Single-shot quantitative phase microscopy with color-multiplexed differential phase contrast (cDPC).

ABSTRACT: We present a new technique for quantitative phase and amplitude microscopy from a single color image with coded illumination. Our system consists of a commercial brightfield microscope with one hardware modification-an inexpensive 3D printed condenser insert. The method, color-multiplexed Differential Phase Contrast (cDPC), is a single-shot variant of Differential Phase Contrast (DPC), which recovers the phase of a sample from images with asymmetric illumination. We employ partially coherent illumination to achieve resolution corresponding to 2× the objective NA. Quantitative phase can then be used to synthesize DIC and phase contrast images or extract shape and density. We demonstrate amplitude and phase recovery at camera-limited frame rates (50 fps) for various in vitro cell samples and c. elegans in a micro-fluidic channel.

Project description:We present an imaging modality capable of providing high-speed optical dispersion measurements of live cells. The technique permits wide-field measurement of the optical phase shifts introduced by a sample for multiple discrete wavelengths in a single image capture. Utilizing spatial modulation and the wavelength dependence of the interference-fringe spacing, average refractive index as a function of wavelength is obtained, yielding optical dispersion measurements of the sample under observation. Because of its simple and low-cost design, the technique can be readily integrated into a standard microscope to collect additional diagnostic information about biological cells.

Single-shot two-frame π-shifted spatially multiplexed interference phase microscopy.

Project description:Single-shot, two-frame, π-shifted spatially multiplexed interference microscopy (π-SMIM) is presented as an improvement to previous SMIM implementations, introducing a versatile, robust, fast, and accurate method for cumbersome, noisy, and low-contrast phase object analysis. The proposed π-SMIM equips a commercially available nonholographic microscope with a high-speed (video frame rate) enhanced quantitative phase imaging (QPI) capability by properly placing a beam-splitter in the microscope embodiment to simultaneously (in a single shot) record two holograms mutually phase shifted by π radians at the expense of reducing the field of view. Upon subsequent subtractive superimposition of holograms, a π-hologram is generated with reduced background and improved modulation of interference fringes. These features determine superior phase retrieval quality, obtained by employing the Hilbert spiral transform on the π-hologram, as compared with a single low-quality (low signal-to-noise ratio) hologram analysis. In addition, π-SMIM enables accurate in-vivo analysis of high dynamic range phase objects, otherwise measurable only in static regime using time-consuming phase-shifting. The technique has been validated utilizing a 20 × / 0.46 NA objective in a regular Olympus BX-60 upright microscope for QPI of different lines of prostate cancer cells and flowing microbeads.

Single-shot wavelength-multiplexed phase microscopy under Gabor regime in a regular microscope embodiment.

Project description:Phase imaging microscopy under Gabor regime has been recently reported as an extremely simple, low cost and compact way to update a standard bright-field microscope with coherent sensing capabilities. By inserting coherent illumination in the microscope embodiment and producing a small defocus distance of the sample at the input plane, the digital sensor records an in-line Gabor hologram of the target sample, which is then numerically post-processed to finally achieve the sample's quantitative phase information. However, the retrieved phase distribution is affected by the two well-known drawbacks when dealing with Gabor's regime, that is, coherent noise and twin image disturbances. Here, we present a single-shot technique based on wavelength multiplexing for mitigating these two effects. A multi-illumination laser source (including 3 diode lasers) illuminates the sample and a color digital sensor (conventional RGB color camera) is used to record the wavelength-multiplexed Gabor hologram in a single exposure. The technique is completed by presenting a novel algorithm based on a modified Gerchberg-Saxton kernel to finally retrieve an enhanced quantitative phase image of the sample, enhanced in terms of coherent noise removal and twin image minimization. Experimental validations are performed in a regular Olympus BX-60 upright microscope using a 20X 0.46NA objective lens and considering static (resolution test targets) and dynamic (living spermatozoa) phase samples.

Color-coded LED microscopy for multi-contrast and quantitative phase-gradient imaging.

Project description:We present a multi-contrast microscope based on color-coded illumination and computation. A programmable three-color light-emitting diode (LED) array illuminates a specimen, in which each color corresponds to a different illumination angle. A single color image sensor records light transmitted through the specimen, and images at each color channel are then separated and utilized to obtain bright-field, dark-field, and differential phase contrast (DPC) images simultaneously. Quantitative phase imaging is also achieved based on DPC images acquired with two different LED illumination patterns. The multi-contrast and quantitative phase imaging capabilities of our method are demonstrated by presenting images of various transparent biological samples.

Single-Shot Optical Anisotropy Imaging with Quantitative Polarization Interference Microscopy.

Project description:Optical anisotropy measurement is essential for material characterization and biological imaging. In order to achieve single-shot mapping of the birefringence parameters of anisotropic samples, a novel polarized light imaging concept is proposed, namely quantitative polarization interference microscopy (QPIM). QPIM can be realized through designing a compact polarization-resolved interference microscopy system that captures interferograms bearing sample's linear birefringence information. To extract the retardance and the orientation angle maps from a single-shot measurement, a mathematical model for QPIM is further developed. The QPIM system is validated by measuring a calibrated quarter-wave plate, whose fast-axis orientation angle and retardance are determined with great accuracies. The single-shot nature of QPIM further allows to measure the transient dynamics of birefringence changes in material containing anisotropic structures. This application is demonstrated by capturing transient retardance changes in a custom-designed parallel-aligned nematic liquid crystal-based device.

