Project description:Bacteria such as Methicillin-resistant Staphylococcus aureus (MRSA) causes acidic microenvironment during infection. A biomaterial that exhibits tunable antimicrobial property in a pH dependent manner is potentially attractive. Herein, we presented a novel antibacterial hydrogel consisting of pH responsive and reversible catechol-boronate linkage formed between intrinsically bactericidal chlorinated catechol (catechol-Cl) and phenylboronic acid. Fourier transformed infrared spectroscopy (FTIR), oscillatory rheometry, and Johnson Kendall Roberts (JKR) contact mechanics testing confirmed the formation and dissociation of the complex in a pH dependent manner. When the hydrogel was treated with an acidic buffer (pH 3), the hydrogel exhibited excellent antimicrobial property against multiple strains of Gram-positive and negative bacteria including MRSA (up to 4 log10 reduction from 108 colony forming units/mL). At an acidic pH, catechol-Cl was unbound from the phenylboronic acid and available for killing bacteria. Conversely, when the hydrogel was treated with a basic buffer (pH 8.5), the hydrogel lost its antimicrobial property but also became non-cytotoxic. At a basic pH, the formation of catechol-boronate complex effectively reduce the exposure of the cytotoxic catechol-Cl to the surrounding. When further incubating the hydrogel in an acidic pH, the reversible complex dissociated to re-expose catechol-Cl and the hydrogel recovered its antibacterial property. Overall, the combination of catechol-Cl and phenylboronic acid provide a new strategy for designing hydrogels with pH responsive antibacterial activity and reduced cytotoxicity.

Project description:A smart adhesive capable of binding to a wetted surface was prepared by copolymerizing dopamine methacrylamide (DMA) and 3-acrylamido phenylboronic acid (AAPBA). pH was used to control the oxidation state and the adhesive property of the catechol side chain of DMA and to trigger the catechol-boronate complexation. FTIR spectroscopy confirmed the formation of the complex at pH 9, which was not present at pH 3. The formation of the catechol-boronate complex increased the cross-linking density of the adhesive network. Most notably, the loss modulus values of the adhesive were more than an order of magnitude higher for adhesive incubated at pH 9 when compared to those measured at pH 3. This drastic increase in the viscous dissipation property is attributed to the introduction of reversible complexation into the adhesive network. Based on the Johnson Kendall Roberts (JKR) contact mechanics test, adhesive containing both DMA and AAPBA demonstrated strong interfacial binding properties (work of adhesion (Wadh) = 2000 mJ/m2) to borosilicate glass wetted with an acidic solution (pH 3). When the pH was increased to 9, Wadh values (180 mJ/m2) decreased by more than an order of magnitude. During successive contact cycles, the adhesive demonstrated the capability to transition reversibly between its adhesive and nonadhesive states with changing pH. Adhesive containing only DMA responded slowly to repeated changes in pH and became progressively oxidized without the protection of boronic acid. Although adhesive containing only AAPBA also demonstrated strong wet adhesion (Wadh ? 500 mJ/m2), its adhesive properties were not pH responsive. Both DMA and AAPBA are required to fabricate a smart adhesive with tunable and reversible adhesive properties.

Project description:Smart adhesive hydrogels containing 10 mol% each of dopamine methacrylamide (DMA) and 3-acrylamido phenylboronic acid (APBA) were polymerized in situ onto polydimethylsiloxane (PMDS) micropillars with different aspect ratios (AR = 0.4, 1 and 2). Using Johnson-Kendall-Roberts (JKR) contact mechanics tests, the adhesive-coated pillars demonstrated strong wet adhesion at pH 3 (Wadh = 420 mJ m-2) and can be repeatedly deactivated and reactivated by changing the pH value (pH 9 and 3, respectively). When compared to the bulk adhesive hydrogel of the same composition, the adhesive-coated pillars exhibited a significantly faster rate of transition (1 min) between strong and weak adhesion. This was attributed to an increased surface area to volume ratio of the adhesive hydrogel-coated pillars, which permitted rapid diffusion of ions into the adhesive matrix to form or break the catechol-boronate complex.

Project description:Endoscopic mucosal resection (EMR) and endoscopic submucosal dissection (ESD) are well-established therapeutics for gastrointestinal neoplasias, but complications after EMR/ESD, including bleeding and perforation, result in additional treatment morbidity and even threaten the lives of patients. Thus, designing biomaterials to treat gastric bleeding and wound healing after endoscopic treatment is highly desired and remains a challenge. Herein, a series of injectable pH-responsive self-healing adhesive hydrogels based on acryloyl-6-aminocaproic acid (AA) and AA-g-N-hydroxysuccinimide (AA-NHS) were developed, and their great potential as endoscopic sprayable bioadhesive materials to efficiently stop hemorrhage and promote the wound healing process was further demonstrated in a swine gastric hemorrhage/wound model. The hydrogels showed a suitable gelation time, an autonomous and efficient self-healing capacity, hemostatic properties, and good biocompatibility. With the introduction of AA-NHS as a micro-cross-linker, the hydrogels exhibited enhanced adhesive strength. A swine gastric hemorrhage in vivo model demonstrated that the hydrogels showed good hemostatic performance by stopping acute arterial bleeding and preventing delayed bleeding. A gastric wound model indicated that the hydrogels showed excellent treatment effects with significantly enhanced wound healing with type I collagen deposition, α-SMA expression, and blood vessel formation. These injectable self-healing adhesive hydrogels exhibited great potential to treat gastric wounds after endoscopic treatment.

