First Report of Anthracnose Caused by Colletotrichum fioriniae on Chinese Matrimony Vine in Korea.

ABSTRACT: A fungus, Colletotrichum fioriniae, was isolated for the first time from fruits of Chinese matrimony vine (Lycium chinense Mill.) in Korea. It was classified as C. fioriniae based on the morphological characteristics and nucleotide sequence of glyceraldehyde-3-phosphate-dehydrogenase and ?-tubulin. To the best of our knowledge, this is the first report of C. fioriniae causing anthracnose of Chinese matrimony vine in Korea.

Project description:Anthracnose is one of the major problems for cultivating many crops, including vegetables, fruits, and trees. It is a continual threat for fruits grower worldwide. Colletotrichum fructicola was isolated from Shine Muscat berries showing typical anthracnose symptom in Korea. It was identified as C. fructicola based on morphology, pathological signs and concatenated sequences of internal transcribed spacer region of rDNA, glyceraldehyde-3-phosphate dehydrogenase, ?-tubulin-2, chitin synthase-1, calmodulin, and the Apn2-Mat1-2 intergenic spacer and partial mating type (Mat1-2) gene. Spot Anthracnose Disease Caused by Colletotrichum gloeosporioides on Tulip Tree in Korea.

Project description:The tulip tree (Liriodendron chinense) has been widely cultivated in Korea as a street or garden tree for its large flowers, which have a superficial resemblance to tulips. Occurrence of anthracnose disease on the leaves of tulip trees growing on the campus of Gyeongsang National University, Jinju, Korea, has been observed. Based on mycological characteristics, pathogenicity, and internal transcribed spacer sequence, the causal fungus was identified as Colletotrichum gloeosporioides. This is the first report on anthracnose disease caused by C. gloeosporioides on tulip trees in Korea.

First Report on Colletotrichum fructicola Causing Anthracnose in Chinese Sorghum and Its Management Using Phytochemicals.

Project description:Sorghum bicolor is cultivated worldwide. Leaf spots on sorghum, which lead to leaf lesions and impaired growth, are prevalent and severe in Guizhou Province, Southwest China. In August 2021, new leaf spot symptoms were observed on sorghum plants growing in agricultural fields. We used conventional tissue isolation methods and pathogenicity determination tests. Inoculations of sorghum with isolate 022ZW resulted in brown lesions similar to those observed under field conditions. The original inoculated isolates were reisolated and fulfilled Koch's postulates. Based on the morphological character and phylogenetic analyses of the combined sequences of the internal transcribed spacer (ITS) region and the β-tubulin (TUB2) and glyceraldehyde-3-phosphate dehydrogenase (GAPDH) genes, we identified the isolated fungus as C. fructicola. This paper is the first to report this fungus-causing disease in sorghum leaves. We studied the sensitivity of the pathogen to various phytochemicals. The sensitivity of C. fructicola to seven phytochemicals was measured using the mycelial growth rate method. Honokiol, magnolol, thymol, and carvacrol displayed good antifungal effects, with EC50 (concentration for 50% of the maximal effect) values of 21.70 ± 0.81, 24.19 ± 0.49, 31.97 ± 0.51, and 31.04 ± 0.891 µg/mL, respectively. We tested the control effect of the seven phytochemicals on the anthracnose caused by C. fructicola: honokiol and magnolol displayed good field efficacy. In this study, we expand the host range of C. fructicola, providing a basis for controlling sorghum leaf diseases caused by C. fructicola.

Characterization and Pathogenicity of New Record of Anthracnose on Various Chili Varieties Caused by Colletotrichum scovillei in Korea.

Project description:The anthracnose disease caused by Colletotrichum species is well-known as a major plant pathogen that primarily causes fruit rot in pepper and reduces its marketability. Thirty-five isolates representing species of Colletotrichum were obtained from chili fruits showing anthracnose disease symptoms in Chungcheongnam-do and Chungcheongbuk-do, South Korea. These 35 isolates were characterized according to morphological characteristics and nucleotide sequence data of internal transcribed spacer, glyceraldehyde-3-phosphate-dehydrogenase, and β-tubulin. The combined dataset shows that all of these 35 isolates were identified as C. scovillei and morphological characteristics were directly correlated with the nucleotide sequence data. Notably, these isolates were recorded for the first time as the causes of anthracnose caused by C. scovillei on pepper in Korea. Forty cultivars were used to investigate the pathogenicity and to identify the possible source of resistance. The result reveals that all of chili cultivars used in this study are susceptible to C. scovillei.

Soybean anthracnose caused by Colletotrichum species: Current status and future prospects.

Project description:Soybean (Glycine max) is one of the most important cultivated plants worldwide as a source of protein-rich foods and animal feeds. Anthracnose, caused by different lineages of the hemibiotrophic fungus Colletotrichum, is one of the main limiting factors to soybean production. Losses due to anthracnose have been neglected, but their impact may threaten up to 50% of the grain production.TaxonomyWhile C. truncatum is considered the main species associated with soybean anthracnose, recently other species have been reported as pathogenic on this host. Until now, it has not been clear whether the association of new Colletotrichum species with the disease is related to emerging species or whether it is due to the undergoing changes in the taxonomy of the genus.Disease symptomsTypical anthracnose symptoms are pre- and postemergence damping-off; dark, depressed, and irregular spots on cotyledons, stems, petioles, and pods; and necrotic laminar veins on leaves that can result in premature defoliation. Symptoms may evolve to pod rot, immature opening of pods, and premature germination of grains.ChallengesAs accurate species identification of the causal agent is decisive for disease control and prevention, in this work we review the taxonomic designation of Colletotrichum isolated from soybean to understand which lineages are pathogenic on this host. We also present a comprehensive literature review of soybean anthracnose, focusing on distribution, symptomatology, epidemiology, disease management, identification, and diagnosis. Colletotrichum Species Associated with Japanese Plum (Prunus salicina) Anthracnose in South Korea.

