Project description:Many bacteria in the environment have adapted to the presence of toxic heavy metals. Over the last 30 years, this heavy metal tolerance was the subject of extensive research. The bacterium Cupriavidus metallidurans strain CH34, originally isolated by us in 1976 from a metal processing factory, is considered a major model organism in this field because it withstands milli-molar range concentrations of over 20 different heavy metal ions. This tolerance is mostly achieved by rapid ion efflux but also by metal-complexation and -reduction. We present here the full genome sequence of strain CH34 and the manual annotation of all its genes. The genome of C. metallidurans CH34 is composed of two large circular chromosomes CHR1 and CHR2 of, respectively, 3,928,089 bp and 2,580,084 bp, and two megaplasmids pMOL28 and pMOL30 of, respectively, 171,459 bp and 233,720 bp in size. At least 25 loci for heavy-metal resistance (HMR) are distributed over the four replicons. Approximately 67% of the 6,717 coding sequences (CDSs) present in the CH34 genome could be assigned a putative function, and 9.1% (611 genes) appear to be unique to this strain. One out of five proteins is associated with either transport or transcription while the relay of environmental stimuli is governed by more than 600 signal transduction systems. The CH34 genome is most similar to the genomes of other Cupriavidus strains by correspondence between the respective CHR1 replicons but also displays similarity to the genomes of more distantly related species as a result of gene transfer and through the presence of large genomic islands. The presence of at least 57 IS elements and 19 transposons and the ability to take in and express foreign genes indicates a very dynamic and complex genome shaped by evolutionary forces. The genome data show that C. metallidurans CH34 is particularly well equipped to live in extreme conditions and anthropogenic environments that are rich in metals.

Biomimetic fabrication of superhydrophobic loofah sponge: robust for highly efficient oil-water separation in harsh environments.

Project description:Oil/water separation has become an increasingly important field due to frequent industrial oily wastewater emission and crude oil spill accidents. Herein, we fabricate a robust superhydrophobic loofah sponge via a versatile, environmentally friendly, and low-cost dip coating strategy, which involves the modification of commercial loofah sponge with waterborne polyurea and fused SiO2 nanoparticles without the modification of any toxic low-surface-energy compound. The as-prepared loofah sponge showed excellent superhydrophobic/superoleophilic properties and exhibited robustness for effective oil-water separation in extremely harsh environments (such as 1 M HCl, 1 M NaOH, saturated NaCl solution and hot water higher than 95 °C) due to the remarkably high chemical stability. In addition, the as-prepared loofah sponge was capable of excellent anti-fouling, has self-cleaning ability and could act as the absorber for effective separation of surfactant-free oil-in-water emulsions. More importantly, the as-prepared loofah sponge demonstrated remarkable robustness against strong sandpaper abrasion and finger wipes, while retaining its superhydrophobicity and efficient oil/water separation efficiency even after more than 50 abrasion cycles. This facile and green synthesis approach presented here has the advantage of large-scale fabrication of a multifunctional biomass-based adsorbent material as a promising candidate in anti-fouling, self-cleaning, and versatile water-oil separation.

Genetics, adaptation, and invasion in harsh environments.

Project description:We analyze mathematical models to examine how the genetic basis of fitness affects the persistence of a population suddenly encountering a harsh environment where it would go extinct without evolution. The results are relevant for novel introductions and for an established population whose existence is threatened by a sudden change in the environment. The models span a range of genetic assumptions, including identical loci that contribute to absolute fitness, a two-locus quantitative genetic model with nonidentical loci, and a model with major and minor genes affecting a quantitative trait. We find as a general (though not universal) pattern that prospects for persistence narrow as more loci contribute to fitness, in effect because selection per locus is increasingly weakened with more loci, which can even overwhelm any initial enhancement of fitness that adding loci might provide. When loci contribute unequally to fitness, genes of small effect can significantly reduce extinction risk. Indeed, major and minor genes can interact synergistically to reduce the time needed to evolve growth. Such interactions can also increase vulnerability to extinction, depending not just on how genes interact but also on the initial genetic structure of the introduced, or newly invaded, population.

Project description:We report here the complete genome sequence (2.08 Mb) of Methanohalophilus portucalensis strain FDF-1T, a halophilic methylotrophic methanogen isolated from the sediment of a saltern in Figeria da Foz, Portugal. The average nucleotide identity and DNA-DNA hybridization analyses show that Methanohalophilus mahii, M. halophilus, and M. portucalensis are three different species within the Methanosarcinaceae family.

Electron Communication of Bacillus subtilis in Harsh Environments.

Project description:Elucidating the effect of harsh environments on the activities of microorganisms is important in revealing how microbes withstand unfavorable conditions or evolve mechanisms to counteract those effects, many of which involve electron transfer phenomena. Here we show that the non-acidophilic and non-thermophilic Bacillus subtilis is able to maintain activity after being subjected to extreme temperatures (100°C for up to 8 h) and acidic environments (pH = 1.50 for over 2 years). In the process, our results suggest that B. subtilis utilizes an extracellular electron transfer as an electron communication pathway between B. subtilis and the environment that involves the cofactor nicotinamide adenine dinucleotide as an essential participant to maintain viability. Elucidation of the capability of the non-acidophilic and non-thermophilic strain to maintain viability under these extreme conditions could aid in understanding the cell responses to different environments from the perspective of energy conservation pathways.

Harsh environments promote alloparental care across human societies.

Project description:Alloparental care is central to human life history, which integrates exceptionally short interbirth intervals and large birth size with an extended period of juvenile dependency and increased longevity. Formal models, previous comparative research, and palaeoanthropological evidence suggest that humans evolved higher levels of cooperative childcare in response to increasingly harsh environments. Although this hypothesis remains difficult to test directly, the relative importance of alloparental care varies across human societies, providing an opportunity to assess how local social and ecological factors influence the expression of this behaviour. We therefore, investigated associations between alloparental infant care and socioecology across 141 non-industrialized societies. We predicted increased alloparental care in harsher environments, due to the fitness benefits of cooperation in response to shared ecological challenges. We also predicted that starvation would decrease alloparental care, due to prohibitive energetic costs. Using Bayesian phylogenetic multilevel models, we tested these predictions while accounting for potential confounds as well as for population history. Consistent with our hypotheses, we found increased alloparental infant care in regions characterized by both reduced climate predictability and relatively lower average temperatures and precipitation. We also observed reduced alloparental care under conditions of high starvation. These results provide evidence of plasticity in human alloparenting in response to ecological contexts, comparable to previously observed patterns across avian and mammalian cooperative breeders. This suggests convergent social evolutionary processes may underlie both inter- and intraspecific variation in alloparental care.

A Fully Transparent Resistive Memory for Harsh Environments.

Project description:A fully transparent resistive memory (TRRAM) based on Hafnium oxide (HfO2) with excellent transparency, resistive switching capability, and environmental stability is demonstrated. The retention time measured at 85 °C is over 3 × 10(4) sec, and no significant degradation is observed in 130 cycling test. Compared with ZnO TRRAM, HfO2 TRRAM shows reliable performance under harsh conditions, such as high oxygen partial pressure, high moisture (relative humidity = 90% at 85 °C), corrosive agent exposure, and proton irradiation. Moreover, HfO2 TRRAM fabricated in cross-bar array structures manifests the feasibility of future high density memory applications. These findings not only pave the way for future TRRAM design, but also demonstrate the promising applicability of HfO2 TRRAM for harsh environments.
