Project description:Mitochondrial superoxide (O2⋅-) underlies much oxidative damage and redox signaling. Fluorescent probes can detect O2⋅-, but are of limited applicability in vivo, while in cells their usefulness is constrained by side reactions and DNA intercalation. To overcome these limitations, we developed a dual-purpose mitochondrial O2⋅- probe, MitoNeoD, which can assess O2⋅- changes in vivo by mass spectrometry and in vitro by fluorescence. MitoNeoD comprises a O2⋅--sensitive reduced phenanthridinium moiety modified to prevent DNA intercalation, as well as a carbon-deuterium bond to enhance its selectivity for O2⋅- over non-specific oxidation, and a triphenylphosphonium lipophilic cation moiety leading to the rapid accumulation within mitochondria. We demonstrated that MitoNeoD was a versatile and robust probe to assess changes in mitochondrial O2⋅- from isolated mitochondria to animal models, thus offering a way to examine the many roles of mitochondrial O2⋅- production in health and disease.

Project description:We found mitoflash has function in reprogramming. So we examined the genes expression after activation of mitoflash during somatic cell reprogramming. There are two ways to improve the mitoflash. First we overexpressed CypD, a mPTP component, to improve the mitoflash. Second we used the durgs, mastoparan and paraquat to increase the mitoflash. The MEF cells were reprogrammed, and treatment with mastoparan, paraquat or overexpressed with CypD. After activation of mitoflash at different times, the cells were collected for RNA-Seq.

Project description:Here we report the synthesis and characterization of a membrane-impermeant fluorogenic probe, hydropropidine (HPr(+)), the reduction product of propidium iodide, for detecting extracellular superoxide (O(2)(•-)). HPr(+) is a positively charged water-soluble analog of hydroethidine (HE), a fluorogenic probe commonly used for monitoring intracellular O(2)(•-). We hypothesized that the presence of a highly localized positive charge on the nitrogen atom would impede cellular uptake of HPr(+) and allow for exclusive detection of extracellular O(2)(•-). Our results indicate that O(2)(•-) reacts with HPr(+) (k=1.2×10(4) M(-1) s(-1)) to form exclusively 2-hydroxypropidium (2-OH-Pr(2+)) in cell-free and cell-based systems. This reaction is analogous to the reaction between HE and O(2)(•-) (Zhao et al., Free Radic. Biol. Med.34:1359-1368; 2003). During the course of this investigation, we also reassessed the rate constants for the reactions of O(2)(•-) with HE and its mitochondria targeted analog (Mito-HE or MitoSOX Red) and addressed the discrepancies between the present values and those reported previously by us. Our results indicate that the rate constant between O(2)(•-) and HPr(+) is slightly higher than that of HE and O(2)(•-) and is closer to that of Mito-HE and O(2)(•-). Similar to HE, HPr(+) undergoes oxidation in the presence of various oxidants (peroxynitrite-derived radicals, Fenton's reagent, and ferricytochrome c) forming the corresponding propidium dication (Pr(2+)) and the dimeric products (e.g., Pr(2+)-Pr(2+)). In contrast to HE, there was very little intracellular uptake of HPr(+). We conclude that HPr(+) is a useful probe for detecting O(2)(•-) and other one-electron oxidizing species in an extracellular milieu.

Project description:Various chemical probes for the detection of reactive oxygen species have been developed to examine oxidative stress associated with different pathologies. L-012, a luminol-based chemiluminescent probe, is widely used to detect extracellular superoxide because of its high sensitivity. We herein demonstrated that the co-application of the peptide boronic acid proteasome inhibitor, bortezomib, with L-012 significantly increased its luminescence without affecting the background. More than a 5-fold increase was detected in the total luminescence of L-012 in both NADPH oxidase-expressing cells and the xanthine oxidase-dependent cell-free superoxide generation system, but not in their background. Therefore, bortezomib increased the signal-to-background ratio and improved the detection of low levels of superoxide. The application of MLN2238, another peptide boronic acid proteasome inhibitor, also enhanced the luminescence of L-012. In contrast, carfilzomib, an epoxyketone proteasome inhibitor, did not increase luminescence, suggesting that the effects of bortezomib depend on the chemical structure of the peptide boronic acid, but not on its pharmacological effects. Bortezomib-induced enhancements appeared to be specific to the detection of superoxide because the detection of H2O2 by Amplex Red/HRP was not affected by the application of bortezomib. In the quantitative detection of the superoxide-specific oxidative product 2-hydroxyethidium (2-OH-E+), the application of bortezomib resulted in a 2-fold increase in the level of 2-OH-E+. Therefore, bortezomib sensitizes the detection of superoxide in both cell-based and cell-free systems, highlighting a novel feature of compounds containing the peptide boronic acid as powerful enhancers for the detection of superoxide.

Project description:All aerobic cells contain reactive oxygen species (ROSs) in balance with biochemical antioxidants. Oxidative stress is developed when this balance gets disturbed because of excessive production of ROSs or depletion of antioxidants. Here, in this work, we have developed the first cyclic diselenide BODIPY-based (organoselenium-containing) probe for the selective detection of superoxide. The probe demonstrates excellent selective response for superoxide over other ROSs with nine-fold increase in fluorescence intensity. The detection limit was found to be 0.924 μM. The plausible "turn-on" mechanism has been proposed based on the spectroscopic and quantum chemical data. Usefulness of the probe for selective detection of superoxide was confirmed in mammalian breast cancer cell lines.

