Project description:Experimental endotoxemia is a translational model to study inflammatory mechanisms involved in the pathophysiology of mood disorders including depression. Disturbed affective cognition constitutes a core aspect in depression, but has never been studied in the context of inflammation. We combined experimental endotoxemia with an established experimental mood induction procedure to assess the interaction between acute inflammation and sad mood and their effects on affective cognition. In this randomized cross-over study, N?=?15 healthy males received endotoxin (0.8?ng/kg lipopolysaccharide iv) on one study day and placebo an otherwise identical study day. The affective Go/Nogo task was conducted after experimental induction of neutral and sad mood. Inflammatory markers were assessed hourly. Endotoxin application induced a transient systemic inflammation, characterized by increased leukocyte counts, TNF-alpha and interleukin-6 plasma concentrations (all p?<?0.01, interaction effects). Mood induction led to greater sadness ratings, with highest ratings when sad mood was induced during inflammation (p?<?0.05, interaction effect). Based on a 2 (endotoxin vs. placebo)?×?2 (sad vs. neutral mood)?×?2 (sad vs. happy Go/Nogo target words) factorial design, we observed a significant target?×?endotoxin condition interaction (p?<?0.01) reflecting slower responses to sad targets during endotoxemia. Additionally, we found a valence?×?mood interaction (p?<?0.05), reflecting slower reaction times to sad targets in sad mood. In summary, acute inflammation and sad mood are risk factors for disturbed affective cognition. The results may reflect a mood-congruency effect, with prolonged and sustained processing of mood-congruent information during acute inflammation, which may contribute to depression risk.

Project description:Insecure attachment in couples is negatively associated with relationship functioning. Similarity of partner attachment on the other hand might attenuate such relationship outcomes. We tested the opposing insecurity and similarity hypotheses by examining associations of attachment with relationship satisfaction and instability in a representative community sample of couples (N = 1,014). We expected: (a) negative effects of insecure attachment (main effects of actor and partner avoidance and anxiety, and interaction effects consistent with fearful-avoidance and demand-withdraw patterns); and (b) positive effects of similarity in avoidance and anxiety. Actor-Partner-Interdependence Models showed clear support for the insecurity hypothesis. Main effects of avoidance and anxiety, explained 46.2% of the variance of satisfaction, and 17.9% of instability. We conclude that reducing insecurity of attachment, in particular avoidance, must be a central target in couple therapy. Interestingly, similarity of attachment can partially buffer the negative effects of attachment insecurity. Clinical implications are discussed.

Project description:BackgroundThere is growing interest in the nature of slow variations of the blood oxygen level-dependent (BOLD) signal observed in functional MRI resting-state studies. In humans, these slow BOLD variations are thought to reflect an underlying or intrinsic form of brain functional connectivity in discrete neuroanatomical systems. While these 'resting-state networks' may be relatively enduring phenomena, other evidence suggest that dynamic changes in their functional connectivity may also emerge depending on the brain state of subjects during scanning.Methodology/principal findingsIn this study, we examined healthy subjects (n = 24) with a mood induction paradigm during two continuous fMRI recordings to assess the effects of a change in self-generated mood state (neutral to sad) on the functional connectivity of these resting-state networks (n = 24). Using independent component analysis, we identified five networks that were common to both experimental states, each showing dominant signal fluctuations in the very low frequency domain (approximately 0.04 Hz). Between the two states, we observed apparent increases and decreases in the overall functional connectivity of these networks. Primary findings included increased connectivity strength of a paralimbic network involving the dorsal anterior cingulate and anterior insula cortices with subjects' increasing sadness and decreased functional connectivity of the 'default mode network'.Conclusions/significanceThese findings support recent studies that suggest the functional connectivity of certain resting-state networks may, in part, reflect a dynamic image of the current brain state. In our study, this was linked to changes in subjective mood.

Project description:Attachment insecurity is determined early in life, is a risk factor for psychopathology, and can be measured on two separate continuous dimensions: attachment anxiety and attachment avoidance. Therapeutic changes toward more secure attachment correlate with reduction in psychiatric symptoms. Psilocybin-assisted psychotherapy has demonstrated promise in the treatment of psychopathology, such as treatment-resistant depression and substance use disorders. We hypothesized that psilocybin-assisted psychotherapy would reduce attachment anxiety and attachment avoidance, thus increasing attachment security. We also hypothesized that baseline measures of attachment insecurity, which can reflect a diminished capacity for trust and exploration, would inform the quality of the psilocybin session. Participants were male long-term AIDS survivors with moderate-severe demoralization (n = 18). Using the Experiences in Close Relationships scale, we measured attachment insecurity at baseline as well as immediately, and 3 months, after completion of a brief group therapy course, which included a single midtreatment open-label psilocybin session conducted individually. Clinically important aspects of the psilocybin session were assessed using the revised Mystical Experience Questionnaire and the Challenging Experience Questionnaire the day following psilocybin administration. Self-reported ratings of attachment anxiety decreased significantly from baseline to 3-months post-intervention, t(16) = -2.2; p = 0.045; d rm = 0.45; 95% CI 0.01, 0.87. Attachment avoidance did not change significantly. Baseline attachment anxiety was strongly correlated with psilocybin-occasioned mystical-type experiences, r(15) = 0.53, p = 0.029, and baseline attachment avoidance was strongly correlated with psilocybin-related challenging experiences, r(16) = 0.62, p = 0.006. These findings have important implications for the general treatment of psychopathology as well as optimizing psilocybin-assisted psychotherapy as a broadly applicable treatment modality.

