crosslinker mobility. The careful design of strand rigidity presents a new direction to reduce topological defects for optimizing polymeric materials.

Shape memory polymer network with thermally distinct elasticity and plasticity.

Project description:Stimuli-responsive materials with sophisticated yet controllable shape-changing behaviors are highly desirable for real-world device applications. Among various shape-changing materials, the elastic nature of shape memory polymers allows fixation of temporary shapes that can recover on demand, whereas polymers with exchangeable bonds can undergo permanent shape change via plasticity. We integrate the elasticity and plasticity into a single polymer network. Rational molecular design allows these two opposite behaviors to be realized at different temperature ranges without any overlap. By exploring the cumulative nature of the plasticity, we demonstrate easy manipulation of highly complex shapes that is otherwise extremely challenging. The dynamic shape-changing behavior paves a new way for fabricating geometrically complex multifunctional devices.

Effect of Polymer Gel Elasticity on Complex Coacervate Phase Behavior.

Project description:Gels are key materials in biological systems such as tissues and may control biocondensate formation and structure. To further understand the effects of elastic environments on biomacromolecular assembly, we have investigated the phase behavior and radii of complex coacervate droplets in polyacrylamide (PAM) networks as a function of gel modulus. Poly-l-lysine (PLL) and sodium hyaluronate (HA) complex coacervate phases were prepared in PAM gels with moduli varying from 0.035 to 15.0 kPa. The size of the complex coacervate droplets is reported from bright-field microscopy and confocal fluorescence microscopy. Overall, the complex coacervate droplet volume decreases inversely with the modulus. Fluorescence microscopy is also used to determine the phase behavior and concentration of fluorescently tagged HA in the complex coacervate phases as a function of ionic strength (100-270 mM). We find that the critical ionic strength and complex coacervate stability are nonmonotonic as a function of the network modulus and that the local gel concentration can be used to control phase behavior and complex coacervate droplet size scale. By understanding how elastic environments influence simple electrostatic assembly, we can further understand how biomacromolecules exist in complex, crowded, and elastic cellular environments.

Effect of Chain Polydispersity on the Elasticity of Disordered Polymer Networks.

Project description:Due to their unique structural and mechanical properties, randomly cross-linked polymer networks play an important role in many different fields, ranging from cellular biology to industrial processes. In order to elucidate how these properties are controlled by the physical details of the network (e.g., chain-length and end-to-end distributions), we generate disordered phantom networks with different cross-linker concentrations C and initial densities ρinit and evaluate their elastic properties. We find that the shear modulus computed at the same strand concentration for networks with the same C, which determines the number of chains and the chain-length distribution, depends strongly on the preparation protocol of the network, here controlled by ρinit. We rationalize this dependence by employing a generic stress-strain relation for polymer networks that does not rely on the specific form of the polymer end-to-end distance distribution. We find that the shear modulus of the networks is a nonmonotonic function of the density of elastically active strands, and that this behavior has a purely entropic origin. Our results show that if short chains are abundant, as it is always the case for randomly cross-linked polymer networks, the knowledge of the exact chain conformation distribution is essential for correctly predicting the elastic properties. Finally, we apply our theoretical approach to literature experimental data, qualitatively confirming our interpretations.

Tyramine functionalization of poly(glycerol sebacate) increases the elasticity of the polymer.

Project description:Poly(glycerol sebacate) (PGS) is an elastomer used widely in tissue engineering studies due to good biocompatibility. Hereby we report a tyramine functionalized PGS called PGS-TA. Tyramine adds a stronger physical bonding capability to PGS-TA. Tensile tests showed that the softness and toughness of the material were similar to PGS. However, PGS-TA demonstrated 16-folds increase of elastic deformations compared to PGS processed under identical conditions. The in vitro studies demonstrated that the viability, and metabolic activity of baboon smooth muscle cells were the same as those on tissue culture polystyrene. Porous subcutaneous implants of PGS-TA substantially degraded in vivo over two weeks, showing good biodegradability and biocompatibility. We expect PGS-TA to be useful for applications in tissues and organs that are subjected to large reversible mechanical deformations.

Entropic elasticity controls nanomechanics of single tropocollagen molecules.