Project description:A total of 24 Colletotrichum isolates were isolated from diseased Japanese plum (Prunus salicina) fruits showing chlorotic regions with whitish-brown sunken necrotic lesions and phylogenetic relationships among the collected Colletotrichum isolates were determined. A subset of 11 isolates was chosen for further taxonomic study based on morphology and molecular characteristics identified using the internal transcribed spacer (ITS) and beta-tubulin (TUB2) genes. Isolates in the C. acutatum complex were analyzed using partial sequencing of five gene regions (ITS, GAPDH, ACT, TUB2, and CHS), and C. gloeosporioides sensu lato (s.l.) isolates were analyzed using seven gene regions (ITS, TUB2, GAPDH, ACT, CAL, CHS-1, and ApMat). Morphological assessments in combination with phylogenetic analysis delineated four species of Colletotrichum including C. gloeosporioides sensu stricto (s.s.), C. nymphaeae, C. foriniae, and C. siamense; these data identify Colletotrichum fioriniae and C. siamense two new species associated with plum anthracnose in South Korea. Finally, the pathogenicity of these four species in the development of plum anthracnose in South Korea was confirmed by inoculations of plum fruit.

Identification, Characterization, and Pathogenicity of Colletotrichum Species Causing Anthracnose of Peach in Korea.

Project description:Peach (Prunus persica L.) is one of the major fruit crops in South Korea, along with apple, persimmon, and Asian pears. Peach anthracnose is a continuing threat to growers and is accountable for enormous economic loss. In July 2018, anthracnose of peach appeared at different peach orchards in Gyeongsangbuk-do region, Korea. The typical anthracnose symptoms (brown, circular, and necrotic lesions) were observed on the fruits. Anthracnose of peach was surveyed in different peach orchards of Gyeongsangbuk-do, and 20 fungal isolates from 19 diseased fruits were collected. Multigene phylogenetic analyses coupled with morphological characteristic analysis approaches were used for identifying the fungal species isolated from diseased fruits. This study confirmed three Colletotrichum species. Based on the results, Colletotrichum siamense are reported for the first time as causal agents of peach anthracnose alongside C. fructicola and C. fioriniae, which has been reported previously. Pathogenicity assays were performed for the three isolates representing all the species identified, and Koch's postulates on detached healthy peach fruits were verified. All the identified species were pathogenic on peach fruits as the typical anthracnose symptoms were reproduced. Significant variations in the virulence were observed among fungal species on peach fruit.

Colletotrichum cymbidiicola Causing Anthracnose on Cymbidium Orchids in Korea.

Project description:A Colletotrichum species was isolated from leaves of Cymbidium exhibiting symptoms of anthracnose. In this study, the isolates obtained were identified based on recent taxonomic approaches for the genus Colletotrichum. The identity of the causal pathogen was confirmed using morphological data and phylogenetic analysis of combined multi-gene dataset (internal transcribed spacer, glyceraldehyde 3-phosphate dehydrogenase, chitin synthase-1, actin, histone3, beta-tubulin, and calmodulin). Pathogenicity testing revealed that the isolates were pathogenic to Cymbidium. Based on these results, the fungal pathogen occurring on Cymbidium orchids was identified as Colletotrichum cymbidiicola, which is a newly recorded species in Korea.

First report of feline intestinal trichomoniasis caused by Tritrichomonas foetus in Korea.

Project description:Feline intestinal tritrichomoniasis by Tritrichomonas foetus was first recognized in USA in 1999 and has so far been reported from UK, Norway, Switzerland, and Australia, but not from the Far East Asian countries. In November 2008, 2 female and male littermate Siamese cats, 6-month old, raised in a household in Korea were referred from a local veterinary clinic with a history of chronic persistent diarrhea. A direct smear examination of fecal specimens revealed numerous trichomonad trophozoites which were isolated by the fecal culture in InPouch TF-Feline medium. A PCR testing of the isolate based on the amplification of a conserved portion of the T. foetus internal transcribed spacer (ITS) regions (ITS1 and ITS2) and the 5.8S rRNA gene, and the molecular sequencing of the PCR amplicons confirmed infection with T. foetus. This is the first clinical case of feline intestinal trichomoniasis caused by T. foetus in Korea.

First Record of Colletotrichum anthrisci Causing Anthracnose on Avocado Fruits in Chile.

Project description:Anthracnose caused by Colletotrichum species is one of the most frequent and damaging fungal diseases affecting avocado fruits (Persea americana Mill.) worldwide. In Chile, the disease incidence has increased over the last decades due to the establishment of commercial groves in more humid areas. Since 2018, unusual symptoms of anthracnose have been observed on Hass avocado fruits, with lesions developing a white to gray sporulation. Morphological features and multi-locus phylogenetic analyses using six DNA barcodes (act, chs-1, gapdh, his3, ITS, and tub2) allowed the identification of the causal agent as Colletotrichum anthrisci, a member of the dematium species complex. Pathogenicity was confirmed by inoculating healthy Hass avocado fruits with representative isolates, reproducing the same symptoms initially observed, and successfully reisolating the same isolates from the margin of the necrotic pulp. Previously, several Colletotrichum species belonging to other species complexes have been associated with avocado anthracnose in other countries. To our knowledge, this is the first record of C. anthrisci and of a species of the dematium species complex causing anthracnose on avocado fruits in Chile and worldwide.
