Catechol-Functionalized Synthetic Polymer as a Dental Adhesive to Contaminated Dentin Surface for a Composite Restoration.

ABSTRACT: This study reports a synthetic polymer functionalized with catechol groups as dental adhesives. We hypothesize that a catechol-functionalized polymer functions as a dental adhesive for wet dentin surfaces, potentially eliminating the complications associated with saliva contamination. We prepared a random copolymer containing catechol and methoxyethyl groups in the side chains. The mechanical and adhesive properties of the polymer to dentin surface in the presence of water and salivary components were determined. It was found that the new polymer combined with an Fe(3+) additive improved bond strength of a commercial dental adhesive to artificial saliva contaminated dentin surface as compared to a control sample without the polymer. Histological analysis of the bonding structures showed no leakage pattern, probably due to the formation of Fe-catechol complexes, which reinforce the bonding structures. Cytotoxicity test showed that the polymers did not inhibit human gingival fibroblast cells proliferation. Results from this study suggest a potential to reduce failure of dental restorations due to saliva contamination using catechol-functionalized polymers as dental adhesives.

Project description:ObjectiveAssess the ability of an antimicrobial drug-releasing resin adhesive, containing octenidine dihydrochloride (OCT)-silica co-assembled particles (DSPs), to enhance the biostability and preserve the interfacial fracture toughness (FT) of composite restorations bonded to dentin. Enzyme-catalyzed degradation compromises the dental restoration-tooth interface, increasing cariogenic bacterial infiltration. In addition to bacterial ingress inhibition, antimicrobial-releasing adhesives may exhibit direct interfacial biodegradation inhibition as an additional benefit.MethodsMini short-rod restoration bonding specimens with total-etch adhesive with/without 10% wt. DSPs were made. Interfacial fracture toughness (FT) was measured as-manufactured or post-incubation in simulated human salivary esterase (SHSE) for up to 6-months. Effect of OCT on SHSE and whole saliva/bacterial enzyme activity was assessed. Release of OCT outside the restoration interface was assessed.ResultsNo deleterious effect of DSPs on initial bonding capacity was observed. Aging specimens in SHSE reduced FT of control but not DSP-adhesive-bonded specimens. OCT inhibited SHSE degradation of adhesive monomer and may inhibit endogenous proteases. OCT inhibited bacterial esterase and collagenase. No endogenous collagen breakdown was detected in the present study. OCT increased human saliva degradative esterase activity below its minimum inhibitory concentration towards S. mutans (MIC), but inhibited degradation above MIC. OCT release outside restoration margins was below detection.SignificanceDSP-adhesive preserves the restoration bond through a secondary enzyme-inhibitory effect of released OCT, which is virtually confined to the restoration interface microgap. Enzyme activity modulation may produce a positive-to-negative feedback switch, by increasing OCT concentration via biodegradation-triggered release to an effective dose, then subsequently slowing degradation and degradation-triggered release.

Project description:Uranium (U) contamination of drinking water often affects communities with limited resources, presenting unique technology challenges for U6+ treatment. Here, we develop a suite of chemically functionalized polymer (polyacrylonitrile; PAN) nanofibers for low pressure reactive filtration applications for U6+ removal. Binding agents with either nitrogen-containing or phosphorous-based (e.g., phosphonic acid) functionalities were blended (at 1-3 wt.%) into PAN sol gels used for electrospinning, yielding functionalized nanofiber mats. For comparison, we also functionalized PAN nanofibers with amidoxime (AO) moieties, a group well-recognized for its specificity in U6+ uptake. For optimal N-based (Aliquat® 336 or Aq) and P-containing [hexadecylphosphonic acid (HPDA) and bis(2-ethylhexyl)phosphate (HDEHP)] binding agents, we then explored their use for U6+ removal across a range of pH values (pH 2-7), U6+ concentrations (up to 10 μM), and in flow through systems simulating point of use (POU) water treatment. As expected from the use of quaternary ammonium groups in ion exchange, Aq-containing materials appear to sequester U6+ by electrostatic interactions; while uptake by these materials is limited, it is greatest at circumneutral pH where positively charged N groups bind negatively charged U6+ complexes. In contrast, HDPA and HDEHP perform best at acidic pH representative of mine drainage, where surface complexation of the uranyl cation likely drives uptake. Complexation by AO exhibited the best performance across all pH values, although U6+ uptake via surface precipitation may also occur near circumneutral pH value and at high (10 μM) dissolved U6+ concentrations. In simulated POU treatment studies using a dead-end filtration system, we observed U removal in AO-PAN systems that is insensitive to common co-solutes in groundwater (e.g., hardness and alkalinity). While more research is needed, our results suggest that only 80 g (about 0.2 lbs.) of AO-PAN filter material would be needed to treat an individual's water supply (contaminated at ten-times the U.S. EPA Maximum Contaminant Level for U) for one year.
