Yucca-derived synthesis of gold nanomaterial and their catalytic potential.

ABSTRACT: AuNPs ranging in 20 to 300 nm size were synthesized at a room temperature using Yucca filamentosa leaf extract. Diverse nanomaterial morphologies were obtained by varying the extract concentration, reaction pH, and temperature. While low volumes of extract (0.25 and 0.5 mL) induced the formation of microscale Au sheets with edge length greater than 1 μm, high volumes yielded spherical particles ranging from 20 to 200 nm. Varying pH of the solution significantly influenced the particle shape with the production of largely spherical particles at pH 5 to 6 and truncated triangles at pH 2. Separation of multidimensional nanostructures was achieved using a novel method of sucrose density gradient centrifugation. The catalytic function of Yucca-derived AuNPs was demonstrated by degradation of a wastewater dye: methylene blue using spectrophotometric measurements over time. Treatment with Au nanosheets and spheres demonstrated methylene blue degradation approximately 100% greater than the activity in control at 60 min.

Project description:Thin metal particles with two-dimensional (2D) symmetry are attractive for multiple applications but are difficult to synthesize in a reproducible manner. Although molecules that selectively adsorb to facets have been used to control nanoparticle shape, there is still limited research into the temporal control of growth processes to control these structural outcomes. Moreover, much of the current research into the growth of thin 2D particles lacks mechanistic details. In this work, we study why the substitution of isoleucine for methionine in a gold-binding peptide (Z2, RMRMKMK) results in an increase in gold nanoparticle anisotropy. Nanoplatelet growth in the presence of Z2M246I (RIRIKIK) is characterized using in situ small-angle X-ray scattering (SAXS) and UV-vis spectroscopy. Fitting time-resolved SAXS profiles reveal that 10 nm-thick particles with 2D symmetry are formed within the first few minutes of the reaction. Next, through a combination of electron diffraction and molecular dynamics simulations, we show that substitution of methionine for isoleucine increases the (111) facet selectivity in Z2M246I, and we conclude that this is key to the growth of nanoplatelets. However, the potential application of nanoplatelets formed using Z2M246I is limited due to their uncontrolled lateral growth, aggregation, and rapid sedimentation. Therefore, we use a liquid-handling robot to perform temporally controlled synthesis and dynamic intervention through the addition of Z2 to nanoplatelets grown in the presence of Z2M246I at different times. UV-vis spectroscopy, dynamic light scattering, and electron microscopy show that dynamic intervention results in control over the mean size and stability of plate-like particles. Finally, we use in situ UV-vis spectroscopy to study plate-like particle growth at different times of intervention. Our results demonstrate that both the selectivity and magnitude of binding free energy toward lattices are important for controlling nanoparticle growth pathways.

Project description:Background:In the emerging field of nanotechnology, copper oxide (CuO) nanomaterials are considered to be one of the most important transition metal oxides owing to its fascinating properties. Its synthesis from green chemistry principles is gaining importance as next-generation antibiotics due to its simplicity, eco-friendliness, and cost-effectiveness. In the present study, CuO nanorods (CuO NRs) were synthesized from the aqueous fruit extract of Momordica charantia and characterized using different analytical techniques. Further, the biomedical therapeutic potential was evaluated against multi-drug resistant microbial strains. Materials and Methods:To synthesize CuO NRs, 0.1M of CuSO4.5H2O solution was added to aqueous extract of Momordica charantia in a 1:3 (v/v) ratio (pH=11) and heated at 50°C followed by washing and drying. The synthesized CuO NRs were subjected to characterization using different analytical techniques such as UV visible spectroscopy, zeta sizer equipped with zeta potential, Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), scanning electron microscopy (SEM) equipped with energy-dispersive X-ray spectroscopy (EDS) and transmission electron microscopy (TEM). Further, the application as a biomedical therapeutic potential was evaluated in vitro using well diffusion method against eleven multidrug-resistant clinical bacterial strains, a fungus- Trichophyton rubrum and in ovo against the R2B virus using haemagglutination (HA) test. Results:Characterization was preliminarily done by the spectral study that confirms the absorbance band at 245nm. FTIR analysis at 628 cm-1 peak identified copper oxide vibration. SEM analysis revealed agglomerated particle clusters. However, with TEM clear nanorods of average diameter of 61.48 ± 2 nm were observed. EDAX confirmed CuO formation while XRD showed a typical monoclinic structure with 6 nm crystallite size. Biological screening of CuO NRs showed significant results against both in vitro and in ovo methods. Significant inhibitory activity (p<0.0001) was noted against most of the resistant human pathogenic strains including both Gram-positive and Gram-negative bacteria. The highest efficacy was observed against Bacillus cereus with a 31.66 mm zone of inhibition. Besides, the therapeutic potential of CuO NRs against Corynebacterium xerosis, Streptococcus viridians and R2B strain of Newcastle disease is reported for the first time. Conclusion:Based on the present results, it could be expected that green synthesized CuO NRs would find potential applications in the field of nanomedicine.

Project description:The demand for safer design and synthesis of gold nanoparticles (AuNPs) is on the increase with the ultimate goal of producing clean nanomaterials for biological applications. We hereby present a rapid, greener, and photochemical synthesis of gold nanoplates with sizes ranging from 10 to 200 nm using water-soluble quercetin diphosphate (QDP) macromolecules. The synthesis was achieved in water without the use of surfactants, reducing agents, or polymers. The edge length of the triangular nanoplates ranged from 50 to 1200 nm. Furthermore, the reduction of methylene blue was used to investigate the catalytic activity of AuNPs. The catalytic activity of triangular AuNPs was three times higher than that of the spherical AuNPs based on kinetic rate constants (k). The rate constants were 3.44 × 10-2 and 1.11 × 10-2 s-1 for triangular and spherical AuNPs, respectively. The X-ray diffraction data of gold nanoplates synthesized by this method exhibited that the nanocrystals were mainly dominated by (111) facets which are in agreement to the nanoplates synthesized by using thermal and chemical approaches. The calculated relative diffraction peak intensity of (200), (220), and (311) in comparison with (111) was found to be 0.35, 0.17, and 0.15, respectively, which were lower than the corresponding standard values (JCPDS 04-0784). For example, (200)/(111) = 0.35 compared to 0.52 obtained from the standard (JCPDS 04-0784), indicating that the gold nanoplates are dominated by (111) facets. The calculated lattice from selected area electron diffraction data of the as-synthesized and after 1 year nanoplates was 4.060 and 4.088 Å, respectively. Our calculations were found to be in agreement with 4.078 Å for face-centered cubic gold (JCPDS 04-0784) and literature values of 4.07 Å. The computed QDP-Au complex demonstrated that the reduction process took place in the B ring of QDP. This approach contributes immensely to promoting the ideals of sustainable nanotechnology by eradicating the use of hazardous and toxic organic solvents.
