Draft Genome Sequence of Raoultella ornithinolytica TNT, a Trinitrotoluene-Denitrating and Plant Growth-Promoting Strain Isolated from Explosive-Contaminated Soil.

ABSTRACT: We report the draft genome of Raoultella ornithinolytica TNT, a Gram-negative bacterium of the Enterobacteriaceae isolated from military soil in Belgium. Strain TNT uses nitrite released from trinitrotoluene (TNT) for growth and is a potent plant growth promoter. An analysis of its 5.6-Mb draft genome will bring insights into TNT degradation-reinforcing bioremediation applications.

Project description:We report the whole-genome sequence of Gordonia sp. strain YY1, which was isolated from the surface soil in an explosive-contaminated site in Israel and cultivated with hexahydro-1,3,5-trinitro-1,3,5-triazine, i.e., royal demolition explosive (RDX), as a nitrogen source. This genome sequence will improve our understanding of the genes for RDX degradation. In addition, this research will reveal metabolic pathways in order to develop new bioremediation methods for polluted soil and groundwater.

Draft Genome Sequences of Rhodococcus sp. Strains YH1 and T7, Isolated from Explosive-Contaminated Environments.

Project description:We report the draft genome sequences for Rhodococcus sp. strains YH1 and T7. These strains are both capable of degrading hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX) and were isolated from explosive-contaminated soil and groundwater, respectively. Further genomic analysis might facilitate an understanding of the degradation of RDX and will contribute to the development of bioremediation methods for polluted soil and groundwater.

<i>Raoultella ornithinolytica</i> as a Potential Candidate for Bioremediation of Heavy Metal from Contaminated Environments.

Project description:Disposal of waste containing heavy metals into the environment is a major threat to human health and can result in toxic or chronic poisoning in aquatic life. In the current study, metal-resistant Raoultella ornithinolytica was isolated from metal-contaminated samples collected from the Tanjaro River, located southwest of Sulaymaniyah, Iraq. R. ornithinolytica was identified by partial amplification of 16S rRNA. The uptake potency of heavy metals was assessed using inductively coupled plasma-optical emission spectroscopy (ICP-OES) and indicated that R. ornithinolytica removed 67, 89, 63.4, 55.6, 56.5, 65, and 61.9% of Cd, Pb, Cr, Ni, Zn, Co, and Fe, respectively. These removal rates were influenced by temperature, pH, and contact time; at 35°C and pH 5 with a change in the incubation time, the reduction rate improved from 89 to 95% for Pb, from 36.4 to 45% for Cu, and from 55.6 to 64% for Ni. Gene analysis indicated that R. ornithinolytica contained pbrT, chrB, nccA, iroN, and czcA genes, but the pcoD gene was absent. Energy-dispersive X-ray spectroscopy (EDS) images showed evidence of metal ion binding on the cell wall surface with different rates of binding. Transmission electron microscopy (TEM) detected different mechanisms for metal particle localization; cell surface adsorption was the main mechanism for Pb, Zn, and Co uptake, while Cd, Ni, and Fe were accumulated inside the cell. The current study describes, for the first time, the isolation of R. ornithinolytica from metal-contaminated water, which can be used as an eco-friendly biological expedient for the remediation and detoxification of metals from contaminated environments.

Data on biosurfactant assisted removal of TNT from contaminated soil.

Project description:Contamination of environment, especially soil, is in great concern and can cause health problems. Thus, remediation of these pollutants through environmentally friendly methods should be considered. The aim of this data was bioremediation of TNT from contaminated soil. Two plastic pans were used as bioreactor. In each pan, 3 kg of soil was used. Concentration of TNT in contaminated soil was 1000 mg/kg. Rhamnolipid in concentration of 60 mg/l was added to intended pan. Sampling was done in each two weeks. In order to assessment of TNT degradation, samples were analyzed with HPLC. The data showed that after 154 days of experiment, TNT removal in soil that amended with rhamnolipid was 73% and in experiment with no addition of rhamnolipid was 58%. Based on the obtained data rhamnolipid was effective in remediation of TNT contaminated soil.

Explosive detonation causes an increase in soil porosity leading to increased TNT transformation.

Project description:Explosives are a common soil contaminant at a range of sites, including explosives manufacturing plants and areas associated with landmine detonations. As many explosives are toxic and may cause adverse environmental effects, a large body of research has targeted the remediation of explosives residues in soil. Studies in this area have largely involved spiking 'pristine' soils using explosives solutions. Here we investigate the fate of explosives present in soils following an actual detonation process and compare this to the fate of explosives spiked into 'pristine' undetonated soils. We also assess the effects of the detonations on the physical properties of the soils. Our scanning electron microscopy analyses reveal that detonations result in newly-fractured planes within the soil aggregates, and novel micro Computed Tomography analyses of the soils reveal, for the first time, the effect of the detonations on the internal architecture of the soils. We demonstrate that detonations cause an increase in soil porosity, and this correlates to an increased rate of TNT transformation and loss within the detonated soils, compared to spiked pristine soils. We propose that this increased TNT transformation is due to an increased bioavailability of the TNT within the now more porous post-detonation soils, making the TNT more easily accessible by soil-borne bacteria for potential biodegradation. This new discovery potentially exposes novel remediation methods for explosive contaminated soils where actual detonation of the soil significantly promotes subsequent TNT degradation. This work also suggests previously unexplored ramifications associated with high energy soil disruption.
