SAVI: Synthetic apertures for long-range, subdiffraction-limited visible imaging using Fourier ptychography.

ABSTRACT: Synthetic aperture radar is a well-known technique for improving resolution in radio imaging. Extending these synthetic aperture techniques to the visible light domain is not straightforward because optical receivers cannot measure phase information. We propose to use macroscopic Fourier ptychography (FP) as a practical means of creating a synthetic aperture for visible imaging to achieve subdiffraction-limited resolution. We demonstrate the first working prototype for macroscopic FP in a reflection imaging geometry that is capable of imaging optically rough objects. In addition, a novel image space denoising regularization is introduced during phase retrieval to reduce the effects of speckle and improve perceptual quality of the recovered high-resolution image. Our approach is validated experimentally where the resolution of various diffuse objects is improved sixfold.

Project description:To a large extent, the performance of imaging systems is determined by their objectives, which affect properties as varied as collection efficiency, resolving power, and image distortions. Such limitations can be addressed by so-called aperture synthesis, a technique used, for instance, in radar, astronomy, and, increasingly, microscopy. Here, we apply such techniques to x-ray imaging and demonstrate how Fourier ptychography can be used at transmission x-ray microscopes to increase resolution, provide quantitative absorption and phase contrast, and allow for corrections of lens aberrations. We anticipate that such methods will find common and frequent applications, alleviating a number of limitations imposed by x-ray optical elements, offering an alternative approach to phase contrast imaging, and providing novel opportunities to mitigate radiation damage.

Diffraction tomography with Fourier ptychography.

Project description:This paper presents a technique to image the complex index of refraction of a sample across three dimensions. The only required hardware is a standard microscope and an array of LEDs. The method, termed Fourier ptychographic tomography (FPT), first captures a sequence of intensity-only images of a sample under angularly varying illumination. Then, using principles from ptychography and diffraction tomography, it computationally solves for the sample structure in three dimensions. The experimental microscope demonstrates a lateral spatial resolution of 0.39 ?m and an axial resolution of 3.7 ?m at the Nyquist-Shannon sampling limit (0.54 and 5.0 ?m at the Sparrow limit, respectively) across a total imaging depth of 110 ?m. Unlike competing methods, this technique quantitatively measures the volumetric refractive index of primarily transparent and contiguous sample features without the need for interferometry or any moving parts. Wide field-of-view reconstructions of thick biological specimens suggest potential applications in pathology and developmental biology.

Visible/near-infrared subdiffraction imaging reveals the stochastic nature of DNA walkers.

Project description:DNA walkers are designed with the structural specificity and functional diversity of oligonucleotides to actively convert chemical energy into mechanical translocation. Compared to natural protein motors, DNA walkers' small translocation distance (mostly <100 nm) and slow reaction rate (<0.1 nm s-1) make single-molecule characterization of their kinetics elusive. An important indication of single-walker kinetics is the rate-limiting reactions that a particular walker design bears. We introduce an integrated super-resolved fluorescence microscopy approach that is capable of long-term imaging to investigate the stochastic behavior of DNA walkers. Subdiffraction tracking and imaging in the visible and second near-infrared spectra resolve walker structure and reaction rates. The distributions of walker kinetics are analyzed using a stochastic model to reveal reaction randomness and the rate-limiting biochemical reaction steps.

In situ printing of liquid superlenses for subdiffraction-limited color imaging of nanobiostructures in nature.

Project description:The nanostructures and patterns that exist in nature have inspired researchers to develop revolutionary components for use in modern technologies and our daily lives. The nanoscale imaging of biological samples with sophisticated analytical tools, such as scanning electron microscopy (SEM) and transmission electron microscopy (TEM), has afforded a precise understanding of structures and has helped reveal the mechanisms contributing to the behaviors of the samples but has done so with the loss of photonic properties. Here, we present a new method for printing biocompatible "superlenses" directly on biological objects to observe subdiffraction-limited features under an optical microscope in color. We demonstrate the nanoscale imaging of butterfly wing scales with a super-resolution and larger field-of-view (FOV) than those of previous dielectric microsphere techniques. Our approach creates a fast and flexible path for the direct color observation of nanoscale biological features in the visible range and enables potential optical measurements at the subdiffraction-limited scale.