Project description:Mitochondria are highly dynamic organelles undergoing constant network reorganization and exhibiting stochastic signaling events in the form of mitochondrial flashes (mitoflashes). Here we investigate whether and how mitochondrial network dynamics regulate mitoflash biogenesis and signaling. We found that mitoflash frequency was largely invariant when network fragmentized or redistributed in the absence of mitofusin (Mfn) 1, Mfn2, or Kif5b. However, Opa1 deficiency decreased spontaneous mitoflash frequency due to superimposing changes in respiratory function, whereas mitoflash response to non-metabolic stimulation was unchanged despite network fragmentation. In Drp1- or Mff-deficient cells whose mitochondria hyperfused into a single whole-cell reticulum, the frequency of mitoflashes of regular amplitude and duration was again unaltered, although brief and low-amplitude "miniflashes" emerged because of improved detection ability. As the network reorganized, however, the signal mass of mitoflash signaling was dynamically regulated in accordance with the degree of network connectivity. These findings demonstrate a novel functional role of mitochondrial network dynamics and uncover a magnitude- rather than frequency-modulatory mechanism in the regulation of mitoflash signaling. In addition, our data support a stochastic trigger model for the ignition of mitoflashes.

Project description:Mitochondrial flashes mediated by optic atrophy 1 (OPA1) fusion protein are bioenergetic responses to stochastic drops in mitochondrial membrane potential (Δψm) whose origin is unclear. Using structurally distinct genetically encoded pH-sensitive probes, we confirm that flashes are matrix alkalinization transients, thereby establishing the pH nature of these events, which we renamed "mitopHlashes". Probes located in cristae or intermembrane space as verified by electron microscopy do not report pH changes during Δψm drops or respiratory chain inhibition. Opa1 ablation does not alter Δψm fluctuations but drastically decreases the efficiency of mitopHlash/Δψm coupling, which is restored by re-expressing fusion-deficient OPA1K301A and preserved in cells lacking the outer-membrane fusion proteins MFN1/2 or the OPA1 proteases OMA1 and YME1L, indicating that mitochondrial membrane fusion and OPA1 proteolytic processing are dispensable. pH/Δψm uncoupling occurs early during staurosporine-induced apoptosis and is mitigated by OPA1 overexpression, suggesting that OPA1 maintains mitopHlash competence during stress conditions. We propose that OPA1 stabilizes respiratory chain supercomplexes in a conformation that enables respiring mitochondria to compensate a drop in Δψm by an explosive matrix pH flash.

Project description:UnlabelledCentral to bioenergetics and reactive oxygen species (ROS) signaling, the mitochondrion plays pivotal roles in the pathogenesis of metabolic diseases. Recent advances have shown that mitochondrial flash ("mitoflash") visualized by the biosensor mt-cpYFP affords a frequency-coded, optical readout linked to mitochondrial ROS production and energy metabolism, at the resolution of a single mitochondrion. To investigate possible mitoflash responses to metabolic stress in insulin resistance (IR), we generated an mt-cpYFP-expressing db/db mouse model with the obesity and IR phenotypes unaltered. In conjunction with in vivo imaging of skeletal muscles, we uncovered a progressive increase of mitoflash frequency along with its morphological changes. Interestingly, enhanced mitochondrial networking occurred at 12 weeks of age, and this was followed by mitochondrial fragmentation at 34 weeks. Pioglitazone treatment normalized mitoflash frequency and morphology while restored mitochondrial respiratory function and insulin sensitivity in 12 weeks mt-cpYFP db/db mice. Mechanistic study revealed that the mitoflash remodeling was associated with altered expression of proteins involved in mitochondrial dynamics and quality control. These findings indicate that mitoflash activity may serve as an optical functional readout of the mitochondria, a robust and sensitive biomarker to gauge IR stresses and their amelioration by therapeutic interventions.Key message• In vivo detection of mitochondrial flashes in mt-cpYFP-expressing db/db mouse. • Mitoflash frequency increased progressively with disease development. • Mitoflash morphology revealed a biphasic change in mitochondrial networking. • Mitoflash abnormalities and mitochondrial defects are restored by pioglitazone. • Mitoflash may serve as a unique biomarker to gauge metabolic stress in insulin resistance.

Project description:Mitoflashes are spontaneous transients of the biosensor mt-cpYFP. In cardiomyocytes, mitoflashes are associated with the cyclophilin D (CypD) mediated opening of mitochondrial permeability transition pore (mPTP), while in skeletal muscle they are considered hallmarks of mitochondrial respiration burst under physiological conditions. Here, we evaluated the potential association between mitoflashes and the mPTP opening at different CypD levels and phosphorylation status by generating three CypD derived fusion constructs with a red shifted, pH stable Ca2+ sensor jRCaMP1b. We observed perinuclear mitochondrial Ca2+ efflux accompanying mitoflashes in CypD and CypDS42A (a phosphor-resistant mutation at Serine 42) overexpressed myofibers but not the control myofibers expressing the mitochondria-targeting sequence of CypD (CypDN30). Assisted by a newly developed analysis program, we identified shorter, more frequent mitoflash activities occurring over larger areas in CypD and CypDS42A overexpressed myofibers than the control CypDN30 myofibers. These observations provide an association between the elevated CypD expression and increased mitoflash activities in hindlimb muscles in an amyotrophic lateral sclerosis (ALS) mouse model previously observed. More importantly, feeding the mice with sodium butyrate reversed the CypD-associated mitoflash phenotypes and protected against ectopic upregulation of CypD, unveiling a novel molecular mechanism underlying butyrate mediated alleviation of ALS progression in the mouse model.