Project description:We use satellite ozone records and Global Modeling Initiative chemistry transport model simulations integrated with Modern Era Retrospective for Research and Analysis 2 meteorology to identify a metric that accurately captures the trend in Antarctic ozone attributable to the decline in ozone depleting substances (ODSs). The GMI CTM Baseline simulation with realistically varying ODS levels closely matches observed interannual to decadal scale variations in Antarctic September ozone over the past four decades. The expected increase or recovery trend is obtained from the differences between the Baseline simulation and one with identical meteorology and fixed 1995 ODS levels. The differences show that vortex-averaged column O3 has the greatest sensitivity to ODS change from 1 to 20 September. The observed vortex-averaged column O3 during this period produces a trend consistent with the expected recovery attributable to ODS decline. Trends from dates after 20 September have smaller sensitivity to ODS decline and are more uncertain due to transport variability. Simulations show that the greatest decrease in O3 loss (i.e., recovery) occurs inside the vortex near the edge. The polar cap metrics have vortex size-dependent bias and do not consistently sample this region. Because the 60-90°S 220 Dobson unit O3 mass deficit metric does not sample the edge region, its trend is lower than the expected trend; this is improved by area weighting. The 250-Dobson unit O3 mass deficit metric samples more of the edge region, which increases its trend. Approximately 25% of the September Antarctic O3 increase is due to higher O3 levels in June prior to winter depletion.

Project description:Behavioral studies suggest a relationship between autobiographical memory, rumination and depression. The objective of this study was to determine whether remitted depressed patients show alterations in connectivity of the posterior cingulate cortex (PCC, a node in the default mode network) with the parahippocampal gyri (PHG, a region associated with autobiographical memory) while intensively recalling negative memories and whether this is related to daily life symptoms and to the further course of depression. Sad mood was induced with keywords of personal negative life events in participants with remitted depression (n = 29) and matched healthy controls (n = 29) during functional magnetic resonance imaging. Additionally, daily life assessments of mood and rumination and a 6-month follow-up were conducted. Remitted depressed participants showed greater connectivity than healthy controls of the PCC with the PHG, which was even stronger in patients with more previous episodes. Furthermore, patients with increased PCC-PHG connectivity showed a sadder mood and more rumination in daily life and a worsening of rumination and depression scores during follow-up. A relationship of negative autobiographical memory processing, rumination, sad mood and depression on a neural level seems likely. The identified increased connectivity probably indicates a 'scar' of recurrent depression and may represent a prognostic factor for future depression.

Project description:ObjectivesCognitive reactivity (CR) to sad mood is a risk factor for major depressive disorder (MDD). CR is usually measured by assessing change on the Dysfunctional Attitudes Scale (DAS-change) after sad mood-induction. It has, however, been suggested that the versions of the DAS (A/B) are not interchangeable, impacting the reliability and validity of the change score. The Leiden Index of Depression Sensitivity-Revised (LEIDS-R) is an alternative self-report measure of CR. Studies examining the relationship between LEIDS-R and DAS-change have shown mixed results. We examined whether scores of these CR measures differed between remitted MDD and controls, the relationship between these CR measures, and the effect of order of DAS administration on DAS-change.DesignCross-sectional design with two groups (remitted MDD and controls).MethodsSixty-eight MDD patients remitted from ?2 previous episodes, not taking antidepressants, and 43 never-depressed controls participated in a mood-induction and filled in the DAS-A/B in randomized order before and after mood-induction, and LEIDS-R separately.ResultsLEIDS-R scores and pre-mood-induction DAS scores were significantly higher in remitted MDD than controls (p < .001, Cohen's d = 1.48; p = .001, Cohen's d = 0.66, respectively). DAS-change did not differ between these groups (p = .67, Cohen's d = 0.08). LEIDS-R correlated with DAS-change (r = .30, p = .042), but only in the group that filled in DAS-B before DAS-A. In remitted MDD, DAS-change was dependent on the order of DAS versions before and after mood-induction (10.6 ± 19.0 vs. -1.2 ± 10.5, for order B-A and A-B, respectively), with a significant group × order interaction (p = .012).ConclusionsExisting DAS versions are not interchangeable, which compromises the usefulness of mood-inductions in clinical practice. The LEIDS-R seems a valid measure of cognitive vulnerability to depression.Practitioner pointsClinical implications: Cognitive reactivity (CR) is a risk factor of depressive recurrence. The current measurement of CR, by assessing change on the Dysfunctional Attitudes Scale (DAS) after mood-induction, is not reliable. The Leiden Index Depression Sensitivity-Revised (LEIDS-R) is an alternative CR measure. In contrast to mood-induction, it reliably assesses depression vulnerability. The use of mood-inductions for clinical/research purposes is unnecessary.Limitations of the studyWe were not able to examine the effect of previous treatment, which could have affected results as psychological treatments probably have differential effects on CR. Examining un-medicated patients may have led to selection of a sample not completely representative for the general MDD population. We did not administer both parallel versions of the DAS (A/B) before and after mood-induction. This might have provided better understanding of their differential sensitivity to change.

