First report of lily mottle virus on lily (Lilium sp.) in southern India.

ABSTRACT: Lily samples collected from commercial cut flower growing field at Nilgiris, Tamil Nadu, India were serologically tested through direct antigen coating-ELISA and dot immuno binding assay for the infection of potyvirus through potyvirus group specific polyclonal antibody. The RT-PCR using potyvirus universal degenerate primers (PNIbF1/PCPR1) showed the presence of a potyvirus and the sequencing results showed the association of lily mottle virus (LMoV). Further LMoV was confirmed by RT-PCR amplification using newly designed primer pair covering entire coat protein gene and sequence results showed 100% nucleotide identity with the LMoV reported from South Korean. To our knowledge, this is the first confirmed evidence for the occurrence of LMoV on lily in southern India.

Rapid Detection of Lily mottle virus and Arabis mosaic virus Infecting Lily (Lilium spp.) Using Reverse Transcription Loop-Mediated Isothermal Amplification.

Project description:The Lily mottle virus (LMoV) impedes the growth and quality of lily crops in Lanzhou, China. Recently Arabis mosaic virus (ArMV) has been detected in LMoV-infected plants in this region, causing plant stunting as well as severe foliar symptoms, and likely posing a threat to lily production. Consequently, there is a need to develop simple, sensitive, and reliable detection methods for these two viruses to prevent them from spreading. Reverse transcription (RT) loop-mediated isothermal amplification (LAMP) assays have been developed to detect LMoV and ArMV using two primer pairs that match six conserved sequences of LMoV and ArMV coat proteins, respectively. RT-LAMP assay results were visually assessed in reaction tubes using green fluorescence and gel electrophoresis. Our assays successfully detected both LMoV and ArMV in lily plants without the occurrence of viral cross-reactivity from other lily viruses. Optimal conditions for LAMP reactions were 65°C and 60°C for 60 min for LMoV and ArMV, respectively. Detection sensitivity for both RT-LAMP assays was a hundredfold greater than that of our comparative RT-polymerase chain reaction assays. We have also found this relatively rapid, target specific and sensitive method can also be used for samples collected in the field and may be especially useful in regions with limited or no laboratory facilities.

First report of Chilli veinal mottle virus in Naga chilli (Capsicum chinense) in Meghalaya, India.

Project description:The present study confirms the occurrence of Chilli veinal mottle virus (ChiVMV) under the genus Potyvirus in Naga chilli (Capsicum chinense) in Meghalaya based on mechanical transmission assay, transmission electron microscopy, RT-PCR and sequence analysis. This is the first record of Chivmv in Naga chilli in North-East India.

First report of Amaranthus sp. as a natural host of capsicum chlorosis virus in India.

Project description:Capsicum chlorosis virus (CaCV) is emerging as economically important tospovirus, being a recently identified virus its complete host range was still not completely known. Virus infected leaves of amaranth plants grown in the vicinity of Capsicum fields showing characteristic symptoms of tospoviruses were collected, confirmed by tospovirus group specific antibody for the presence of a tospovirus and then the presence of CaCV was confirmed by virus specific antibodies. Nucleocapsid protein (NP) specific primer were used for amplification of NP gene and amplified cDNA products were confirmed by sequencing that the virus infecting amaranth was an isolate of CaCV. A comparison of the NP sequence with other isolate confirmed that it was 100 % similar to the reported Indian isolate of CaCV. To best of our knowledge this is the first report of natural infection of CaCv in Amaranthus sp. in India.

The complete mitogenome sequence of the coral lily (Lilium pumilum) and the Lanzhou lily (Lilium davidii) in China.

