Poly(ethylene imine)s as antimicrobial agents with selective activity.

ABSTRACT: We report the structure-activity relationship in the antimicrobial activity of linear and branched poly(ethylene imine)s (L- and B-PEIs) with a range of molecular weights (MWs) (500-12,000). Both L- and B-PEIs displayed enhanced activity against Staphylococcus aureus over Escherichia coli. Both B- and L-PEIs did not cause any significant permeabilization of E. coli cytoplasmic membrane. L-PEIs induced depolarization of S. aureus membrane although B-PEIs did not. The low MW B-PEIs caused little or no hemolysis while L-PEIs are hemolytic. The low MW B-PEIs are less cytotoxic to human HEp-2 cells than other PEIs. However, they induced significant cell viability reduction after 24 h incubation. The results presented here highlight the interplay between polymer size and structure on activity.

Project description:The interaction of the G-2 poly(ethylene imine) dendrimer L, derived from ammonia as initiating core, with Hg(II) and HgCl42- was studied in aqueous solution by means of potentiometric (pH-metric) measurements. Speciation of these complex systems showed that L is able to form a wide variety of complexes including 1:1, 2:1, 3:1 and 3:2 metal-to-ligand species, of different protonation states, as well as the anion complexes [(H7L)HgCl4]5+ and [(H8L)HgCl4]6+. The stability of the metal complexes is very high, making L an excellent sequestering agent for Hg(II), over a large pH range, and a promising ligand for the preparation of functionalized activated carbons to be employed in the remediation and the prevention of environmental problems.

Grafting of Poly(ethylene imine) to Silica Nanoparticles for Odor Removal from Recycled Materials.

Project description:One of the major obstacles to the reuse of recycled plastic materials is the emanation of after-process odors from recycled polymers and composites. Typically, recycled polymers are blended with an off-odor adsorbent additive in the recycling chain to eliminate these smells. This article describes an innovative ultrasonically assisted method of grafting poly(ethylene imine) (PEI) to silica nanoparticles (SiO2) initiated by benzoyl peroxide (BP) which acts as an odor remover. To prepare the PEI/Si, the branched PEI was grafted onto the silica surface without a coupling agent. This made the grafting process straightforward, easy and low in cost. Fourier Transform Infrared (FTIR) analysis confirmed the successful grafting of PEI to silica. The thermogravimetric analysis (TGA) indicated the formation of two different fractions: a polymeric fraction covalently attached to the nanoparticle surface and a non-grafted PEI fraction that was removed during extraction. Up to 30% of the grafted-PEI fractions were produced at the lowest BP concentration with the highest PEI molecular weight at silica-to-PEI weight ratios of (1:1) to (3:1). The sensory assessment showed a substantial reduction in overall odor intensity for 30% of the recycled plastic-containing materials and a ~75% reduction in volatile organic compounds (VOCs) for 100% of the recycled plastics. These results strongly suggest that this innovative PEI/Si nanocomposite can be successfully commercialized for odor removal. To the authors' best knowledge, this is the first reported work describing a one-pot reaction for grafting PEI to different nanoparticle surfaces.

Poly(ethylene-imine)-Functionalized Magnetite Nanoparticles Derivatized with Folic Acid: Heating and Targeting Properties.

Project description:Magnetite nanoparticles (MNPs) coated by branched poly (ethylene-imine) (PEI) were synthesized in a one-pot. Three molecular weights of PEI were tested, namely, 1.8 kDa (sample MNP-1), 10 kDa (sample MNP-2), and 25 kDa (sample MNP-3). The MNP-1 particles were further functionalized with folic acid (FA) (sample MNP-4). The four types of particles were found to behave magnetically as superparamagnetic, with MNP-1 showing the highest magnetization saturation. The particles were evaluated as possible hyperthermia agents by subjecting them to magnetic fields of 12 kA/m strength and frequencies ranging between 115 and 175 kHz. MNP-1 released the maximum heating power, reaching 330 W/g at the highest frequency, in the high side of reported values for spherical MNPs. In vitro cell viability assays of MNP-1 and MNP-4 against three cell lines expressing different levels of FA receptors (FR), namely, HEK (low expression), and HeLa (high expression), and HepG2 (high expression), demonstrated that they are not cytotoxic. When the cells were incubated in the presence of a 175 kHz magnetic field, a significant reduction in cell viability and clone formation was obtained for the high expressing FR cells incubated with MNP-4, suggesting that MNP-4 particles are good candidates for magnetic field hyperthermia and active targeting.