Project description:We report molecular modeling of stretching single molecules of tropocollagen, the building block of collagen fibrils and fibers that provide mechanical support in connective tissues. For small deformation, we observe a dominance of entropic elasticity. At larger deformation, we find a transition to energetic elasticity, which is characterized by first stretching and breaking of hydrogen bonds, followed by deformation of covalent bonds in the protein backbone, eventually leading to molecular fracture. Our force-displacement curves at small forces show excellent quantitative agreement with optical tweezer experiments. Our model predicts a persistence length xi(p) approximately 16 nm, confirming experimental results suggesting that tropocollagen molecules are very flexible elastic entities. We demonstrate that assembly of single tropocollagen molecules into fibrils significantly decreases their bending flexibility, leading to decreased contributions of entropic effects during deformation. The molecular simulation results are used to develop a simple continuum model capable of describing an entire deformation range of tropocollagen molecules. Our molecular model is capable of describing different regimes of elastic and permanent deformation, without relying on empirical parameters, including a transition from entropic to energetic elasticity.

Thee-Dimensional Single-Track-Location Shear Wave Elasticity Imaging.

Project description:Conventional multiple-track-location shear wave elasticity imaging (MTL-SWEI) is a powerful tool for noninvasively estimating tissue elasticity. The resolution and noise levels of MTL-SWEI systems, however, are limited by ultrasound speckle. Single-track-location SWEI (STL-SWEI) is a novel variant which fixes the position of the tracking beam and modulates the push location to effectively cancel out the effects of speckle-induced bias. We present here a 3-D STL-SWEI system, which provides full suppression of lateral and elevation speckle bias for high-resolution volumetric elasticity imaging, and requires no spatial smoothing to make accurate measurements of shear wave speed. We demonstrate and analyze the system's performance in homogeneous and layered elasticity phantoms.

Single crystal elasticity of natural topaz at high-temperatures.

Project description:Topaz is an aluminosilicate mineral phase stable in the hydrothermally altered pegmatitic rocks and also in subducted sedimentary lithologies. In nature, topaz often exhibits solid solution between fluorine and hydrous end members. We investigated elasticity of naturally occurring single crystal topaz (Al2SiO4F1.42(OH)0.58) using Resonant Ultrasound Spectroscopy. We also explored the temperature dependence of the full elastic constant tensor. We find that among the various minerals stable in the Al2O3-SiO2-H2O ternary system, topaz exhibits moderate elastic anisotropy. As a function of temperature, the sound velocity of topaz decreases with [Formula: see text] and [Formula: see text] being -3.10 and -2.30 × 10-4 km/s/K. The elasticity and sound velocity of topaz also vary as a function of OH and F content. The effect of composition ([Formula: see text]) on the velocity is equally important as that of the effect of temperature. We also note that the Debye temperature ([Formula: see text]) of topaz at room temperature condition is 910 K and decreases at higher temperature. The Debye temperature shows positive correlation with density of the mineral phases in the Al2O3-SiO2-H2O ternary system.

Nonlinear elasticity and damping govern ultrafast dynamics in click beetles.

Project description:Many small animals use springs and latches to overcome the mechanical power output limitations of their muscles. Click beetles use springs and latches to bend their bodies at the thoracic hinge and then unbend extremely quickly, resulting in a clicking motion. When unconstrained, this quick clicking motion results in a jump. While the jumping motion has been studied in depth, the physical mechanisms enabling fast unbending have not. Here, we first identify and quantify the phases of the clicking motion: latching, loading, and energy release. We detail the motion kinematics and investigate the governing dynamics (forces) of the energy release. We use high-speed synchrotron X-ray imaging to observe and analyze the motion of the hinge's internal structures of four Elater abruptus specimens. We show evidence that soft cuticle in the hinge contributes to the spring mechanism through rapid recoil. Using spectral analysis and nonlinear system identification, we determine the equation of motion and model the beetle as a nonlinear single-degree-of-freedom oscillator. Quadratic damping and snap-through buckling are identified to be the dominant damping and elastic forces, respectively, driving the angular position during the energy release phase. The methods used in this study provide experimental and analytical guidelines for the analysis of extreme motion, starting from motion observation to identifying the forces causing the movement. The tools demonstrated here can be applied to other organisms to enhance our understanding of the energy storage and release strategies small animals use to achieve extreme accelerations repeatedly.
