Draft whole-genome sequence of the Diaporthe helianthi 7/96 strain, causal agent of sunflower stem canker.

ABSTRACT: Diaporthe helianthi is a fungus pathogenic to sunflower. Virulent strains of this fungus cause stem canker with important yield losses and reduction of oil content. Here we present the first draft whole-genome sequence of the highly virulent isolate D. helianthi strain 7/96, thus providing a useful platform for future research on stem canker of sunflower and fungal genomics. The genome sequence of the D. helianthi isolate 7/96 was deposited at DDBJ/ENA/GenBank under the accession number MAVT00000000 (BioProject PRJNA327798).

Draft genome sequence of Diaporthe aspalathi isolate MS-SSC91, a fungus causing stem canker in soybean.

Project description:Diaporthe aspalathi (Syn. Diaporthe phaseolorum var. meridionalis) is the causal agent of the southern stem canker (SSC) disease in soybean. This disease can kill plants from the middle to the end of the growing season resulting in severe yield loss. The mechanisms of SSC disease development and pathogen invasion of soybean are not fully understood. The genome sequence of D. aspalathi has not been described. In this article, we report the successful assembly of the draft genome sequence of a D. aspalathi isolate, designated MS-SSC91, that was isolated from the stem of a field-grown soybean plant in Mississippi, USA in 2006. This study represents the first reported genome sequence of D. aspalathi in the Diaporthe-Phomopsis complex. The whole genome shotgun sequence of the MS-SSC91 isolate has been deposited at DDBJ/EMBL/GenBank under the accession LJJS00000000 and the sequences could be found at the site http://www.ncbi.nlm.nih.gov/assembly/GCA_001447215.1/. The MS-SSC91 genome sequences will provide information on the genetic basis of fungal infection of the soybean stem. It is valuable for studying soybean-fungal interactions and developing new control strategies for this pathogen.

Ecology of Diaporthe eres, the causal agent of hazelnut defects.

Project description:Diaporthe eres has been recently reported as the causal agent of hazelnut defects, with characteristic brown spots on the kernels surface and internal fruit discoloration. Knowledge regarding the ecology of this fungus is poor but, is critical to support a rationale and effective hazelnut crop protection strategy. Therefore, a study was performed to describe and model the effect of different abiotic factors such as temperature (T, 5-35°C, step 5°C) and water activity (aw 0.83-0.99, step 0.03) regimes on D. eres mycelial growth, pycnidial conidiomata development and asexual spore production during a 60-day incubation period. Alpha conidia germination was tested in the same T range and at different relative humidities (RH = 94, 97 and 100%) over 48 h incubation period. Fungal growth was observed from the first visual observation; regarding pycnidia and cirrhi, their development started after 8 and 19 days of incubation, respectively and increased over time. The optimum T for growth was 20-25°C and for pycnidia and cirrhi development was 30°C; aw ≥ 0.98 was optimal for the tested steps of the fungal cycle. The best condition for conidial germination of D. eres was at 25°C with RH = 100%. Quantitative data obtained were fitted using non- linear regression functions (Bete, logistic and polynomial), which provided a very good fit of the biological process (R2 = 0.793-0.987). These functions could be the basis for the development of a predictive model for the infection of D. eres of hazelnuts.

Draft genome sequence of Venturia carpophila, the causal agent of peach scab.

Project description:Venturia carpophila causes peach scab, a disease that renders peach (Prunus persica) fruit unmarketable. We report a high-quality draft genome sequence (36.9 Mb) of V. carpophila from an isolate collected from a peach tree in central Georgia in the United States. The genome annotation is described and a phylogenetic analysis of the pathogen is presented. The genome sequence will be a useful resource for various studies on the pathogen, including the biology and ecology, taxonomy and phylogeny, host interaction and coevolution, isolation and characterization of genes of interest, and development of molecular markers for genotyping and mapping.
