ABSTRACT: Spontaneous formation of heterogeneous patches on the surface of lipid-based nanoparticles (NPs) and microparticles (MPs) due to the segregation of two different functional groups. Patch formation is observed when tracing the functional groups with quantum dots, gold nanoparticles, and fluorescent dyes. This discovery could have important implications for the future design of self-assembled NPs and MPs for different biomedical applications.

Highly effective heterogeneous chemosensors of luminescent silica@coordination polymer core-shell micro-structures for metal ion sensing.

Project description:Heterogeneous solid sensors are regarded as promising next-generation sensor due to their excellent chemical stability, low contamination, and excellent recyclability, despite their low sensitivity and weak signal. The dispersity and signals specifically from the exterior of solid sensors are critical aspects which define the sensing sensitivity and selectivity. A novel strategy for the preparation of ideal heterogeneous sensors based upon luminescent lanthanide coordination polymers (LnCP) has been demonstrated. Ideal heterogeneous sensors are systematically achieved by producing the sensors in small, uniform, and thin core-shell particles (silica@LnCP, Ln = Eu, Tb). Eventually, we found that the extremely small amount of well-structured silica@LnCP microsphere, less than ca. 1/400 compared to the amount of several known coordination polymer-based sensors, was sufficient to achieve a reliable Cu(2+) sensing with even much greater sensitivity (ca. 550% improvement).

Core-shell and segmented polymer-metal composite nanostructures.

Project description:Composite nanostructures (approximately 200 nm wide and several micrometers long) of metal and polyaniline (PANI) in two new variations of core-shell (PANI-Au) and segmented (Au-PANI and Ni-Au-PANI) architectures were fabricated electrochemically within anodized aluminum oxide (AAO) membranes. Control over the structure of these composites (including the length of the gold shells in the core-shell structures) was accomplished by adjusting the time and rate of electrodeposition and the pH of the solution from which the materials were grown. Exposure of the core-shell structures to oxygen plasma removed the PANI and yielded aligned gold nanotubes. In the segmented structures, a self-assembled monolayer (SAM) of thioaniline nucleated the growth of PANI on top of metal nanorods and acted as an adhesion layer between the metal and PANI components.

A universal polymer shell-isolated nanoparticle (SHIN) design for single particle spectro-electrochemical SERS sensing using different core shapes.

Project description:Shell-isolated nanoparticles (SHINs) have attracted increasing interest for non-interfering plasmonic enhanced sensing in fields such as materials science, biosensing, and in various electrochemical systems. The metallic core of these nanoparticles is isolated from the surrounding environment preventing direct contact or chemical interaction with the metal surface, while still being close enough to enable localized surface plasmon enhancement of the Raman scattering signal from the analyte. This concept forms the basis of the shell isolated nanoparticle-enhanced Raman spectroscopy (SHINERS) technique. To date, the vast majority of SHIN designs have focused on SiO2 shells around spherical nanoparticle cores and there has been very limited published research considering alternatives. In this article, we introduce a new polymer-based approach which provides excellent control over the layer thickness and can be applied to plasmonic metal nanoparticles of various shapes and sizes without compromising the overall nanoparticle morphology. The SHIN layers are shown to exhibit excellent passivation properties and robustness in the case of gold nanosphere (AuNP) and anisotropic gold nanostar (AuNS) core shapes. In addition, in situ SHINERS spectro-electrochemistry measurements performed on both SHIN and bare Au nanoparticles demonstrate the utility of the SHIN coatings. Correlated confocal Raman and SEM mapping was achieved to clearly establish single nanoparticle SERS sensitivity. Finally, confocal in situ SERS mapping enabled visualisation of the redox related molecular structure changes occurring on an electrode surface in the vicinity of individual SHIN-coated nanoparticles.

Spontaneous droplets gyrating via asymmetric self-splitting on heterogeneous surfaces.

Project description:Droplet impacting and bouncing off solid surface plays a vital role in various biological/physiological processes and engineering applications. However, due to a lack of accurate control of force transmission, the maneuver of the droplet movement and energy conversion is rather primitive. Here we show that the translational motion of an impacting droplet can be converted to gyration, with a maximum rotational speed exceeding 7300 revolutions per minute, through heterogeneous surface wettability regulation. The gyration behavior is enabled by the synergetic effect of the asymmetric pinning forces originated from surface heterogeneity and the excess surface energy of the spreading droplet after impact. The findings open a promising avenue for delicate control of liquid motion as well as actuating of solids.

Janus gold nanoparticles obtained via spontaneous binary polymer shell segregation.

Project description:Janus gold nanoparticles are of high interest because they allow directed self-assembly and display plasmonic properties. We succeeded in coating gold nanoparticles with two different polymers that form a Janus shell. The spontaneous segregation of two immiscible polymers at the surface of the nanoparticles was verified by NOESY NMR and most importantly by electron microscopy analysis in two and three dimensions. The Janus structure is additionally shown to affect the aggregation behavior of the nanoparticles.

Catechol Redox Induced Formation of Metal Core-Polymer Shell Nanoparticles.

Project description:A novel strategy was developed to synthesize polymer-coated metal nanoparticles (NPs) through reduction of metal cations with 3,4-dihydroxyphenylalanine (DOPA)-containing polyethylene glycol (PEG) polymers. Catechol redox chemistry was used to both synthesize metal NPs and simultaneously form a cross-linked shell of PEG polymers on their surfaces. DOPA reduced gold and silver cations into neutral metal atoms, producing reactive quinones that covalently cross-linked the PEG molecules around the surface of the NP. Importantly, these PEG-functionalized metal NPs were stable in physiological ionic strengths and under centrifugation, and hold broad appeal since they absorb and scatter light in aqueous solutions.