Project description:The ability to detect ultrafast waveforms arising from randomly occurring events is essential to such diverse fields as bioimaging, spectroscopy, radio-astronomy, sensing and telecommunications. However, noise remains a significant challenge to recover the information carried by such waveforms, which are often too weak for detection. The key issue is that most of the undesired noise is contained within the broad frequency band of the ultrafast waveform, such that it cannot be alleviated through conventional methods. In spite of intensive research efforts, no technique can retrieve the complete description of a noise-dominated ultrafast waveform of unknown parameters. Here, we propose a signal denoising concept involving passive enhancement of the coherent content of the signal frequency spectrum, which enables the full recovery of arbitrary ultrafast waveforms buried under noise, in a real-time and single-shot fashion. We experimentally demonstrate the retrieval of picosecond-resolution waveforms that are over an order of magnitude weaker than the in-band noise. By granting access to previously undetectable information, this concept shows promise for advancing various fields dealing with weak or noise-dominated broadband waveforms.

Project description:Research has suggested that nostalgia is a mixed, albeit predominantly positive emotion. One proposed function of nostalgia is to attenuate the negative consequences of loneliness. This restorative effect of nostalgia, however, has been demonstrated with cross sectional and experimental methods that lack ecological validity. In studies that have measured nostalgia in daily life, however, nostalgia has been negatively related to well-being. We propose an alternative theory that posits that the effect of nostalgia on well-being depends on the event or experience that elicits nostalgia. We tested this theory by measuring daily states of nostalgia, loneliness, and affect across five daily diary studies (N = 504; 6,004 daily reports) that lasted for 14 days. Using multilevel modeling, we found that nostalgia and loneliness were negatively related to positive affect and positively related to negative affect. The negative effects of nostalgia on affective well-being were significantly stronger on days when people felt more lonely as opposed to less lonely. Viewed alternatively, the negative effects of loneliness on affective well-being were stronger on days when people felt more vs. less nostalgic. Thus, in contrast to experimental findings, nostalgia did not attenuate, but rather exaggerated the negative effects of loneliness on affective well-being. These findings support a theoretical account that proposes that the effect of nostalgia on well-being depends on the natural context in which nostalgia is elicited.

Project description:Background and objectivesDepression and cognitive impairment are common and often coexist in older adults. The network theory of mental disorders provides a novel approach to understanding the pathways between depressive symptoms and cognitive domains and the potential "bridge" that links and perpetuates both conditions. This study aimed to identify pathways and bridge symptoms between depressive symptoms and cognitive domains in older adults.Research design and methodsData were derived from 2,792 older adults aged 60 years and older with mild and more severe depressive symptoms from the community in Hong Kong. Depressive symptoms were assessed using the Patient Health Questionnaire (PHQ-9) and cognition using the Montreal Cognitive Assessment 5-minute protocol (MoCA-5min). Summary descriptive statistics were calculated, followed by network estimation using graphical LASSO, community detection, centrality analysis using bridge expected influence (BEI), and network stability analyses to assess the structure of the PHQ-9 and MoCA-5min items network, the pathways, and the bridge symptoms.ResultsParticipants (mean age = 77.3 years, SD = 8.5) scored 8.2 (SD = 3.4) on PHQ-9 and 20.3 (SD = 5.4) on MoCA-5min. Three independent communities were identified in PHQ-9 and MoCA-5min items, suggesting that depression is not a uniform entity (2 communities) and has differential connections with cognition. The network estimation results suggested that the 2 most prominent connections between depressive symptoms and cognitive domains were: (1) anhedonia with executive functions/language and (2) sad mood with memory. Among all depressive symptoms, sad mood had the highest BEI, bridging depressive symptoms and cognitive domains.Discussion and implicationsSad mood seems to be the pathway between depression and cognition in this sample of older Chinese. This finding highlights the importance of sad mood as a potential mechanism for the co-occurrence of depression and cognitive impairment, implying that intervention targeting sad mood might have rippling effects on cognitive health.
