Project description:Pachyptera DC. is a small genus of neotropical lianas included in tribe Bignonieae (Bignoniaceae). The genus has a complicated taxonomic history but currently includes species distributed from Belize to Southern Amazon. Pachyptera is characterised by four main synapomorphies, namely, a papery peeling bark, prophylls of the axillary buds organised in a series of three, patelliform glands arranged in lines in the upper portions of the calyx and corolla tube. Furthermore, members of the genus also have stems with four phloem wedges in cross-section and conspicuous extrafloral nectaries between the interpetiolar region and at the petiole apex, although these characters are also shared with other genera of tribe Bignonieae. Here, we present a taxonomic revision of Pachyptera, which includes a complete list of synonyms, detailed morphological descriptions of species and an identification key, as well as information on the habitat, distribution and phenology, nomenclatural notes, taxonomic comments and illustrations of all the species. In addition, we designate three lectotypes, propose one new combination, raise one variety to species status and describe a new species. After these adjustments, a Pachyptera with five well-defined species is recognised.

Project description:Chloroplast (cp) genome organization, gene order, and content have long been considered conserved among land plants. Despite that, the generation of thousands of complete plastomes through next-generation sequencing (NGS) has challenged their conserved nature. In this study, we analyze 11 new complete plastomes of Amphilophium (Bignonieae, Bignoniaceae), a diverse genus of Neotropical lianas, and that of Anemopaegma prostratum. We explored the structure and content of the assembled plastomes and performed comparative analyses within Amphilophium and among other plastomes available for Bignoniaceae. The overall gene content and orientation of plastomes is similar in all species studied. Plastomes are not conserved among Amphilophium, showing significant differences in length (155,262-164,786 bp), number of genes duplicated in the IRs (eight, 18, or 19), and location of the SC/IR boundaries (i.e., LSC/IRa junction between rps19 and rpl2 genes, within petD, or within petB). Length differences reflect expansions of the IRs and contractions of the LSC regions. The plastome of A. prostratum is 168,172 bp, includes 19 duplicated genes, and has the LSC/IRa boundary located within the petB gene. Amphilophium plastomes show high nucleotide diversity, with many hypervariable regions, and 16 genes with signatures of positive selection. Multiple SSRs and repeat regions were identified for Amphilophium and Anemopaegma prostratum. The differences in structure detected within Amphilophium plastomes in terms of LSC/IR and IR/SSC boundaries, number of duplicated genes, and genome sizes are mostly shared between taxa that belong to the same clade. Our results bring new insights into the evolution of plastomes at low taxonomic levels.

Project description:Tanaecium Sw. emend L.G. Lohmann (Bignonieae, Bignoniaceae) is a genus of Neotropical lianas that is morphologically variable, especially in floral features. The genus is distributed from Mexico and the Antilles to Argentina, and centered in Amazonia. Here, we present an updated overview for Tanaecium that recognizes 21 species within the genus. Species delimitation was based on a detailed analysis of protologues and herbarium specimens, including type collections of all taxa. We present a detailed description for the genus and a key for the identification of all species. For each of the 21 species recognized, we present information on the nomenclature, phenology, habitat, distribution, and taxonomic notes. Furthermore, Spathicalyx kuhlmannii J.C. Gomes is transferred into Tanaecium kuhlmannii (J.C. Gomes) Frazão & L.G. Lohmann. A lectotype is proposed for Tanaecium crucigerum Seem.

Project description:Tynanthus is a genus of lianas that is broadly distributed through the Neotropics. Two new species of Tynanthus from Brazil are here described and illustrated: Tynanthusdensiflorus, from Amazonas, and Tynanthusespiritosantensis, from Espírito Santo. Tynanthusdensiflorus is recognized by the conspicuous interpetiolar glandular fields, a feature rarely found in Tynanthus, and the dense thyrses. Tynanthusespiritosantensis, on the other hand, is recognized by the bromeliad-like prophylls of the axillary buds and the lax thyrses. Information on the distribution, conservation status and morphologically similar species are provided.

Project description:The chloroplast is one of the most important organelles of plants. This organelle has a circular DNA with approximately 130 genes. The use of plastid genomic data in phylogenetic and evolutionary studies became possible with high-throughput sequencing methods, which allowed us to rapidly obtain complete genomes at a reasonable cost. Here, we use high-throughput sequencing to study the "Adenocalymma-Neojobertia" clade (Bignonieae, Bignoniaceae). More specifically, we use Hi-Seq Illumina technology to sequence 10 complete plastid genomes. Plastomes were assembled using selected plastid reads and de novo approach with SPAdes. The 10 assembled genomes were analyzed in a phylogenetic context using five different partition schemes: (1) 91 protein-coding genes ("coding"); (2) 76 introns and spacers with alignment manually edited ("non-coding edited"); (3) 76 non-coding regions with poorly aligned regions removed using T-Coffee ("non-coding filtered"); (4) 91 coding regions plus 76 non-coding regions edited ("coding + non-coding edited"); and, (5) 91 protein-coding regions plus the 76 filtered non-coding regions ("coding + non-coding filtered"). Fragmented regions were aligned using Mafft. Phylogenetic analyses were conducted using Maximum Likelihood (ML) and Bayesian Criteria (BC). The analyses of the individual plastomes consistently recovered an expansion of the Inverted Repeated (IRs) regions and a compression of the Small Single Copy (SSC) region. Major genomic translocations were observed at the Large Single Copy (LSC) and IRs. ML phylogenetic analyses of the individual datasets led to the same topology, with the exception of the analysis of the "non-coding filtered" dataset. Overall, relationships were strongly supported, with the highest support values obtained through the analysis of the "coding + non-coding edited" dataset. Four regions at the LSC, SSC, and IR were selected for primer development. The "Adenocalymma-Neojobertia" clade shows an unusual pattern of plastid structure variation, including four major genomic translocations. These rearrangements challenge the current view of conserved plastid genome architecture in terms of gene order. It also complicates both genomic assemblies using reference genomes and sequence alignments using whole plastomes. Therefore, strategies that employ de novo assemblies and manual evaluation of sequence alignments are required to prevent assembly and alignment errors.