Project description:BackgroundThe mitogenomes of higher plants are conserved. This study was performed to complete the mitogenome of two China Lilium species (Lilium pumilum Redouté and Lilium davidii var. unicolor (Hoog) cotton).MethodsGenomic DNA was separately extracted from the leaves of L. pumilum and L. davidii in triplicate and used for sequencing. The mitogenome of Allium cepa was used as a reference. Genome assembly, annotation and phylogenetic tree were analyzed.ResultsThe mitogenome of L. pumilum and L. davidii was 988,986 bp and 924,401 bp in length, respectively. There were 22 core protein-coding genes (including atp1, atp4, atp6, atp9, ccmB, ccmC, ccmFc, ccmFN1, ccmFN2, cob, cox3, matR, mttB, nad1, nad2, nad3, nad4, nad4L, nad5, nad6, nad7 and nad9), one open reading frame and one ribosomal protein-coding gene (rps12) in the mitogenomes. Compared with the A. cepa mitogenome, the coding sequence of the 24 genes and intergenic spacers in L. pumilum and L. davidii mitogenome contained 1,621 and 1,617 variable sites, respectively. In the phylogenetic tree, L. pumilum and L. davidii were distinct from A. cepa (NC_030100).ConclusionsL. pumilum and L. davidii mitogenomes have far distances from other plants. This study provided additional information on the species resources of China Lilium.

Occurrence of pepper mottle virus on tomato in India.

Project description:Tomato plants exhibiting mosaic and mottling symptoms were collected from the research field of Punjab Agricultural University during 2016-17. DAS-ELISA test using pepper mottle virus (PepMoV) antiserum showed positive reaction to 5 of 25 tomato samples collected. Further molecular characterization of virus was carried out by using potyvirus specific primer pairs corresponding to most conserved and variable genes viz., Nuclear inclusion b (NIb) and P1 respectively. RT-PCR assay confirmed the association of PepMoV with tomato crop under field conditions in India.

First Report of Tobacco streak virus Infecting Kenaf (Hibiscus cannabinus) in India.

Project description:The natural occurrence of Tobacco streak virus (TSV) in Hibiscus cannabinus was detected by enzyme-linked immunosorbent assay using an antiserum raised against TSV and reverse transcription polymerase chain reaction (PCR) using primers specific for the coat protein gene of the virus. Sequence analysis of the PCR products showed 99.6 and 99.5% of maximum identity at nucleotide and amino acid levels, respectively with TSV onion isolate from Kurnool (HM131490).This is the first report of the natural occurrence of TSV on kenaf in India.

First Report of Zucchini yellow mosaic virus Infecting Gherkin (Cucumis anguira) in India.

Project description:A field visit in September 2011 to the Cucumis anguira (Gherkin) growing regions of Kuppam, Chittoor district of Andhra Pradesh, India revealed occurrence of mosaic, blistering and fruit malformation leading to the crop losses. Analysis of field samples revealed association of Zucchini yellow mosaic virus (ZYMV) with the disease. This is the first confirmed report of natural occurrence of ZYMV on Gherkin in India.

First report of cucumber mosaic virus infecting antamul vine (Tylophora indica) in India.

Project description:Tylophora indica (Burm f.) Merrill; commonly known as antamul, is an important medicinal herb. Typical yellow rings or irregular yellow spot and in severe condition necrotic rings were observed on the leaves of the crop. The examination of symptomatic leaf samples under transmission electron microscopy revealed the presence of spherical virions confirmed the association of cucumo like virus group. A novel degenerate primers pair was designed by multiple sequence alignment of RdRP region and used in RT-PCR to amplify a ~?410 bp genomic fragment. The sequence of the amplified fragment shared 97-98% sequence identity with cucumber mosaic virus (CMV). This study is the first report of the association of CMV with the yellow ring symptom of antamul in India.

Desmodium mottle virus, the first legumovirus (genus Begomovirus) from East Africa.

Project description:A novel bipartite legumovirus (genus Begomovirus, family Geminiviridae), that naturally infects the wild leguminous plant Desmodium sp. in Uganda, was molecularly characterized and named Desmodium mottle virus. The highest nucleotide identities for DNA-A, obtained from two field-collected samples, were 79.9% and 80.1% with the legumovirus, soybean mild mottle virus. DNA-B had the highest nucleotide identities (65.4% and 66.4%) with a typical non-legumovirus Old World begomovirus, African cassava mosaic virus. This is the first report of a legumovirus in East Africa and extends the known diversity of begomoviruses found infecting wild plants in this continent.
