A Smart pH-Responsive Three Components Luminescent Hydrogel.

ABSTRACT: In this study, we report a novel three-component luminescent hydrogel, which is composed of amino acid derivatives (N,N'-di valine-3,4,9,10-perylenetetracarboxylic acid, NVPD), riboflavin (RF), and melamine (MM). The three-component hydrogel is attributed to multiple hydrogen bonds and the strong π-π stacking interaction between these molecules. Based on the strong hydrogen bonding of the gelator, when the reversible process between the gel and the solution take places it changes the pH of the system from 6.1 to 10.6. In addition, green fluorescence could be the emissive of the hydrogel under 498 nm and the conversion process of the aggregation state repeated reversibly by altering the value of ambient pH. This pH-responsive luminescent gel may display potential for use in nano pH sensors.

A smart thermo- and pH-responsive microfiltration membrane based on three-dimensional inverse colloidal crystals.

Project description:In this paper, a thermo- and pH-responsive microfiltration membrane was prepared based on three-dimensional (3D) inverse colloidal crystals (ICC). To manufacture the smart ICC membrane, the typical thermo-responsive N-isopropylacrylamide (NIPAM) and pH-responsive methacrylic acid (MAA) were polymerized inside silica colloidal crystals. The smart ICC membranes were characterized by SEM, IR and contact angle measurements. Moreover, the permeability of smart microfiltration membrane was carried out by the KCl diffusion tests. The result showed that effective diameter of the polymer ICC membrane can be reversible tuned by temperature and pH. Besides, the functional ICC membrane showed outstanding temperature- and pH-responsive gating property, which was applied to separate particles of different sizes. The savvy environment-responsive gating membranes have potential uses in filtration, separation, purification, sensor and other applications.

A pH-sensitive, stimuli-responsive, superabsorbent, smart hydrogel from psyllium (<i>Plantago ovata</i>) for intelligent drug delivery.

Project description:Herein, we report a polysaccharide-based hydrogel isolated from psyllium husk (a well-known dietary fiber) and evaluated for its swelling properties in deionized water (DW) at different physiological pH values, i.e., 1.2, 6.8 and 7.4. Swelling of psyllium hydrogel (PSH) in DW under the influence of temperature and at different concentrations of NaCl and KCl solutions was also examined. A pH-dependent swelling pattern of PSH was observed following the order DW > pH 7.4 > pH 6.8 > pH 1.2. Stimuli-responsive swelling and deswelling (on-off switching) behavior of PSH was observed in DW and ethanol, DW and normal saline, at pH 7.4 and pH 1.2 environments, respectively. Similar swelling behavior and on-off switching attribute of PSH-containing tablets indicated the unaltered nature of PSH even after compression. Scanning electron micrographs of swollen and then freeze-dried PSH via transverse and longitudinal cross-sections revealed hollow channels with an average pore size of 6 ± 2 μm. Furthermore, PSH concentration-dependent sustained release of theophylline from tablet formulation was witnessed for >15 h following the non-Fickian diffusion mechanism. Subacute toxicity studies revealed the non-toxic nature of PSH. Therefore, dietary fiber-based material, i.e., PSH could be a valuable pharmaceutical excipient for intelligent and targeted drug delivery.

Antibacterial and Cytocompatible pH-Responsive Peptide Hydrogel.

Project description:A short peptide, FHHF-11, was designed to change stiffness as a function of pH due to changing degree of protonation of histidines. As pH changes in the physiologically relevant range, G' was measured at 0 Pa (pH 6) and 50,000 Pa (pH 8). This peptide-based hydrogel is antimicrobial and cytocompatible with skin cells (fibroblasts). It was demonstrated that the incorporation of unnatural AzAla tryptophan analog residue improves the antimicrobial properties of the hydrogel. The material developed can have a practical application and be a paradigm shift in the approach to wound treatment, and it will improve healing outcomes for millions of patients each year.

Biofilm interfacial acidity evaluation by pH-Responsive luminescent nanoparticle films.