Nanocarriers for Protein Delivery to the Cytosol: Assessing the Endosomal Escape of Poly(Lactide-co-Glycolide)-Poly(Ethylene Imine) Nanoparticles.

Project description:Therapeutic proteins and enzymes are a group of interesting candidates for the treatment of numerous diseases, but they often require a carrier to avoid degradation and rapid clearance in vivo. To this end, organic nanoparticles (NPs) represent an excellent choice due to their biocompatibility, and cross-linked enzyme aggregates (CLEAs)-loaded poly (lactide-co-glycolide) (PLGA) NPs have recently attracted attention as versatile tools for targeted enzyme delivery. However, PLGA NPs are taken up by cells via endocytosis and are typically trafficked into lysosomes, while many therapeutic proteins and enzymes should reach the cellular cytosol to perform their activity. Here, we designed a CLEAs-based system implemented with a cationic endosomal escape agent (poly(ethylene imine), PEI) to extend the use of CLEA NPs also to cytosolic enzymes. We demonstrated that our system can deliver protein payloads at cytoplasm level by two different mechanisms: Endosomal escape and direct translocation. Finally, we applied this system to the cytoplasmic delivery of a therapeutically relevant enzyme (superoxide dismutase, SOD) in vitro.

Chromium-Based Complexes Bearing Aminophosphine and Phosphine-Imine-Pyrryl Ligands for Selective Ethylene Tri/Tetramerization.

Project description:A series of Cr-based complexes 6-10 bearing aminophosphine (P,N) ligands Ph2P-L-NH2 [L = CH2CH2 (1), L = CH2CH2CH2 (2), and L = C6H4CH2 (3)] and phosphine-imine-pyrryl (P,N,N) ligands 2-(Ph2P-L-N=CH)C4H3NH [L = CH2CH2CH2 (4) and L = C6H4CH2 (5)] were prepared, and their catalytic properties were examined for ethylene tri/tetramerization. X-ray crystallographic analysis of complex 8 indicated the κ2-P,N bidentate coordination mode at the Cr(III) center and the distorted octahedral geometry of monomeric P,N-CrCl3. Upon activation by methylaluminoxane (MAO), complexes 7-8 bearing P,N (PC3N backbone) ligands 2-3 showed good catalytic reactivity for ethylene tri/tetramerization. On the other hand, complex 6 bearing the P,N (PC2N backbone) ligand 1 was found active for non-selective ethylene oligomerization, while complexes 9-10 bearing P,N,N ligands 4-5 only produced polymerization products. In particular, the high catalytic activity of 458.2 kg/(g·Cr·h), excellent selectivity of 90.9% (1-hexene and 1-octene combined), and extremely low PE content of 0.1% were obtained with complex 7 in toluene at 45 °C and 45 bar. These results suggest that rational control of P,N and P,N,N ligand backbones, including a carbon spacer and rigidity of a carbon bridge, can lead to the high-performance catalyst for the ethylene tri/tetramerization process.

Cation, Anion and Ion-Pair Complexes with a G-3 Poly(ethylene imine) Dendrimer in Aqueous Solution.

Project description:The G-3 poly(ethylene imine) ligand L2 shows a multifaceted coordination ability, being able to bind metal cations, anions and ion-pairs. The equilibrium constants for the formation of metal (Cu2+, Zn2+), anion (SO₄2-) and ion-pair (Cu2+/SO₄2-) complexes were determined in 0.1 M Me₄NCl aqueous solution at 298.1 ± 0.1 K by means of potentiometric titrations. Thanks to its dendrimeric nature, L2 can form highly nucleated metal complexes, such as Cu₅L210+ and Zn₄L28+, in successive and well-defined complexation steps. Protonated forms of L2 give rise to relatively weak anion complexes with SO₄2-, but the addition of Cu2+ significantly enhances the binding ability of the ligand toward this anion below pH 9. In more alkaline solutions, an opposite trend is observed. The coordination properties of L2 are discussed with the support of modelling calculations. According to results, L2 is a promising molecule for the preparation of solid supported materials for the recovery of cations and anions from aqueous media and/or for applications in heterogeneous catalysis.

Poly(ethylene imine)-chitosan carbon dots: study of its physical-chemical properties and biological in vitro performance.