Dual-energy CT imaging with limited-angular-range data.

Project description:In dual-energy computed tomography (DECT), low- and high-kVp data are collected often over a full-angular range (FAR) of 360°. While there exists strong interest in DECT with low- and high-kVp data acquired over limited-angular ranges (LARs), there remains little investigation of image reconstruction in DECT with LAR data.Objective: we investigate image reconstruction with minimized LAR artifacts from low- and high-kVp data over LARs of ≤180° by using a directional-total-variation (DTV) algorithm.Methods: image reconstruction from LAR data is formulated as a convex optimization problem in which data-ℓ2is minimized with constraints on image's DTVs along orthogonal axes. We then achieve image reconstruction by applying the DTV algorithm to solve the optimization problem. We conduct numerical studies from data generated over arcs of LARs, ranging from 14° to 180°, and perform visual inspection and quantitative analysis of images reconstructed.Results: monochromatic images of interest obtained with the DTV algorithm from LAR data show substantially reduced artifacts that are observed often in images obtained with existing algorithms. The improved image quality also leads to accurate estimation of physical quantities of interest, such as effective atomic number and iodine contrast concentration.Conclusion: our study reveals that from LAR data of low- and high-kVp, monochromatic images can be obtained that are visually, and physical quantities can be estimated that are quantitatively, comparable to those obtained in FAR DECT.Significance: as LAR DECT is of high practical application interest, the results acquired in the work may engender insights into the design of DECT with LAR scanning configurations of practical application significance.

A plasmonic route for the integrated wireless communication of subdiffraction-limited signals.

Project description:Perfect lenses, superlenses and time-reversal mirrors can support and spatially separate evanescent waves, which is the basis for detecting subwavelength information in the far field. However, the inherent limitations of these methods have prevented the development of systems to dynamically distinguish subdiffraction-limited signals. Utilizing the physical merits of spoof surface plasmon polaritons (SPPs), we demonstrate that subdiffraction-limited signals can be transmitted on planar integrated SPP channels with low loss, low channel interference, and high gain and can be radiated with a very low environmental sensitivity. Furthermore, we show how deep subdiffraction-limited signals that are spatially coupled can be distinguished after line-of-sight wireless transmission. For a visualized demonstration, we realize the high-quality wireless communication of two movies on subwavelength channels over the line of sight in real time using our plasmonic scheme, showing significant advantages over the conventional methods.

Distinguishing Single-Metal Nanoparticles with Subdiffraction Spatial Resolution Using Variable-Polarization Fourier Transform Nonlinear Optical Microscopy.

Project description:The development and use of interferometric variable-polarization Fourier transform nonlinear optical (vpFT-NLO) imaging to distinguish colloidal nanoparticles colocated within the optical diffraction limit is described. Using a collinear train of phase-stabilized pulse pairs with orthogonal electric field vectors, the polarization of nonlinear excitation fields are controllably modulated between linear, circular, and various elliptical states. Polarization modulation is achieved by precise control over the time delay separating the orthogonal pulse pairs to within hundreds of attoseconds. The resultant emission from gold nanorods is imaged to a 2D array detector and correlated to the excitation field polarization and plasmon resonance frequency by Fourier transformation. Gold nanorods with length-to-diameter aspect ratios of 2 support a longitudinal surface plasmon resonance at approximately 800 nm, which is resonant with the excitation fundamental carrier wavelength. Differences in the intrinsic linear and circular dichroism resulting from variation in their relative alignment with respect to the laboratory frame enable optical differentiation of nanorods separated within 50 nm, which is an approximate 5-fold improvement over the diffraction limit of the microscope. The experimental results are supported by analytical simulations. In addition to subdiffraction spatial resolution, the vpFT-NLO method intrinsically provides the polarization- and frequency-dependent resonance response of the nanoparticles-providing spectroscopic information content along with super-resolution imaging capabilities.