Project description:Biofilms are dense bacterial colonies that may adhere to the surfaces of medical devices and are major contributors to infections. These colonies are characterized by a self-produced matrix of extracellular polymeric substances (EPS). Bacterial biofilms are difficult to treat with the commonly used antibiotics partially because of their poor diffusion through the EPS and therefore require new targeted strategies to effectively fight them. Biofilms may produce an acidic microenvironment which can be exploited to design such targeted treatment strategies. However, there is currently a lack of high-throughput ways to determine the acidity of biofilms at their interface with the medical device. Here, a novel all-inorganic pH responsive system is developed from luminescent carbonated hydroxyapatite nanoparticles doped with Eu3+ ions which can determine the biofilm acidity fluorometrically due to carbonate removal in acidic environments that directly affects the nanoparticle luminescence. The pH responsive nanoparticles are in-situ deposited during their production onto substrates on which a variety of clinically-relevant biofilms are grown. The acidity of their interfacial (micro)environment depends on the bacterial species and strain even when differences in biofilm biomass are considered.

pH Responsive Antibacterial Hydrogel Utilizing Catechol-Boronate Complexation Chemistry.

Project description:Bacteria such as Methicillin-resistant Staphylococcus aureus (MRSA) causes acidic microenvironment during infection. A biomaterial that exhibits tunable antimicrobial property in a pH dependent manner is potentially attractive. Herein, we presented a novel antibacterial hydrogel consisting of pH responsive and reversible catechol-boronate linkage formed between intrinsically bactericidal chlorinated catechol (catechol-Cl) and phenylboronic acid. Fourier transformed infrared spectroscopy (FTIR), oscillatory rheometry, and Johnson Kendall Roberts (JKR) contact mechanics testing confirmed the formation and dissociation of the complex in a pH dependent manner. When the hydrogel was treated with an acidic buffer (pH 3), the hydrogel exhibited excellent antimicrobial property against multiple strains of Gram-positive and negative bacteria including MRSA (up to 4 log10 reduction from 108 colony forming units/mL). At an acidic pH, catechol-Cl was unbound from the phenylboronic acid and available for killing bacteria. Conversely, when the hydrogel was treated with a basic buffer (pH 8.5), the hydrogel lost its antimicrobial property but also became non-cytotoxic. At a basic pH, the formation of catechol-boronate complex effectively reduce the exposure of the cytotoxic catechol-Cl to the surrounding. When further incubating the hydrogel in an acidic pH, the reversible complex dissociated to re-expose catechol-Cl and the hydrogel recovered its antibacterial property. Overall, the combination of catechol-Cl and phenylboronic acid provide a new strategy for designing hydrogels with pH responsive antibacterial activity and reduced cytotoxicity.

Reversible pH Responsive Bovine Serum Albumin Hydrogel Sponge Nanolayer.

Project description:A pH dependent reversible sponge like behavior of a bovine serum albumin (BSA) nanolayer adsorbed at the gold-saline interface is revealed by quartz crystal microbalance with dissipation (QCM-D), atomic force microscope (AFM) and contact angle measurements. During the saline rinsing cycles, the BSA layer adsorbs water molecules at pH 7.0 and releases them at pH 4.5. The phenomenon remains constant and reproducible upon multiple rinsing cycles. The BSA layer thickness also increases upon rinsing with saline at pH 7.0 and reverses back to its original thickness at pH 4.5. Varying ionic strength with water further desorbs more water molecules from the BSA layer, which decreases its mass and thickness. However, upon both pH and ionic strength changes, all the BSA molecules remain adsorbed irreversibly at the gold interface and only the sorption of water molecules occurs. The study aims at engineering high efficiency pH-responsive biodiagnostics and drug delivery systems.

Smart Material Hydrogel Transfer Devices Fabricated with Stimuli-Responsive Silk-Elastin-Like Proteins.

Project description:Three-dimensional organoid tissue culture models are a promising approach for the study of biological processes including diseases. Advances in these tissue culture technologies improve in vitro analysis compared to standard 2D cellular approaches and are more representative of the physiological environment. However, a major challenge associated with organoid systems stems from the laborious processing involved in the analysis of large numbers of organoids. Here the design, characterization, and application of silk-elastin-like protein-based smart carrier arrays for processing organoids is presented. Fabrication of hydrogel-based carrier systems at room temperature result in organized arrays of organoids that maintain tissue culture plate orientation and could be processed simultaneously for histology. The system works by transfer of the organoids to the hydrogel arrays after which the material is subjected to 65 °C to induce hydrogel contraction to secure the organoids, resulting in multisample constructs and allowing for placement on a microscope slide. Histological processing and immunostaining of these arrayed cerebral organoids analyzed within the contracted silk-elastin-like proteins (SELP) show retention of native organoid features compared to controls without the hydrogel carrier system, thus avoiding any artifacts. These SELP carriers present a useful approach for improving efficiency of scaled organoid screening and processing.

