Project description:Quorum sensing (QS) is a mechanism dependent on bacterial density. This coordinated process is mediated by the synthesis and the secretion of signal molecules, called autoinducers (AIs). N-acyl-homoserine lactones (AHLs) are the most common AIs that are used by Gram-negative bacteria and are involved in biofilm formation. Quorum Quenching (QQ) is the interference of QS by producing hydrolyzing enzymes, among other strategies. The main objective of the present study was to identify QS and QQ strains from MBR wastewater treatment plants. A total of 99 strains were isolated from two Spanish plants that were intended to treat leachate from municipal solid waste. Five AHL producers were detected using AHL biosensor strains (Chromobacterium violaceum CV026 and Agrobacterium tumefaciens NT1). Fifteen strains of seventy-one Gram-positive were capable of eliminating or reducing at least one AHL activity. The analysis of 16S rRNA gene sequence showed the importance of the Pseudomonas genus in the production of biofilms and the relevance of the genus Bacillus in the disruption of the QS mechanism, in which the potential activity of lactonase or acylase enzymes was investigated with the aim to contribute to solve biofouling problems and to increase the useful lifespan of membranes.

Project description:This work aimed at studying the photochemical treatment of a landfill leachate using ultraviolet light, hydrogen peroxide, and ferrous or ferric ions, in a batch recycle photoreactor. The effect of inorganic carbon presence, pH, initial H2O2 amount (0-9990 mg L-1) as well as Fe(II) (200-600 ppm) and Fe(III) (300-700 ppm) concentrations on the total carbon removal and color change was studied. Prior to the photochemical treatment, a pretreatment process was applied; inorganic nitrogen and inorganic carbon were removed by means of air stripping and initial pH regulation, respectively. The leachate sent subsequently for photochemical treatment was free of inorganic carbon and contained only organic carbon with concentration 1200±100 mg L-1 at pH 5.1-5.3. The most favorable concentrations of H2O2 and ferric ions for carbon removal were 6660 mg L-1 and 400 ppm, respectively. Adjusting the initial pH value in the range of 2.2-5.3 had a significant effect on the organic carbon removal. The photo-Fenton-like process was more advantageous than the photo-Fenton one for leachate treatment. By applying the most favorable operating conditions, 88.7% removal of total organic carbon, 100% removal of total inorganic carbon, 96.5% removal of total nitrogen, and 98.2% color removal were achieved.

Project description:Occupational hearing loss is an increasingly prevalent occupational condition worldwide, and has been reported to occur in a wide range of workplaces; however, its prevalence among workers from municipal solid waste landfills (MSWLs) remains less clear. This study aimed to investigate the occupational hearing loss among Chinese MSWL workers.A cross-sectional study of 247 workers from 4 Chinese MSWLs was conducted. Noise and total volatile organic compounds (TVOCs) levels at worksites were determined. We conducted hearing examinations to determine hearing thresholds. A worker was identified as having hearing loss if the mean threshold at 2000, 3000 and 4000 Hz in either ear was equal to or greater than 25 dB. Prevalence of occupational hearing loss was then evaluated. Using unconditional Logistic regression models, we estimated the odds ratios (ORs) of MSWL work associated with hearing loss.According to the job title for each worker, the study subjects were divided into 3 groups, including group 1 of 63 workers without MSWL occupational hazards exposure (control group), group 2 of 84 workers with a few or short-period MSWL occupational hazards exposure, and group 3 of 100 workers with continuous MSWL occupational hazards exposure. Both noise and TVOCs levels were significantly higher at worksites for group 3. Significantly poorer hearing thresholds at frequencies of 2000, 3000 and 4000 Hz were found in group 3, compared with that in group 1 and group 2. The overall prevalence rate of hearing loss was 23.5%, with the highest in group 3 (36.0%). The OR of MSWL work associated with hearing loss was 3.39 (95% confidence interval [CI]: 1.28-8.96).The results of this study suggest significantly higher prevalence of hearing loss among MSWL workers. Further studies are needed to explore possible exposure-response relationship between MSWL occupational hazards exposure and hearing loss.

