Methods to Assess Measurement Error in Questionnaires of Sedentary Behavior.

ABSTRACT: Sedentary behavior has already been associated with mortality, cardiovascular disease, and cancer. Questionnaires are an affordable tool for measuring sedentary behavior in large epidemiological studies. Here, we introduce and evaluate two statistical methods for quantifying measurement error in questionnaires. Accurate estimates are needed for assessing questionnaire quality. The two methods would be applied to validation studies that measure a sedentary behavior by both questionnaire and accelerometer on multiple days. The first method fits a reduced model by assuming the accelerometer is without error, while the second method fits a more complete model that allows both measures to have error. Because accelerometers tend to be highly accurate, we show that ignoring the accelerometer's measurement error, can result in more accurate estimates of measurement error in some scenarios. In this manuscript, we derive asymptotic approximations for the Mean-Squared Error of the estimated parameters from both methods, evaluate their dependence on study design and behavior characteristics, and offer an R package so investigators can make an informed choice between the two methods. We demonstrate the difference between the two methods in a recent validation study comparing Previous Day Recalls (PDR) to an accelerometer-based ActivPal.

Project description:Studies on the effects of air pollution and more generally environmental exposures on health require measurements of pollutants, which are affected by measurement error. This is a cause of bias in the estimation of parameters relevant to the study and can lead to inaccurate conclusions when evaluating associations among pollutants, disease risk and biomarkers. Although the presence of measurement error in such studies has been recognized as a potential problem, it is rarely considered in applications and practical solutions are still lacking. In this work, we formulate Bayesian measurement error models and apply them to study the link between air pollution and omic signals. The data we use stem from the "Oxford Street II Study", a randomized crossover trial in which 60 volunteers walked for two hours in a traffic-free area (Hyde Park) and in a busy shopping street (Oxford Street) of London. Metabolomic measurements were made in each individual as well as air pollution measurements, in order to investigate the association between short-term exposure to traffic related air pollution and perturbation of metabolic pathways. We implemented error-corrected models in a classical framework and used the flexibility of Bayesian hierarchical models to account for dependencies among omic signals, as well as among different pollutants. Models were implemented using traditional Markov Chain Monte Carlo (MCMC) simulative methods as well as integrated Laplace approximation. The inclusion of a classical measurement error term resulted in variable estimates of the association between omic signals and traffic related air pollution measurements, where the direction of the bias was not predictable a priori. The models were successful in including and accounting for different correlation structures, both among omic signals and among different pollutant exposures. In general, more associations were identified when the correlation among omics and among pollutants were modeled, and their number increased when a measurement error term was additionally included in the multivariate models (particularly for the associations between metabolomics and NO2).

Project description:Measurement errors commonly occur in 24-h hormonal data and may affect the outcomes of such studies. Measurement errors often appear as outliers in such data sets; however, no well-established method is available for their automatic detection. In this study, we aimed to compare performances of different methods for outlier detection in hormonal serial data. Hormones (glucose, insulin, thyroid-stimulating hormone, cortisol, and growth hormone) were measured in blood sampled every 10 min for 24 h in 38 participants of the Leiden Longevity Study. Four methods for detecting outliers were compared: (1) eyeballing, (2) Tukey's fences, (3) stepwise approach, and (4) the expectation-maximization (EM) algorithm. Eyeballing detects outliers based on experts' knowledge, and the stepwise approach incorporates physiological knowledge with a statistical algorithm. Tukey's fences and the EM algorithm are data-driven methods, using interquartile range and a mathematical algorithm to identify the underlying distribution, respectively. The performance of the methods was evaluated based on the number of outliers detected and the change in statistical outcomes after removing detected outliers. Eyeballing resulted in the lowest number of outliers detected (1.0% of all data points), followed by Tukey's fences (2.3%), the stepwise approach (2.7%), and the EM algorithm (11.0%). In all methods, the mean hormone levels did not change materially after removing outliers. However, their minima were affected by outlier removal. Although removing outliers affected the correlation between glucose and insulin on the individual level, when averaged over all participants, none of the 4 methods influenced the correlation. Based on our results, the EM algorithm is not recommended given the high number of outliers detected, even where data points are physiologically plausible. Since Tukey's fences is not suitable for all types of data and eyeballing is time-consuming, we recommend the stepwise approach for outlier detection, which combines physiological knowledge and an automated process.