Project description:Star-shaped poly(ethylene glycol) (PEG) chain termini were functionalized with alendronate to create transient networks with reversible crosslinks upon addition of calcium ions. The gelation ability of alendronate-functionalized PEG was greatly dependent on the number of arms and arm molecular weight. After mixing polymer and calcium solutions, the formed hydrogels could be cut and then brought back together without any visible interface. After 2 minutes of contact, their connection was strong enough to allow for stretching without tearing through the previous fracture surface. Oscillatory rheology showed that the hydrogels recovered between 70 and 100% of the original storage and loss modulus after rupture. Frequency sweep measurements revealed a liquid-like behavior at lower frequencies and solid-like at high frequencies. Shifting frequency curves obtained at different calcium and polymer concentrations, all data collapsed in a single common master curve. This time-concentration superposition reveals a common relaxation mechanism intrinsically connected to the calcium-bisphosphonate complexation equilibrium.

Project description:We developed a new muco-adhesive hydrogel composed of cationic guar gum (CGG) and boric acid (BA). The CGG-BA precursor solution of 0.5-2% w/v concentration exhibited fluidity at low pH (3-5), while gelation occurred within 1 min at physiological pH (7-8) conditions. Scanning electron microscopy and Fourier-transform infrared spectroscopy results confirmed the change in physical and chemical behavior, respectively, with change in pH. The pH-responsive self-healing ability was analyzed through microscopy and rheology. CGG-BA hydrogels showed good self-healing property at pH 7.4. The in vitro biocompatibility test of the hydrogel studied using NIH3T3 and NHEK cells showed that it was non-toxic at concentrations of CGG-BA below 2% w/v. Ex vivo mucoadhesive tests confirmed the hydrogel's potential for use as a muco-adhesive. Burst pressure tests were conducted using pig esophageal mucosa and the results showed that at pH 7.4, 1% w/v CGG-BA self-healable hydrogel resisted about 8 ± 2 kPa pressure, comparable to that of Fibrin glue. This was higher than that at solution (pH 5) and brittle gel (pH 10) conditions. To confirm the good adhesive strength of the self-healable hydrogels, lap shear tests conducted, resulted in adhesive strengths measured in the range of 1.0 ± 0.5-2.0 ± 0.6 kPa, which was also comparable to fibrin glue control 1.8 ± 0.6 kPa. Hydrogel weight measurements showed that 40-80% gel lasted under physiological conditions for 10 h. The results suggest that CGG-BA hydrogel has potential as a pH responsive mucosal protectant biomaterial.

Project description:Synthetic diblock copolypeptides were designed to incorporate oppositely charged ionic segments that form β-sheet-structured hydrogel assemblies via polyion complexation when mixed in aqueous media. The observed chain conformation directed assembly was found to be required for efficient hydrogel formation and provided distinct and useful properties to these hydrogels, including self-healing after deformation, microporous architecture, and stability against dilution in aqueous media. While many promising self-assembled materials have been prepared using disordered or liquid coacervate polyion complex (PIC) assemblies, the use of ordered chain conformations in PIC assemblies to direct formation of new supramolecular morphologies is unprecedented. The promising attributes and unique features of the β-sheet-structured PIC hydrogels described here highlight the potential of harnessing conformational order derived from PIC assembly to create new supramolecular materials.

Project description:Expanding the useful lifespan of materials is becoming highly desirable, and self-healing and self-repairing materials may become valuable commodities. The formation of supramolecular materials through host-guest interactions is a powerful method to create non-conventional materials. Here we report the formation of supramolecular hydrogels and their redox-responsive and self-healing properties due to host-guest interactions. We employ cyclodextrin (CD) as a host molecule because it is environmentally benign and has diverse applications. A transparent supramolecular hydrogel quickly forms upon mixing poly(acrylic acid) (pAA) possessing β-CD as a host polymer with pAA possessing ferrocene as a guest polymer. Redox stimuli induce a sol-gel phase transition in the supramolecular hydrogel and can control self-healing properties such as re-adhesion between cut surfaces.

Project description:Inspired by smart biological tissues, artificial muscle-like actuators offer fascinating prospects due to their distinctive shape transformation and self-healing function under external stimuli. However, further practical application is hindered by the lack of simple and general routes to fabricate ingenious soft materials with anisotropic responsiveness. Here, we describe a general in situ polymerization strategy for the fabrication of anisotropic hydrogels composed of highly-ordered lamellar network crosslinked by the metal nanostructure assemblies, accompanied with remarkably anisotropic performances on mechanical, optical, de-swelling and swelling behaviors. Owing to the dynamic thiolate-metal coordination as healing motifs, the composites exhibit rapid and efficient multi-responsive self-healing performance under NIR irradiation and low pH condition. Dependent on well-defined anisotropic structures, the hydrogel presents controllable solvent-responsive mechanical actuating performance. Impressively, the integrated device through a healing-induced assembly way can deliver more complicated, elaborate forms of actuation, demonstrating its great potentials as superior soft actuators like smart robots.