Project description:Municipal solid waste incinerator (MSWI) bottom ash is often used as the protection layer for the geomembrane and intermediate layer in the landfill. In this study, three sets of simulated landfills with different mass proportion of MSWI bottom ash layer to municipal solid waste (MSW) layer were operated. Cu and Zn concentrations in the leachates and MSW were monitored to investigate the effect of MSWI bottom ash layer on the Cu and Zn discharge from the landfill. The results showed that the Zn discharge was dependent on the mass proportion of MSWI bottom ash layer. The pH of landfill was not notably increased when the mass proportion of MSWI bottom ash layer to MSW layer was 1?:?9, resulting in the enhancement of the Zn discharge. However, Zn discharge was mitigated when the mass proportion was 2?:?8, as the pH of landfill was notably promoted. The discharge of Cu was not dependent on the mass proportion, due to the great affinity of Cu to organic matter. Moreover, Cu and Zn contents of the sub-MSW layer increased due to the MSWI bottom ash layer. Therefore, the MSWI bottom ash layer can increase the potential environmental threat of the landfill.

Project description:Average biodegradation rate of newly filled municipal solid waste (MSW) in landfills is relatively fast, and the landfill gas produced by the new MSW biodegradation can cause great variations in gas pressure. To predict the gas pressure distribution in the MSW layer, a one-dimensional gas transport model is established in this study. The following factors are considered in this model: (1) the variation of gas permeability with depth; (2) the anisotropy ratio of gas permeability; (3) the settlement caused by waste biodegradation. Furthermore, a single peak model for gas production is applied as the source term of gas production. The equation for settlement caused by waste biodegradation is presented, and the time of peak gas production rate is obtained by fitting the settlement of the newly filled layer. The stratification of the unsaturated and saturated regions is taken into account by distinguishing the difference in gas saturation. The layering of the new and old waste layers is considered by distinguishing the difference in the length of time that waste has been degraded to produce gas. Based on the method of numerical calculation, the gas pressure distribution in the landfill with layered new and old MSW is well simulated. The position where the maximum gas pressure occurs is found. The sensitivity analysis shows that the influence of the anisotropy ratio on gas pressure distribution is more significant.

Project description:Omics have given rise to research on sparsely studied microbial communities such as the landfill, lignocellulolytic microorganisms and enzymes. The bacterial diversity of Municipal Solid Waste sediments was determined using the illumina MiSeq system after DNA extraction and Polymerase chain reactions. Data analysis was used to determine the community's richness, diversity, and correlation with environmental factors. Physicochemical studies revealed sites with mesophilic and thermophilic temperature ranges and a mixture of acidic and alkaline pH values. Temperature and moisture content showed the highest correlation with the bacteria community. The bacterial analysis of the community DNA revealed 357,030 effective sequences and 1891 operational taxonomic units (OTUs) assigned. Forty phyla were found, with the dominant phyla Proteobacteria, Firmicutes, Actinobacteria, and Bacteroidota, while Aerococcus, Stenotrophomonas, and Sporosarcina were the dominant species. PICRUSt provided insight on community's metabolic function, which was narrowed down to search for lignocellulolytic enzymes' function. Cellulase, xylanase, esterase, and peroxidase were gene functions inferred from the data. This article reports on the first phylogenetic analysis of the Pulau Burung landfill bacterial community. These results will help to improve the understanding of organisms dominant in the landfill and the corresponding enzymes that contribute to lignocellulose breakdown.