Electrically Tunable Bifocal Metalens with Diffraction-Limited Focusing and Imaging at Visible Wavelengths.

Project description:Tunable optical devices powered by metasurfaces provide a new path for functional planar optics. In particular, lenses with tunable focal lengths can play a key role in various fields with applications in imaging, displays, and augmented and virtual reality devices. Here, the authors demonstrate an electrically controllable bifocal metalens at visible wavelengths by incorporating a metasurface designed to focus light at two different focal lengths, with liquid crystals to actively manipulate the focal length of the metalens through the application of an external bias. By utilizing hydrogenated amorphous silicon that is optimized to provide an extremely low extinction coefficient in the visible regime, the metalens is highly efficient with measured focusing efficiencies of around 44%. They numerically design and experimentally realize and characterize tunable focusing and demonstrate electrically tunable active imaging at visible wavelengths using the bifocal metalens combined with liquid crystals. Diffraction limited focusing and imaging is verified through the analysis of the measured optical intensities at the focal points and the modulation transfer function. The bifocal metalens is used to demonstrate electrically modulated focus switching between the two designed focal planes, to display images of positive and negative target objects.

Automated wide-field malaria parasite infection detection using Fourier ptychography on stain-free thin-smears.

Project description:Diagnosis of malaria in endemic areas is hampered by the lack of a rapid, stain-free and sensitive method to directly identify parasites in peripheral blood. Herein, we report the use of Fourier ptychography to generate wide-field high-resolution quantitative phase images of erythrocytes infected with malaria parasites, from a whole blood sample. We are able to image thousands of erythrocytes (red blood cells) in a single field of view and make a determination of infection status of the quantitative phase image of each segmented cell based on machine learning (random forest) and deep learning (VGG16) models. Our random forest model makes use of morphology and texture based features of the quantitative phase images. In order to label the quantitative images of the cells as either infected or uninfected before training the models, we make use of a Plasmodium berghei strain expressing GFP (green fluorescent protein) in all life cycle stages. By overlaying the fluorescence image with the quantitative phase image we could identify the infected subpopulation of erythrocytes for labelling purposes. Our machine learning model (random forest) achieved 91% specificity and 72% sensitivity while our deep learning model (VGG16) achieved 98% specificity and 57% sensitivity. These results highlight the potential for quantitative phase imaging coupled with artificial intelligence to develop an easy to use platform for the rapid and sensitive diagnosis of malaria.

Subdiffraction-resolution live-cell imaging for visualizing thylakoid membranes.

Project description:The chloroplast is the chlorophyll-containing organelle that produces energy through photosynthesis. Within the chloroplast is an intricate network of thylakoid membranes containing photosynthetic membrane proteins that mediate electron transport and generate chemical energy. Historically, electron microscopy (EM) has been a powerful tool for visualizing the macromolecular structure and organization of thylakoid membranes. However, an understanding of thylakoid membrane dynamics remains elusive because EM requires fixation and sectioning. To improve our knowledge of thylakoid membrane dynamics we need to consider at least two issues: (i) the live-cell imaging conditions needed to visualize active processes in vivo; and (ii) the spatial resolution required to differentiate the characteristics of thylakoid membranes. Here, we utilize three-dimensional structured illumination microscopy (3D-SIM) to explore the optimal imaging conditions for investigating the dynamics of thylakoid membranes in living plant and algal cells. We show that 3D-SIM is capable of examining broad characteristics of thylakoid structures in chloroplasts of the vascular plant Arabidopsis thaliana and distinguishing the structural differences between wild-type and mutant strains. Using 3D-SIM, we also visualize thylakoid organization in whole cells of the green alga Chlamydomonas reinhardtii. These data reveal that high light intensity changes thylakoid membrane structure in C. reinhardtii. Moreover, we observed the green alga Chromochloris zofingiensis and the moss Physcomitrella patens to show the applicability of 3D-SIM. This study demonstrates that 3D-SIM is a promising approach for studying the dynamics of thylakoid membranes in photoautotrophic organisms during photoacclimation processes.