Smart Microparticles with a pH-responsive Macropore for Targeted Oral Drug Delivery.

Project description:The development of a smart microencapsulation system programmed to actively respond to environmental pH change has long been recognized a key technology in pharmaceutical and food sciences. To this end, we developed hollow microparticles (MPs) with self-controlled macropores that respond to environmental pH change, using an Oil-in-Water emulsion technique, for oral drug delivery. We observed that freeze-drying of MPs induced closure of macropores. The closing/opening behavior of macropores was confirmed by exposing MPs encapsulating different ingredients (sulforhodamine b, fluorescent nanoparticles, and lactase) to simulated gastrointestinal (GI) fluids. MPs maintained their intact, closed pore structure in gastric pH, and subsequent exposure to intestinal pH resulted in pore opening and ingredients release. Further, MPs displayed higher protection (>15 times) than commercial lactase formulation, indicating the protective ability of the system against harsh GI conditions. This study showed development of a hybrid MP system combining the advantages of solid particles and hollow capsules, exhibiting easy solvent-free loading mechanism and smart protection/release of encapsulates through controllable macropores. Ultimately, our MPs system strives to usher a new research area in smart drug delivery systems and advance the current oral drug delivery technology by solving major challenges in targeted delivery of pH-sensitive therapeutics.

Bioinspired High-Performance Bilayer, pH-Responsive Hydrogel with Superior Adhesive Property.

Project description:Soft actuators have attracted extensive attention for promising applications in drug delivery, microfluidic switches, artificial muscles and flexible sensors. However, the performance of pH-responsive hydrogel actuators, such as regarding reversible bending property and adhesive property, remains to be improved. In this study, inspired by drosera leaves, we have fabricated high-performance bilayer, pH-responsive poly(acrylamide-acrylic acid-3-acrylamidophenylboronic acid)(P(AAm-AAc-3-AAPBA)) based on the copolymers of AAm, AAc and 3-AAPBA. The pH-sensitive actuators were fabricated by ultraviolet polymerization of the P(AAm-AAc-3-AAPBA) layer as the active actuating layer and the PAAm layer as the auxiliary actuating layer. The effects of pH, glucose concentration and content of 3-AAPBA on bending behavior of P(AAm-AAc-3-AAPBA)/PAAm bilayer actuators were discussed. By tuning the pH of media, the soft actuator could achieve fast and large-amplitude bidirectional bending behaviors. The bending orientation and bending degree can be reversibly and precisely adjusted. More importantly, P(AAm-AAc-3-AAPBA) hydrogel shows good adhesive property in polyvinyl alcohol (PVA) solution; thus, complex structures have been fabricated. In addition, the bilayer hydrogel structures have been demonstrated as soft actuators, bionic flowers and bionic manipulators. The proposed pH-responsive bilayer actuator shows great potential for drug delivery and other medical systems.

Research advances in smart responsive-hydrogel dressings with potential clinical diabetic wound healing properties.

Project description:The treatment of chronic and non-healing wounds in diabetic patients remains a major medical problem. Recent reports have shown that hydrogel wound dressings might be an effective strategy for treating diabetic wounds due to their excellent hydrophilicity, good drug-loading ability and sustained drug release properties. As a typical example, hyaluronic acid dressing (Healoderm) has been demonstrated in clinical trials to improve wound-healing efficiency and healing rates for diabetic foot ulcers. However, the drug release and degradation behavior of clinically-used hydrogel wound dressings cannot be adjusted according to the wound microenvironment. Due to the intricacy of diabetic wounds, antibiotics and other medications are frequently combined with hydrogel dressings in clinical practice, although these medications are easily hindered by the hostile environment. In this case, scientists have created responsive-hydrogel dressings based on the microenvironment features of diabetic wounds (such as high glucose and low pH) or combined with external stimuli (such as light or magnetic field) to achieve controllable drug release, gel degradation, and microenvironment improvements in order to overcome these clinical issues. These responsive-hydrogel dressings are anticipated to play a significant role in diabetic therapeutic wound dressings. Here, we review recent advances on responsive-hydrogel dressings towards diabetic wound healing, with focus on hydrogel structure design, the principle of responsiveness, and the behavior of degradation. Last but not least, the advantages and limitations of these responsive-hydrogels in clinical applications will also be discussed. We hope that this review will contribute to furthering progress on hydrogels as an improved dressing for diabetic wound healing and practical clinical application.
