APPLYING A SPATIOTEMPORAL MODEL FOR LONGITUDINAL CARDIAC IMAGING DATA.

ABSTRACT: Longitudinal imaging studies have both spatial and temporal correlation among the multiple outcome measurements from a subject. Statistical methods of analysis must properly account for this autocorrelation. In this work we discuss how a linear model with a separable parametric correlation structure could be used to analyze data from such a study. The goal of this paper is to provide an easily understood description of how such a model works and discuss how it can be applied to real data. Model assumptions are discussed and the process of selecting a working correlation structure is thoroughly discussed. The steps necessitating collaboration between statistical and scientific investigators have been highlighted, as have considerations for missing data or uneven follow-up. The results from a completed longitudinal cardiac imaging study were considered for illustration purposes. The data comes from a clinical trial for medical therapy for patients with mitral regurgitation, with repeated measurements taken at sixteen locations from the left ventricle to measure disease progression. The spatiotemporal correlation model was compared to previously used summary measures to demonstrate improved power as well as increased flexibility in the use of time- and space-varying predictors.

Project description:We propose a novel framework to learn the spatiotemporal variability in longitudinal 3D shape data sets, which contain observations of objects that evolve and deform over time. This problem is challenging since surfaces come with arbitrary parameterizations and thus, they need to be spatially registered. Also, different deforming objects, hereinafter referred to as 4D surfaces, evolve at different speeds and thus they need to be temporally aligned. We solve this spatiotemporal registration problem using a Riemannian approach. We treat a 3D surface as a point in a shape space equipped with an elastic Riemannian metric that measures the amount of bending and stretching that the surfaces undergo. A 4D surface can then be seen as a trajectory in this space. With this formulation, the statistical analysis of 4D surfaces can be cast as the problem of analyzing trajectories embedded in a nonlinear Riemannian manifold. However, performing the spatiotemporal registration, and subsequently computing statistics, on such nonlinear spaces is not straightforward as they rely on complex nonlinear optimizations. Our core contribution is the mapping of the surfaces to the space of Square-Root Normal Fields (SRNF) where the [Formula: see text] metric is equivalent to the partial elastic metric in the space of surfaces. Thus, by solving the spatial registration in the SRNF space, the problem of analyzing 4D surfaces becomes the problem of analyzing trajectories embedded in the SRNF space, which has a euclidean structure. In this paper, we develop the building blocks that enable such analysis. These include: (1) the spatiotemporal registration of arbitrarily parameterized 4D surfaces even in the presence of large elastic deformations and large variations in their execution rates; (2) the computation of geodesics between 4D surfaces; (3) the computation of statistical summaries, such as means and modes of variation, of collections of 4D surfaces; and (4) the synthesis of random 4D surfaces. We demonstrate the performance of the proposed framework using 4D facial surfaces and 4D human body shapes.

Project description:Cardiac strain imaging (CSI) plays a critical role in the detection of myocardial motion abnormalities. Displacement estimation is an important processing step to ensure the accuracy and precision of derived strain tensors. In this paper, we propose and implement Spatiotemporal Bayesian regularization (STBR) algorithms for two-dimensional (2-D) normalized cross-correlation (NCC) based multi-level block matching along with incorporation into a Lagrangian cardiac strain estimation framework. Assuming smooth temporal variation over a short span of time, the proposed STBR algorithm performs displacement estimation using at least four consecutive ultrasound radio-frequency (RF) frames by iteratively regularizing 2-D NCC matrices using information from a local spatiotemporal neighborhood in a Bayesian sense. Two STBR schemes are proposed to construct Bayesian likelihood functions termed as Spatial then Temporal Bayesian (STBR-1) and simultaneous Spatiotemporal Bayesian (STBR-2). Radial and longitudinal strain estimated from a finite-element-analysis (FEA) model of realistic canine myocardial deformation were utilized to quantify strain bias, normalized strain error and total temporal relative error (TTR). Statistical analysis with one-way analysis of variance (ANOVA) showed that all Bayesian regularization methods significantly outperform NCC with lower bias and errors (p < 0.001). However, there was no significant difference among Bayesian methods. For example, mean longitudinal TTR for NCC, SBR, STBR-1 and STBR-2 were 25.41%, 9.27%, 10.38% and 10.13% respectively An in vivo feasibility study using RF data from ten healthy mice hearts were used to compare the elastographic signal-to-noise ratio (SNR e ) calculated using stochastic analysis. STBR-2 had the highest expected SNR e both for radial and longitudinal strain. The mean expected SNR e values for accumulated radial strain for NCC, SBR, STBR-1 and STBR-2 were 5.03, 9.43, 9.42 and 10.58, respectively. Overall results suggest that STBR improves CSI in vivo.