Project description:Carbon dots (CDs) have been quickly extended for nanomedicine uses because of their multiple applications, such as bioimaging, sensors, and drug delivery. However, the interest in increasing their photoluminescence properties is not always accompanied by cytocompatibility. Thus, a knowledge gap exists regarding their interactions with biological systems linked to the selected formulations and synthesis methods. In this work, we have developed carbon dots (CDs) based on poly (ethylene imine) (PEI) and chitosan (CS) by using microwave irradiation, hydrothermal synthesis, and a combination of both, and further characterized them by physicochemical and biological means. Our results indicate that synthesized CDs have sizes between 1 and 5 nm, a high presence of amine groups on the surface, and increased positive ζ potential values. Further, it is established that the choice and use of different synthesis procedures can contribute to a different answer to the CDs regarding their optical and biological properties. In this regard, PEI-only CDs showed the longest photoluminescent emission lifetime, non-hemolytic activity, and high toxicity against fibroblast. On the other hand, CS-only CDs have higher PL emission, non-cytotoxicity associated with fibroblast, and high hemolytic activity. Interestingly, their combination using the proposed methodologies allow a synergic effect in their CDs properties. Therefore, this work contributes to developing and characterizing CD formulations based on PEI and CS and better understanding the CD's properties and biological interaction.

Actinide Ion (Americium-241 and Uranium-232) Interaction with Hybrid Silica-Hyperbranched Poly(ethylene imine) Nanoparticles and Xerogels.

Project description:The binding of actinide ions (Am(III) and U(VI)) in aqueous solutions by hybrid silica-hyperbranched poly(ethylene imine) nanoparticles (NPs) and xerogels (XGs) has been studied by means of batch experiments at different pH values (4, 7, and 9) under ambient atmospheric conditions. Both materials present relatively high removal efficiency at pH 4 and pH 7 (>70%) for Am(III) and U(VI). The lower removal efficiency for the nanoparticles is basically associated with the compact structure of the nanoparticles and the lower permeability and access to active amine groups compared to xerogels, and the negative charge of the radionuclide species is formed under alkaline conditions (e.g., UO2(CO3)34- and Am(CO3)2-). Generally, the adsorption process is relatively slow due to the very low radionuclide concentrations used in the study and is basically governed by the actinide diffusion from the aqueous phase to the solid surface. On the other hand, adsorption is favored with increasing temperature, assuming that the reaction is endothermic and entropy-driven, which is associated with increasing randomness at the solid-liquid interphase upon actinide adsorption. To the best of our knowledge, this is the first study on hybrid silica-hyperbranched poly(ethylene imine) nanoparticle and xerogel materials used as adsorbents for americium and uranium at ultra-trace levels. Compared to other adsorbent materials used for binding americium and uranium ions, both materials show far higher binding efficiency. Xerogels could remove both actinides even from seawater by almost 90%, whereas nanoparticles could remove uranium by 80% and americium by 70%. The above, along with their simple derivatization to increase the selectivity towards a specific radionuclide and their easy processing to be included in separation technologies, could make these materials attractive candidates for the treatment of radionuclide/actinide-contaminated water.

PEGylation of silver nanoparticles by physisorption of cyclic poly(ethylene glycol) for enhanced dispersion stability, antimicrobial activity, and cytotoxicity.

Project description:Silver nanoparticles (AgNPs) are practically valuable in biological applications. However, no steady PEGylation has been established, which is essential for internal use in humans or animals. In this study, cyclic PEG (c-PEG) without any chemical inhomogeneity is physisorbed onto AgNPs to successfully PEGylate and drastically enhance the dispersion stability against physiological conditions, white light, and high temperature. In contrast, linear HO-PEG-OH and MeO-PEG-OMe do not confer stability to AgNPs, and HS-PEG-OMe, which is often used for gold nanoparticles, sulfidates the surface to considerably degrade the properties. TEM shows an essentially intact nanostructure of c-PEG-physisorbed AgNPs even after heating at 95 °C, while complete disturbance is observed for other AgNPs. Molecular weight- and concentration-dependent stabilization by c-PEG is investigated, and DLS and ζ-potential measurements prove the formation of a c-PEG layer on the surface of AgNPs. Furthermore, c-PEG-physisorbed AgNPs exhibit persistent antimicrobial activity and cytotoxicity.

Surface Modification of Poly(amidoamine) (PAMAM) Dendrimer as Antimicrobial Agents.

Project description:Poly(amidoamine) (PAMAM) (G3) dendrimer was modified into quaternary ammonium salts using tertiary amines with different chain lengths: dimethyldodecyl amine, dimethylhexyl amine, and dimethylbutyl amine using an efficient synthetic route. The antimicrobial activity of these dendrimer ammonium salts against Staphylococcus and E-coli bacteria was examined using the disc diffusion method. It was found that quaternary ammonium salt prepared with the dimethyldodecyl amine exhibits antimicrobial efficacy against Staphalococus and E.coli bacteria.
