The Sequestration of Oxy-Polybrominated Diphenyl Ethers in the Nudibranchs Miamira magnifica and Miamira miamirana.

ABSTRACT: A series of oxy-polybrominated diphenyl ethers (O-PBDEs) has been isolated from the extracts of Miamira magnifica and Miamira miamirana collected from Queensland, Australia. M. magnifica sequesters the new OH-PBDE 1 and six known OH-PBDEs containing four to six bromines (2-7). M. miamirana also accumulates known tribromo- and tetrabromo OMe-PBDEs 8-10 in both mantle and viscera tissues. To date, Miamira is the only genus of the family Chromodorididae that is known to incorporate O-PBDEs, rather than terpenes, in the mantle where the metabolites may play a putative role in chemical defense. The extract of M. magnifica was tested in a brine shrimp lethality assay and exhibited an LD₅₀ of 58 ?g/mL.

Project description:Prenatal exposure to polybrominated diphenyl ethers (PBDEs) may disrupt thyroid function and contribute to adverse neurodevelopmental outcomes. We conducted a pilot study to explore the relationship between serum concentrations of lower-brominated PBDEs (BDE-17 to -154), higher-brominated PBDEs (BDE-183 to -209), and hydroxylated PBDE metabolites (OH-PBDEs) with measures of thyroid function in pregnant women. Concentrations of PBDEs, OH-PBDEs, thyroid-stimulating hormone (TSH), total thyroxine (T(4)), and free T(4) were measured in serum samples collected between 2008 and 2009 from 25 second trimester pregnant women in California. Median concentrations of lower-brominated PBDEs and OH-PBDEs were the highest reported to date in pregnant women. Median concentrations of BDE-47 and the sum of lower-brominated PBDEs (ΣPBDE(5)) were 43.1 ng/g lipid and 85.8 ng/g lipid, respectively, and the sum of OH-PBDEs (ΣOH-PBDE(4)) was 0.084 ng/mL. We observed a positive association between the weighted sum of chemicals known to bind to transthyretin (ΣTTR binders) and TSH levels. We also found positive associations between TSH and ΣPBDE(5), ΣOH-PBDE(4), BDE-47, BDE-85, 5-OH-BDE47, and 4'-OH-BDE49, and an inverse association with BDE-207. Relationships with free and total T(4) were weak and inconsistent. Our results indicate that PBDE exposures are elevated in pregnant women in California and suggest a relationship with thyroid function. Further investigation is warranted to characterize the risks of PBDE exposures during pregnancy.

Project description:Extensive use of polybrominated diphenyl ethers (PBDEs) has resulted in widespread environmental distribution and concerns remain about risks to human health and ecosystems from legacy PBDE contamination. Though bioremediation has been proven to be a cost-effective and efficient technology for treating halogenated pollutants in situ, application of bioremediation technologies at PBDE-contaminated sites is restricted by a lack of knowledge about functional PBDE-dehalogenating microbes. In the current study, we observed distinct PBDE-dehalogenation activity by three previously isolated Dehalococcoides mccartyi strains (CG1, CG4, and 11a5). PBDE debromination proceeded via distinct pathways in each D. mccartyi strain and involved previously characterized (TceA11a5, PcbA1, and PcbA4) and uncharacterized (11a5_e001) reductive dehalogenases (RDase), suggesting that this phenotype may be more widespread among the Dehalococcoides than is currently recognized.

Oxy-Polybrominated Diphenyl Ethers from the Indonesian Marine Sponge, Lamellodysidea herbacea: X-ray, SAR, and Computational Studies.

