Root xylem plasticity to improve water use and yield in water-stressed soybean.

ABSTRACT: We tested the hypothesis that increasing the number of metaxylem vessels would enhance the efficiency of water uptake in soybean (Glycine max) and decrease the yield gap in water-limited environments. A panel of 41 soybean accessions was evaluated in greenhouse, rainout shelter, and rain-fed field environments. The metaxylem number influenced the internal capture of CO2 and improved stomatal conductance, enhancing water uptake/use in soybeans exposed to stress during the reproductive stage. We determined that other root anatomical features, such as cortex cell area and the percentage of stele that comprised cortical cells, also affected seed yield under similar growth parameters. Seed yield was also impacted by pod retention rates under drought stress (24-80 pods/plant). We surmise that effective biomass allocation, that is, the transport of available photosynthates to floral structures at late reproductive growth stages (R6-R7), enables yield protection under drought stress. A mesocosm study of contrasting lines for yield under drought stress and root anatomical features revealed that increases in metaxylem number as an adaptation to drought in the high-yielding lines improved root hydraulic conductivity, which reduced the metabolic cost of exploring water in deeper soil strata and enhanced water transport. This allowed the maintenance of shoot physiological processes under water-limited conditions.

Project description:Drought stress has been identified as the major environmental factor limiting soybean [Glycine max (L.) Merr.] yield worldwide. Current breeding efforts in soybean largely focus on identifying genotypes with high seed yield and drought tolerance. Water use efficiency (WUE) that results in greater yield per unit rainfall is an important parameter in determining crop yields in many production systems, and is often related with crop drought tolerance. Even though roots are major plant organs that perceive and respond to drought stress, their utility in improving soybean yield and WUE under different environmental and management conditions are largely unclear. The objectives of this research was to evaluate soybean cultivars and breeding and germplasm lines for yield, WUE, root penetrability of hardpan, and root morphology. Field experiments were conducted at two locations in South Carolina (southeastern United States) during the 2017 cropping season to test the genotypes for yield and root morphology under irrigated and non-irrigated conditions. Two independent controlled-environmental experiments were conducted to test the genotypes for WUE and root penetrability of synthetic hardpans. The slow wilting lines NTCPR94-5157 and N09-13890 had equal or greater yield than the checks- cultivar NC-Raleigh and the elite South Carolina breeding line SC07-1518RR, under irrigated and non-irrigated conditions. The high yielding genotypes NTCPR94-5157, N09-13890, and SC07-1518RR exhibited root parsimony (reduced root development). This supported the recent hypothesis in literature that root parsimony would have adaptational advantage to improve yield under high input field conditions. The high yielding genotypes NTCPR94-5157, N09-13890, NC-Raleigh, and SC07-1518RR and a cultivar Boggs (intermediate in yield) possessed high WUE and had increased root penetrability of hardpans. These genotypes offer useful genetic materials for soybean breeding programs for improving yield, drought tolerance, and/or hardpan penetrability.

Project description:Enhancing the acquisition of belowground resources has been identified as an opportunity for improving soybean productivity worldwide. Root system architecture is gaining interest as a selection criterion in breeding programs for enhancing soil resource acquisition and developing climate-resilient varieties. Here we are presenting two novel characteristics of soybean root system architecture that improve aboveground growth and yield. Eleven selected soybean genotypes were tested under rain-fed conditions in 2019 and 2020 at two locations in South Carolina, in which one of the locations was characterized by compacted soils. The elite SC breeding line SC07-1518RR, exotic pedigree line N09-12854, and slow wilting line N09-13890 were superior genotypes in terms of biomass production, seed yield, and/or water use efficiency. Genotypes N09-12854 and N09-13890 demonstrated reduced root development (based on total root count and length), likely to restrict belowground growth and allocate more resources for shoot growth. This characteristic, which can be referred as a parsimonious root phenotype, might be advantageous for soybean improvement in high-input production systems (characterized by adequate fertilizer application and soil fertility) that exist in many parts of the world. Genotype SC07-1518RR exhibited a similar strategy: while it maintained its root system at an intermediate size through reduced levels of total root count and length, it selectively distributed more roots at deeper depths (53-70 cm). The increased root distribution of SC07-1518RR at deeper depths in compacted soil indicates its root penetrability and suitability for clayey soils with high penetration resistance. The beneficial root phenotypes identified in this study (parsimonious root development and selective root distribution in deeper depths) and the genotypes that possessed those phenotypes (SC07-1518RR, N09-12854, and N09-13890) will be useful for breeding programs in developing varieties for optimal, drought, and compacted-soil conditions.

