Concentric Magnetic Structures for Magnetophoretic Bead Collection, Cell Trapping and Analysis of Cell Morphological Changes Caused by Local Magnetic Forces.

ABSTRACT: Concentric magnetic structures (ring and square) with domain wall (DW) pinning geometry are designed for biological manipulation. Magnetic beads collection was firstly demonstrated to analyse the local magnetic field generated by DWs and the effective regions to capture magnetic targets of size 1 ?m. Primary mouse embryonic fibroblasts (MEFs) are magnetically labeled by internalizing poly (styrene sulfonic acid) stabilized magnetic nanoparticles (PSS-MNPs) and then are selectively trapped by head-to-tail DWs (HH DWs) or tail-to-tail DWs (TT DWs) to be arranged into linear shape or cross shape. The morphologies and the nuclear geometry of the cells growing on two kinds of concentric magnetic structures are shown to be distinctive. The intracellular magnetic forces generated by the local magnetic field of DWs are found to influence the behaviour of cells.

Magnetophoretic transistors in a tri-axial magnetic field.

Project description:The ability to direct and sort individual biological and non-biological particles into spatially addressable locations is fundamentally important to the emerging field of single cell biology. Towards this goal, we demonstrate a new class of magnetophoretic transistors, which can switch single magnetically labeled cells and magnetic beads between different paths in a microfluidic chamber. Compared with prior work on magnetophoretic transistors driven by a two-dimensional in-plane rotating field, the addition of a vertical magnetic field bias provides significant advantages in preventing the formation of particle clumps and in better replicating the operating principles of circuits in general. However, the three-dimensional driving field requires a complete redesign of the magnetic track geometry and switching electrodes. We have solved this problem by developing several types of transistor geometries which can switch particles between two different tracks by either presenting a local energy barrier or by repelling magnetic objects away from a given track, hereby denoted as "barrier" and "repulsion" transistors, respectively. For both types of transistors, we observe complete switching of magnetic objects with currents of ∼40 mA, which is consistent over a range of particle sizes (8-15 μm). The switching efficiency was also tested at various magnetic field strengths (50-90 Oe) and driving frequencies (0.1-0.6 Hz); however, we again found that the device performance only weakly depended on these parameters. These findings support the use of these novel transistor geometries to form circuit architectures in which cells can be placed in defined locations and retrieved on demand.

Magnetophoretic Conductors and Diodes in a 3D Magnetic Field.

Project description:We demonstrate magnetophoretic conductor tracks that can transport single magnetized beads and magnetically labeled single cells in a 3-dimensional time-varying magnetic field. The vertical field bias, in addition to the in-plane rotating field, has the advantage of reducing the attraction between particles, which inhibits the formation of particle clusters. However, the inclusion of a vertical field requires the re-design of magnetic track geometries which can transport magnetized objects across the substrate. Following insights from magnetic bubble technology, we found that successful magnetic conductor geometries defined in soft magnetic materials must be composed of alternating sections of positive and negative curvature. In addition to the previously studied magnetic tracks taken from the magnetic bubble literature, a drop-shape pattern was found to be even more adept at transporting small magnetic beads and single cells. Symmetric patterns are shown to achieve bi-directional conduction, whereas asymmetric patterns achieve unidirectional conduction. These designs represent the electrical circuit corollaries of the conductor and diode, respectively. Finally, we demonstrate biological applications in transporting single cells and in the size based separation of magnetic particles.

Integration of Magnetic Bead-Based Cell Selection into Complex Isolations.

Project description:Magnetic bead-based analyte capture has emerged as a ubiquitous method in cell isolation, enabling the highly specific capture of target populations through simple magnetic manipulation. To date, no "one-size fits all" magnetic bead has been widely adopted leading to an overwhelming number of commercial beads. Ultimately, the ideal bead is one that not only facilitates cell isolation but also proves compatible with the widest range of downstream applications and analytic endpoints. Despite the diverse offering of sizes, coatings, and conjugation chemistries, few studies exist to benchmark the performance characteristics of different commercially available beads; importantly, these bead characteristics ultimately determine the ability of a bead to integrate into the user's assay. In this report, we evaluate bead-based cell isolation considerations, approaches, and results across a subset of commercially available magnetic beads (Dynabeads FlowComps, Dynabeads CELLection, GE Healthcare Sera-Mag SpeedBeads streptavidin-blocked magnetic particles, Dynabeads M-270s, Dynabeads M-280s) to compare and contrast both capture-specific traits (i.e., purity, capture efficacy, and contaminant isolations) and endpoint compatibility (i.e., protein localization, fluorescence imaging, and nucleic acid extraction). We identify specific advantages and contexts of use in which distinct bead products may facilitate experimental goals and integrate into downstream applications.