Project description:Polybrominated diphenyl ether (PBDE) compounds, derived from marine organisms, originate from symbiosis between marine sponges and cyanobacteria or bacteria. PBDEs have broad biological spectra; therefore, we analyzed structure and activity relationships of PBDEs to determine their potential as anticancer or antibacterial lead structures, through reactions and computational studies. Six known PBDEs (1-6) were isolated from the sponge, Lamellodysdiea herbacea; 13C NMR data for compound 6 are reported for the first time and their assignments are confirmed by their theoretical 13C NMR chemical shifts (RMSE < 4.0 ppm). Methylation and acetylation of 1 (2, 3, 4, 5-tetrabromo-6-(3', 5'-dibromo-2'-hydroxyphenoxy) phenol) at the phenol functional group gave seven molecules (7-13), of which 10, 12, and 13 were new. New crystal structures for 8 and 9 are also reported. Debromination carried out on 1 produced nine compounds (1, 2, 14, 16-18, 20, 23, and 26) of which 18 was new. Debromination product 16 showed a significant IC50 8.65 ± 1.11; 8.11 ± 1.43 µM against human embryonic kidney (HEK293T) cells. Compounds 1 and 16 exhibited antibacterial activity against Gram-positive Staphylococcus aureus and Gram-negative Klebsiella pneumoniae with MID 0.078 µg/disk. The number of four bromine atoms and two phenol functional groups are important for antibacterial activity (S. aureus and K. pneumoniae) and cytotoxicity (HEK293T). The result was supported by analysis of frontier molecular orbitals (FMOs). We also propose possible products of acetylation and debromination using analysis of FMOs and electrostatic charges and we confirm the experimental result.

Aerobic biotransformation of polybrominated diphenyl ethers (PBDEs) by bacterial isolates.

Project description:Polybrominated diphenyl ethers (PBDEs) are flame retardants that have been used in consumer products and furniture for three decades. Currently, very little is known about their fate in the environment and specifically about their susceptibility to aerobic biotransformation. Here, we investigated the ability of the polychlorinated biphenyl (PCB) degrading bacteria Rhodococcus jostii RHA1 and Burkholderia xenovorans LB400 to transform mono- through hexa-BDEs at ppb levels. We also tested the PBDE transforming abilities of the related strain Rhodococcus sp. RR1 and the ether-degrading Pseudonocardia dioxanivorans CB1190. The two PCB-degrading strains transformed all of the mono- through penta-BDEs and strain LB400 transformed one of the hexa-BDEs. The extent of transformation was inversely proportional to the degree of bromination. Strains RR1 and CB1190 were only able to transform the less brominated mono- and di-BDE congeners. RHA1 released stoichiometric quantities of bromide while transforming mono- and tetra-BDE congeners. LB400 instead converted most of a mono-BDE to a hydroxylated mono-BDE. This is the first report of aerobic transformation of tetra-, penta,- and hexa-BDEs as well as the first report of stoichiometric release of bromide during PBDE transformation.

A theoretical study on reductive debromination of polybrominated diphenyl ethers.

Project description:Recent progress has been made in the reductive debromination of polybrominated diphenyl ethers (PBDEs) by nanoscale zero-valent iron (nZVI). To better understand the mechanism of this reaction, seven selected BDE congeners and their anions were investigated at the density functional theory (DFT) level using four different methods, including B3LYP/6-31G(d), B3LYP/6-31+G(d), B3LYP/6-31G(d,p) and B3LYP/6-311G(d,p). The cleaved C-Br bonds observed in the equilibrium structures of anionic PBDEs were adopted as the probe of the susceptible debromination position of PBDEs in the presence of nZVI, and the proposed major reaction pathways based on our calculations can satisfactorily conform to the reported experimental results. The debromination preference is theoretically evaluated as meta-Br > ortho-Br > para-Br. In addition, both the calculated frontier orbital energies and adiabatic electronic affinities were found to be highly related to their experimental reductive debromination rate constants. The highest linear regression coefficient was observed in the case using the energy of lowest unoccupied molecular orbital as the molecular descriptor obtained from B3LYP/6-31G(d) (R(2) = 0.961, n = 7) or B3LYP/6-31G(d,p) (R(2) = 0.961, n = 7). The results clearly showed the evidence of an electron transfer mechanism associated with this reductive debromination reaction.