Project description:Water shortage and low phosphorus (P) availability limit yields in soybean. Roots play important roles in water-limited and P-deficient environment, but the underlying mechanisms are largely unknown. In this study we determined the responses of four soybean [Glycine max (L.) Merr.] genotypes [Huandsedadou (HD), Bailudou (BLD), Jindou 21 (J21), and Zhonghuang 30 (ZH)] to three P levels [applied 0 (P0), 60 (P60), and 120 (P120) mg P kg-1 dry soil to the upper 0.4 m of the soil profile] and two water treatment [well-watered (WW) and water-stressed (WS)] with special reference to root morphology and architecture, we compared yield and its components, root morphology and root architecture to find out which variety and/or what kind of root architecture had high grain yield under P and drought stress. The results showed that water stress and low P, respectively, significantly reduced grain yield by 60 and 40%, daily water use by 66 and 31%, P accumulation by 40 and 80%, and N accumulation by 39 and 65%. The cultivar ZH with the lowest daily water use had the highest grain yield at P60 and P120 under drought. Increased root length was positively associated with N and P accumulation in both the WW and WS treatments, but not with grain yield under water and P deficits. However, in the WS treatment, high adventitious and lateral root densities were associated with high N and P uptake per unit root length which in turn was significantly and positively associated with grain yield. Our results suggest that (1) genetic variation of grain yield, daily water use, P and N accumulation, and root morphology and architecture were observed among the soybean cultivars and ZH had the best yield performance under P and water limited conditions; (2) water has a major influence on nutrient uptake and grain yield, while additional P supply can modestly increase yields under drought in some soybean genotypes; (3) while conserved water use plays an important role in grain yield under drought, root traits also contribute to high nutrient uptake efficiency and benefit yield under drought.

Project description:Typical small-pot culture systems are not ideal for controlled environment phenotyping for drought tolerance, especially for root-related traits. We grew soybean plants in a greenhouse in 1-m rooting columns filled with amended field soil to test the effects of drought stress on water use, root growth, shoot growth, and yield components. There were three watering treatments, beginning at first flower: watered daily to 100% of the maximum soil water holding capacity (control), 75% (mild drought stress), or 50% (drought stress). We also tested whether applying fertilizer throughout the 1-m soil depth instead of only in the top 30 cm would modify root distribution by depth in the soil profile and thereby affect responses to drought stress. Distributing the fertilizer over the entire 1-m soil depth altered the root biomass distribution and volumetric soil water content profile at first flower, but these effects did not persist to maturity and thus did not enhance drought tolerance. Compared to the control (100%) watering treatment, the 50% watering treatment significantly reduced seed yield by 40%, pod number by 42%, seeds per pod by 3%, shoot dry matter by 48%, root dry matter by 53%, and water use by 52%. Effects of the 75% watering treatment were intermittent between the 50 and 100%. The 50% treatment significantly increased root-to-shoot dry matter ratio by 23%, harvest index by 17%, and water-use efficiency by 7%. Seed size was not affected by either fertilizer or watering treatments. More than 65% of the total root dry matter was distributed in the upper 20 cm of the profile in all watering treatments. However, the two drought stress treatments, especially the mild drought stress, had a greater proportion of root dry matter located in the deeper soil layers. The overall coefficient of variation for seed yield was low at 5.3%, suggesting good repeatability of the treatments. Drought stress imposed in this culture system affected yield components similarly to what is observed in the field, with pod number being the component most strongly affected. This system should be useful for identifying variation among soybean lines for a wide variety of traits related to drought tolerance.

Project description:IntroductionArtemisinin is a secondary metabolite well-known for its use in the treatment of malaria. It also displays other antimicrobial activities which further increase its interest. At present, Artemisia annua is the sole commercial source of the substance, and its production is limited, leading to a global deficit in supply. Furthermore, the cultivation of A. annua is being threatened by climate change. Specifically, drought stress is a major concern for plant development and productivity, but, on the other hand, moderate stress levels can elicit the production of secondary metabolites, with a putative synergistic interaction with elicitors such as chitosan oligosaccharides (COS). Therefore, the development of strategies to increase yield has prompted much interest. With this aim, the effects on artemisinin production under drought stress and treatment with COS, as well as physiological changes in A. annua plants are presented in this study.MethodsPlants were separated into two groups, well-watered (WW) and drought-stressed (DS) plants, and in each group, four concentrations of COS were applied (0, 50,100 and 200 mg•L-1). Afterwards, water stress was imposed by withholding irrigation for 9 days.ResultsTherefore, when A. annua was well watered, COS did not improve plant growth, and the upregulation of antioxidant enzymes hindered the production of artemisinin. On the other hand, during drought stress, COS treatment did not alleviate the decline in growth at any concentration tested. However, higher doses improved the water status since leaf water potential (YL) improved by 50.64% and relative water content (RWC) by 33.84% compared to DS plants without COS treatment. Moreover, the combination of COS and drought stress caused damage to the plant's antioxidant enzyme defence, particularly APX and GR, and reduced the amount of phenols and flavonoids. This resulted in increased ROS production and enhanced artemisinin content by 34.40% in DS plants treated with 200 mg•L-1 COS, compared to control plants.ConclusionThese findings underscore the critical role of ROS in artemisinin biosynthesis and suggest that COS treatment may boost artemisinin yield in crop production, even under drought conditions.