In situ single cell detection via microfluidic magnetic bead assay.

Project description:We present a single cell detection device based on magnetic bead assay and micro Coulter counters. This device consists of two successive micro Coulter counters, coupled with a high gradient magnetic field generated by an external magnet. The device can identify single cells in terms of the transit time difference of the cell through the two micro Coulter counters. Target cells are conjugated with magnetic beads via specific antibody and antigen binding. A target cell traveling through the two Coulter counters interacts with the magnetic field, and have a longer transit time at the 1st counter than that at the 2nd counter. In comparison, a non-target cell has no interaction with the magnetic field, and hence has nearly the same transit times through the two counters. Each cell passing through the two counters generates two consecutive voltage pulses one after the other; the pulse widths and magnitudes indicating the cell's transit times through the counters and the cell's size respectively. Thus, by measuring the pulse widths (transit times) of each cell through the two counters, each single target cell can be differentiated from non-target cells even if they have similar sizes. We experimentally proved that the target human umbilical vein endothelial cells (HUVECs) and non-target rat adipose-derived stem cells (rASCs) have significant different transit time distribution, from which we can determine the recognition regions for both cell groups quantitatively. We further demonstrated that within a mixed cell population of rASCs and HUVECs, HUVECs can be detected in situ and the measured HUVECs ratios agree well with the pre-set ratios. With the simple device structure and easy sample preparation, this method is expected to enable single cell detection in a continuous flow and can be applied to facilitate general cell detection applications such as stem cell identification and enumeration.

MAPS-seq: magnetic bead-assisted parallel single-cell gene expression profiling.

Project description:Recently developed single-cell RNA sequencing methods allow the simultaneous profiling of the transcriptomes of thousands of individual cells. However, current methods still require advanced equipment or entail substantial waste of reagents. Here, we introduce magnetic bead-assisted parallel single-cell gene expression sequencing (MAPS-seq), a microwell-based method that pools samples before the reverse transcription step, increasing the ease of sample preparation and reducing reagent waste. Moreover, because this method uses universal reagents and standard molecular biology lab instruments, it is easy to implement, even in labs that have not previously conducted single-cell RNA sequencing. We validated our method by demonstrating that it can generate gene expression data at the single-cell level. We then applied the MAPS-seq method to analyze 237 human myelogenous leukemia cells treated with one of three different drugs or dimethyl sulfoxide. We observed transcriptional changes and identified marker genes that indicate a drug response. Furthermore, the MAPS-seq method produced data of comparable quality to those of existing single-cell RNA sequencing methods. Consequently, we expect that our method will provide researchers with a more accessible, less wasteful, and less burdensome method for investigating the transcriptomes of individual cells.

Solute trapping in Al-Cu alloys caused by a 29 Tesla super high static magnetic field.

Project description:Solidification of Al-Cu alloys has been investigated using a 29 Tesla super high static magnetic field (SHSMF). The results show that, by imposing a 29 Tesla SHSMF, the size of primary phases and spacing of eutectic structure have been refined through the increase of undercooling which results from the suppression of diffusion coefficient. The diffusion coefficient of atoms in the liquid matrix decreases to be about 1.2 × 10-12 m2/s. The lattice constants are reduced and high dislocation density forms in the primary phase, which induces a solute trapping effects. The spacing of (110) plane in Al2Cu is corrected to be 4.3123 Å and 4.2628 Å for Al-40 wt.%Cu alloys treated without and with a SHSMF. The spacing of (111) plane in Al is corrected to be 2.3351 Å and 2.3258 Å for Al-26 wt.%Cu alloys treated without and with a SHSMF. The compression yield strength has been improved by about 42% from 268 MPa to 462 MPa for Al-26 wt.%Cu and 42.5% from 248 MPa to 431 MPa for Al-40 wt.%Cu. The maximum elastic strain increases from about 2% to 4.3% for Al-26 wt.%Cu and from 2% to 4% for Al-40 wt.%Cu. It is expected that SHSMF is beneficial to process materials with high mechanical properties.