Minimizing scattering-induced phase errors in differential interference contrast microscopy.

Project description:Differential interference contrast (DIC) microscopes allow noninvasive in vivo observation of transparent microstructures in tissue without the use of fluorescent dyes or genetic modification. We show how to modify a DIC microscope to measure the sample phase distribution accurately and in real-time even deep inside sample tissue. Our aim is to improve the DIC microscope's phase measurement to remove the phase bias that occurs in the presence of strong scattering. A quarter-wave plate was added in front of the polarization camera, allowing a modified phase calculation to incorporate all four polarization orientation angles (0 deg, 45 deg, 90 deg, and 135 deg) captured simultaneously by the polarization camera, followed by deconvolution. We confirm that the proposed method reduces phase measurement error in the presence of scattering and demonstrate the method using in vivo imaging of a beating heart inside a medaka egg and the whole-body blood circulation in a young medaka fish. Modifying a polarization-camera DIC microscope with a quarter-wave plate allows users to image deep inside samples without phase bias due to scattering effects.

Single-Shot Smartphone-Based Quantitative Phase Imaging Using a Distorted Grating.

Project description:Blood testing has been used as an essential tool to diagnose diseases for decades. Recently, there has been a rapid developing trend in using Quantitative Phase Imaging (QPI) methods for blood cell screening. Compared to traditional blood testing techniques, QPI has the advantage of avoiding dyeing or staining the specimen, which may cause damage to the cells. However, most existing systems are bulky and costly, requiring experienced personnel to operate. This work demonstrates the integration of one QPI method onto a smartphone platform and the application of imaging red blood cells. The adopted QPI method is based on solving the Intensity Transport Equation (ITE) from two de-focused pupil images taken in one shot by the smartphone camera. The device demonstrates a system resolution of about 1 ?m, and is ready to be used for 3D morphological study of red blood cells.

Smartphone-based multi-contrast microscope using color-multiplexed illumination.

Project description:We present a portable multi-contrast microscope capable of producing bright-field, dark-field, and differential phase contrast images of thin biological specimens on a smartphone platform. The microscopy method is based on an imaging scheme termed "color-coded light-emitting-diode (LED) microscopy (cLEDscope)," in which a specimen is illuminated with a color-coded LED array and light transmitted through the specimen is recorded by a color image sensor. Decomposition of the image into red, green, and blue colors and subsequent computation enable multi-contrast imaging in a single shot. In order to transform a smartphone into a multi-contrast imaging device, we developed an add-on module composed of a patterned color micro-LED array, specimen stage, and miniature objective. Simple installation of this module onto a smartphone enables multi-contrast imaging of transparent specimens. In addition, an Android-based app was implemented to acquire an image, perform the associated computation, and display the multi-contrast images in real time. Herein, the details of our smartphone module and experimental demonstrations with various biological specimens are presented.

Single-shot dual-wavelength interferometric microscopy.

Project description:Interferometric microscopy (IM) can provide complex field information of the biological samples with high spatial and temporal resolution with virtually no photodamage. Measuring wavelength-dependent information in particular has a wide range of applications from cell and tissue refractometry to the cellular biophysical measurements. IM measurements at multiple wavelengths are typically associated with a loss in temporal resolution, field of view, stability, sensitivity, and may involve using expensive equipment such as tunable filters or spatial light modulators. Here, we present a novel and simple design for an interferometric microscope that provides single-shot off-axis interferometric measurements at two wavelengths by encoding the two spectral images at two orthogonal spatial frequencies that allows clean separation of information in the Fourier space with no resolution loss. We demonstrated accurate simultaneous quantification of polystyrene bead refractive indices at two wavelengths.

Grating-based phase-contrast and dark-field computed tomography: a single-shot method.

Project description:Grating-based X-ray interferometry offers vast potential for imaging materials and tissues that are not easily visualised using conventional X-ray imaging. Tomographic reconstruction based on X-ray interferometric data provides not only access to the attenuation coefficient of an object, but also the refractive index and information about ultra-small-angle scattering. This improved functionality comes at the cost of longer measurement times because existing projection-based signal extraction algorithms require not only a single measurement per projection angle but several with precise grating movements in between. This obstacle hinders the adaptation of grating-based interferometry into a continuously rotating gantry. Several solutions to this problem have been proposed but all suffer from major drawbacks. We present results using an iterative reconstruction algorithm working directly on the interferograms. The suggested direct approach enables improved image quality, since interpolations and unnecessary assumptions about the object are circumvented. Our results demonstrate that it is possible to successfully reconstruct the linear attenuation coefficient, the refractive index and the linear diffusion coefficient, which is a measure related to ultra-small-angle scattering, using a single measurement per projection angle and without any grating movements. This is a milestone for future clinical implementation of grating-based phase-contrast and dark-field contrast X-ray computed tomography.