Project description:Groundwater pollution resulting from leachate leakage at landfill sites has garnered significant attention. Investigating the migration of pollutants from these landfills to adjacent groundwater is crucial for understanding the diffusion patterns and extent of contamination. It is imperative to develop cost-effective yet highly efficient tracer techniques to aid landfill operators in monitoring groundwater contamination stemming from their operations. The primary objective of this research was to compare the roles of conservative tracers sodium (Na+) and chloride (Cl-), and conventional pollutants permanganate oxidation (CODMn), ammonium nitrogen (NH4 +-N), lead (Pb), and zinc (Zn) in assessing pollution levels from municipal solid waste landfills to groundwater. For this purpose, a typical municipal solid landfill was selected to investigate the origin of Cl-, groundwater quality, and spatiotemporal variations of multiple contaminations. Geochemistry analyses revealed that Na-Cl and Ca-HCO3 were the dominant groundwater type in this study and landfill was the primary source of Cl- in groundwater, with an average contribution of 78 %. Groundwater in proximity to the landfill (5#, 2#, 22#, 23#) exhibited elevated concentrations of Na+ (15.6-914.0 mg/L), Cl- (8.9-1352.0 mg/L), CODMn (0.54-95.9 mg/L), and NH4 +-N (0.33-49.0 mg/L), yet demonstrated reduced levels of Pb (0.2-391.0 μg/L) and Zn (2.0-112.8 μg/L). In contrast, groundwater located at a considerable distance from the landfill (13#, 18#, 15#, 26#) displayed the inverse trend, with relatively low concentration of Na+ (3.2-8.5 mg/L), Cl- (0.1-0.7 mg/L), CODMn (0.28-4.78 mg/L), and NH4 +-N (0.03-0.52 mg/L), but increased levels of Pb (1.2-483.0 μg/L) and Zn (1.6-357.0 μg/L). The primary determinant of groundwater quality near the landfill was NH4 +-N, with the highest pollution index (Pi) of 492.85, whereas Pb was the predominant factor affecting water quality in areas distant from the landfill, with the highest pollution index (Pi) of 10.9. While no discernible seasonal variation was detected for all pollutants, spatial variation can be observed that pollution levels decreased progressively with increasing distance from the landfill, a trend particularly corroborated by the conservative Cl- and Na+ measurements. This research suggests that conservative ions, such as Cl- and Na+, exhibit superior efficacy in tracing the pollution range from municipal solid landfills to groundwater. Therefore, monitoring these conservative ions in groundwater can yield a more precise understanding of the extent of groundwater contamination originated from landfills.

Project description:Based on a field monitored dataset measured at landfill #1 over 21 years, the characteristics of settlement coupling mechanical creep and biodegradation and the residual settlement were analyzed. Since landfill #1 is a multi-stage municipal solid waste (MSW) landfill where dykes are constructed after landfilling for subsequent waste fills, the waste decomposition between the lower and upper lifts was quite different and it is difficult to discern between the mechanical creep and bio-compression on the settlement curves. The compression ratio coupled with mechanical creep and bio-compression and the residual compression ratio were determined as 0.233 and 0.068, respectively. This implies that biodegradation was gradually and significantly reduced in the MSW settlement behavior after the residual settlement began. The starting date of residual settlement was distributed between 3821 and 5402 days from the initial date of landfilling. The settlement coupling mechanical creep and biodegradation (SMB) was 2.9 times larger than the residual settlement (SRS), and the duration of SMB is determined to be 0.3 times that of SRS. In addition, the remnant methane gas content existed in the landfill gas, and low-level biodegradation was still generated in the waste buried for more than 10 years after the residual settlement began.

Project description:Municipal solid waste incineration (MSWI) ash is often managed through co-disposal with unburned wastes in landfills, a practice previously reported to result in enhanced leaching of pollutants (e.g., heavy metals) in landfill leachate. The objective of this study was to evaluate the effect of co-disposed unburned wastes on per- and polyfluoroalkyl substances (PFAS) in MSWI ash landfill leachate. Leachate was collected from four landfills containing MSWI ash, either as a sole waste stream or co-disposed of with sewage sludge and MSW screenings. Samples of ash and unburned materials were collected and assessed separately for leachable PFAS in the laboratory. All samples were analyzed for 26 PFAS. Results showed that greater ash content was associated with lower leachate PFAS concentrations. The pure ash monofill exhibited the lowest PFAS in landfill leachate (290 ng L-1) while the landfill contained a large amount of unburned waste had the highest PFAS (11,000 ng L-1). For laboratory leaching tests, average ∑26PFAS concentration in lab ash leachate (310 ng L-1) was 10 and 24 times lower than observed in lab sewage sludge leachate (3,200 ng L-1) and lab MSW screenings leachate (7,500 ng L-1), respectively. Leachate from the ash-only landfill had ∑26PFAS concentration similar to what was measured in the ash itself. On the contrary, ∑26PFAS concentration in co-disposal landfill leachates were similar to those in PFAS-rich unburned waste itself, regardless of the percentages of landfilled unburned wastes. We hypothesize that leachate generated in co-disposal scenarios preferentially flows through PFAS-rich unburned materials and that biotransformation of precursors enhanced by unburned waste degradation further contributes to higher concentrations of terminal PFAS in ash co-disposal sites. Landfill operators should expect PFAS in leachates to be higher when PFAS-rich unburned wastes are disposed of alongside MSWI ash, even if the unburned fraction is small.