Fabricating Fibers of a Porous-Polystyrene Shell and Particle-Loaded Core.

Project description:Polystyrene (PS) polymers have broad applications in protective packaging for food shipping, containers, lids, bottles, trays, tumblers, disposable cutlery and the making of models. Currently, most PS products, such as foams, are not accepted for recycling due to a low density in the porous structure. This poses a challenge for logistics as well as creating a lack of incentive to invest in high-value products. This study, however, demonstrated the use of a dry-jet wet-spinning technique to manufacture continuous PS fibers enabled by an in-house designed and developed spinning apparatus. The manufactured fibers showed porosity in the shell and the capability to load particles in their core, a structure with high potential use in environmentally relevant applications such as water treatment or CO2 collections. A two-phase liquid-state microstructure was first achieved via a co-axial spinneret. Following coagulation procedures and heat treatment, phase-separation-based selective dissolution successfully generated the porous-shell/particle-core fibers. The pore size and density were controlled by the porogen (i.e., PEG) concentrations and examined using scanning electron microscopy (SEM). Fiber formation dynamics were studied via rheology tests and gelation measurements. The shell components were characterized by tensile tests, thermogravimetric analysis, and differential scanning calorimetry for mechanical durability and thermal stability analyses.

Surfaces and Interfaces of Liquid Metal Core-Shell Nanoparticles under the Microscope.

Project description:Eutectic gallium indium (EGaIn), a Ga-based liquid metal alloy holds great promise for designing next generation core-shell nanoparticles (CSNs). A shearing assisted ligand-stabilization method has shown promise as a synthetic method for these CSNs; however, determining the role of the ligand on stabilization demands an understanding of the surface chemistry of the ligand-nanoparticle interface. EGaIn CSNs have been created functionalized with aliphatic carboxylates of different chain length allowing a fundamental investigation on ligand stabilization of EGaIn CSNs. Raman and diffuse reflectance Fourier transform spectroscopies (DRIFTS) confirm reaction of the ligand with the oxide shell of the EGaIn nanoparticles. Changing the length of the alkyl chain in the aliphatic carboxylates (C2-C18) may influence the size and structural stability of EGaIn CSNs, which is easily monitored using atomic force microscopy (AFM). No matter how large the carboxylate ligand, there is no obvious effect on the size of the EGaIn CSNs, except the particle size got more uniform when coated with longer chain carboxylates. The AFM force distance (F-D) measurements are used to measure the stiffness of the carboxylate coated EGaIn CSN. In corroboration with DRIFTS analysis, the stiffness studies show that the alkyl chains undergo conformational changes upon compression.

Curcumin to Promote the Synthesis of Silver NPs and their Self-Assembly with a Thermoresponsive Polymer in Core-Shell Nanohybrids.

Project description:This work presents a simple one-pot protocol to achieve core-doped shell nanohybrids comprising silver nanoparticles, curcumin and thermoresponsive polymeric shell taking advantage of the reducing properties of phenolic curcumin substance and its ability to decorate metallic surfaces. Silver nanoparticles were synthesized, via sodium citrate and silver nitrate addition into a boiling aqueous solution of curcumin, monomers and surfactant. Curcumin and sodium citrate promoted silver nucleation, acting as reducing and stabilizing agents. These curcumin-capped AgNPs enabled, after adding the radical polymerization initiator, the assembling of the growing polymer chains around the hydrophobic AgNP surface. The resultant core-doped shell nanohybrids exhibit plasmonic, luminescent and volume thermoresponsive properties, with improved possibilities to be used as successful therapeutic platforms. In fact, the possibility to nanoconfine the synergistic antioxidant, antiviral, antibacterial features of silver and curcumin in one bioavailable hybrid paves the way to promising applications in the biomedical field.

Core shell lipid-polymer hybrid nanoparticles with combined docetaxel and molecular targeted therapy for the treatment of metastatic prostate cancer.

Project description:Many prostate cancers relapse after initial chemotherapy treatment. Combining molecular and chemotherapy together with encapsulation of drugs in nanocarriers provides effective drug delivery and toxicity reduction. We developed core shell lipid-polymer hybrid nanoparticles (CSLPHNPs) with poly (lactic-co-glycolic acid) (PLGA) core and lipid layer containing docetaxel and clinically used inhibitor of sphingosine kinase 1 (SK1) FTY720 (fingolimod). We show for the first time that FTY720 (both free and in CSLPHNPs) re-sensitizes castrate resistant prostate cancer cells and tumors to docetaxel, allowing a four-fold reduction in effective dose. Our CSLPHNPs showed high serum stability and a long shelf life. CSLPHNPs demonstrated a steady uptake by tumor cells, sustained intracellular drug release and in vitro efficacy superior to free therapies. In a mouse model of human prostate cancer, CSLPHNPs showed excellent tumor targeting and significantly lower side effects compared to free drugs, importantly, reversing lymphopenia induced by FTY720. Overall, we demonstrate that nanoparticle encapsulation can improve targeting, provide low off-target toxicity and most importantly reduce FTY720-induced lymphopenia, suggesting its potential use in clinical cancer treatment.