Project description:We developed chloroplast microsatellite markers (cpSSRs) to be used to study the patterns of genetic structure and genetic diversity of populations of Stizophyllum riparium (Bignonieae, Bignoniaceae). We used genomic data obtained through an Illumina HiSeq sequencing platform to develop a set of cpSSRs for S. riparium. A total of 36 primer pairs were developed, of which 28 displayed polymorphisms across 59 individuals from three populations. Two to 12 alleles were recorded, and the unbiased haploid diversity per locus ranged from 0.037 to 0.905. All 28 cpSSRs presented transferability to two closely related species, S. inaequilaterum and S. perforatum. We report a set of 28 cpSSRs for S. riparium. All markers were shown to be variable in S. riparium, indicating that these markers will be valuable for population genetic studies across S. riparium and congeneric species.

Project description:In this study, we analyzed the morphological affinities of the 24 species of Amphitecna based on detailed morphological studies and multivariate cluster analyses. Our results suggest that the genus Amphitecna includes six morphological groups that can be easily distinguished based on floral and fruits characteristics: A. donnell-smithii group, A. macrophylla group, A. megalophylla group, A. molinae group, A. spathicalyx group, and A. steyermarkii group. A new species from Mexico, Amphitecna fonceti, is described. This new species is clearly differentiated by the predominantly ramiflorous inflorescences bearing multiple flowers per shoot, buds rounded at the apex, large flowers with a transverse fold in the corolla throat, calyx surface pubescent and strongly costate, and fruits elliptic, apiculate at the apex. We discuss the characteristics of each morphological group and their geographical distribution, provide a detailed description of the new species including ethnobotany notes, and propose the re-establishment of the giant-leaved species A. megalophylla.

Project description:The influence of ecological traits to the distribution and abundance of species is a prevalent issue in biodiversity science. Most studies of plant community assembly have focused on traits related to abiotic aspects or direct interactions among plants, with less attention paid to ignore indirect interactions, as those mediated by pollinators. Here, we assessed the influence of phylogeny, habitat, and floral morphology on ecological community structure in a clade of Neotropical lianas (tribe Bignonieae, Bignoniaceae). Our investigation was guided by the long-standing hypothesis that habitat specialization has promoted speciation in Bignonieae, while competition for shared pollinators influences species co-occurrence within communities. We analyzed a geo-referenced database for 94 local communities occurring across the Neotropics. The effect of floral morphological traits and abiotic variables on species co-occurrence was investigated, taking into account phylogenetic relationships. Habitat filtering seems to be the main process driving community assembly in Bignonieae, with environmental conditions limiting species distributions. Differing specialization to abiotic conditions might have evolved recently, in contrast to the general pattern of phylogenetic clustering found in communities of other diverse regions. We find no evidence that competition for pollinators affects species co-occurrence; instead, pollinator occurrence seems to have acted as an "environmental filter" in some habitats.

Project description:Background:Jacaranda decurrens Cham., known as carobinha, is prevalent in the Cerrado biome and presents popular use in treatment of dermatological diseases. The present study aimed to investigate the healing action of topical formulation of Jacaranda decurrens Cham. (FtEHJ) in mice cutaneous lesions. Methods:Phytochemical analysis of J. decurrens hydroalcoholic extract was carried out by using HPLC-PDA-ESI-MS and FIA-ESI-IT-MSn. Swiss mice were treated topically with formulation base (FtB) or Fibrinase® or ointment FtEHJ (15?mg/g; 50?mg/Kg). At the end of treatment periods, the inflammatory cytokines (TNF-?, IL-1?, and IL-6) in the lesions were measured by using ELISA and gene expression of TGF-?, Collagen I, and Collagen III was demonstrated by RTqPCR method and histological evaluation. Results:Ten compounds were identified in the extract, distributed among the classes of flavonoids and triterpenes. Treatment with FtEHJ increased the wound contraction in 24 hours, such as reduction of TNF-?, IL-1?, and IL-6 (pg/mL) cytokines in the lesion. The TGF-? and collagen gene expression was increased and the wound closure accelerated to nine days, with discrete inflammation, collagenization, and accented reepithelialization. Conclusions. The results obtained suggest chemical compounds present in the FtEHJ accelerates wound healing by being a gene expression modulator, and protein content of different molecules are involved in tissue repair.

Project description:Martinella has traditionally included two species, Martinella iquitoensis and Martinella obovata, that are characterized by the presence of interpetiolar ridges surrounding the stems and minute prophylls of the axillary buds. A third species, Martinella insignis, is here described as new, illustrated and compared to other species in the genus. Martinella insignis is the first record of the genus in the Atlantic Forest of Brazil, and differs from other species of Martinella by the yellow corolla (vs. red to dark purple) and 5-lobed calices (vs. 2-4-lobed).