Soft Polymer Magnetic Nanocomposites: Microstructure Patterning by Magnetophoretic Transport and Self-Assembly.

Project description:A method to produce and pattern magnetic microstructure in a soft-polymer matrix is demonstrated. An externally applied magnetic field is used to influence the dynamics of magnetophoretic transport and dipolar self-assembly of magnetic nanoparticle clusters in the liquid precursor of poly-dimethylsiloxane (PDMS). Magnetic nanoparticles agglomerate by an interplay of van der Waals forces and dipolar interactions to form anisotropic clusters. These clusters are concentrated on a substrate by magnetophoresis, wherein they self-organize by dipolar interactions to form microscopic filaments. The polymer is cured in the presence of the magnetic field to preserve the microstructure shape. The externally applied magnetic field and its gradient are the two main control variables of interest when considering magnetic control during nanoparticle self-assembly. Their influence on microstructure geometry is investigated through correlations with the height of a characteristic self-assembled filament, fraction of the substrate area covered by the microstructure and its shape anisotropy. These relations enable a priori design.

Capturing magnetic bead-based arrays using perpendicular magnetic anisotropy.

Project description:Designing and implementing means of locally trapping magnetic beads and understanding the factors underlying the bead capture force are important steps toward advancing the capture-release process of magnetic particles for biological applications. In particular, capturing magnetically labeled cells using magnetic microstructures with perpendicular magnetic anisotropy (PMA) will enable an approach to cell manipulation for emerging lab-on-a-chip devices. Here, a Co (0.2 nm)/Ni (0.4 nm) multilayered structure was designed to exhibit strong PMA and large saturation magnetization (Ms ). Finite element simulations were performed to assess the dependence of the capture force on the value of Ms . The simulated force profile indicated the largest force at the perimeter of the disks. Arrays of Co/Ni disk structures of (4-7) μm diameter were fabricated and tested in a microchannel with suspended fluorescent magnetic beads. The magnetic beads were captured and localized to the edge of the disks as predicted by the simulations. This approach has been demonstrated to enable uniform assembly of magnetic beads without external fields and may provide a pathway toward precise cell manipulation methods.

Dielectrophoretic Traps for Efficient Bead and Cell Trapping and Formation of Aggregates of Controlled Size and Composition

Project description:We present a microfluidic dielectrophoretic-actuated system designed to trap chosen single-cell and form controlled cell aggregates. A novel method is proposed to characterize the efficiency of the dielectrophoretic trapping, considering the flow speed but also the heat generated by the traps as limiting criteria in cell-safe manipulation. Two original designs with different manufacturing processes are experimentally compared. The most efficient design is selected and the cell membrane integrity is monitored by fluorescence imaging to guarantee a safe-cell trapping. Design rules are suggested to adapt the traps to multiple-cells trapping and are experimentally validated as we formed aggregates of controlled size and composition with two different types of cells. We provide hereby a simple manufactured tool allowing the controlled manipulation of particles for the composition of multicellular assemblies.

Trapping atoms using nanoscale quantum vacuum forces.

Project description:Quantum vacuum forces dictate the interaction between individual atoms and dielectric surfaces at nanoscale distances. For example, their large strengths typically overwhelm externally applied forces, which makes it challenging to controllably interface cold atoms with nearby nanophotonic systems. Here we theoretically show that it is possible to tailor the vacuum forces themselves to provide strong trapping potentials. Our proposed trapping scheme takes advantage of the attractive ground-state potential and adiabatic dressing with an excited state whose potential is engineered to be resonantly enhanced and repulsive. This procedure yields a strong metastable trap, with the fraction of excited-state population scaling inversely with the quality factor of the resonance of the dielectric structure. We analyse realistic limitations to the trap lifetime and discuss possible applications that might emerge from the large trap depths and nanoscale confinement.
