Reducing the critical particle diameter in (highly) asymmetric sieve-based lateral displacement devices.

ABSTRACT: Deterministic lateral displacement technology was originally developed in the realm of microfluidics, but has potential for larger scale separation as well. In our previous studies, we proposed a sieve-based lateral displacement device inspired on the principle of deterministic lateral displacement. The advantages of this new device is that it gives a lower pressure drop, lower risk of particle accumulation, higher throughput and is simpler to manufacture. However, until now this device has only been investigated for its separation of large particles of around 785 µm diameter. To separate smaller particles, we investigate several design parameters for their influence on the critical particle diameter. In a dimensionless evaluation, device designs with different geometry and dimensions were compared. It was found that sieve-based lateral displacement devices are able to displace particles due to the crucial role of the flow profile, despite of their unusual and asymmetric design. These results demonstrate the possibility to actively steer the velocity profile in order to reduce the critical diameter in deterministic lateral displacement devices, which makes this separation principle more accessible for large-scale, high throughput applications.

Project description:The lateral displacement of cells orthogonal to a flow stream by rolling on asymmetrical receptor patterns presents a new opportunity for the label-free separation and analysis of cells. Understanding the nature of cell rolling trajectories on such substrates is necessary to the engineering of substrates and the design of devices for cell separation and analysis. Here, we investigate the statistical nature of cell rolling and the effect of pattern geometry and flow shear stress on cell rolling trajectories using micrometer-scale patterns of biomolecular receptors with well-defined edges. Leukemic myeloid HL60 cells expressing the PSGL-1 ligand were allowed to flow across a field of patterned lines fabricated using microcontact printing and functionalized with the P-selectin receptor, leveraging both the specific adhesion of this ligand-receptor pair and the asymmetry of the receptor pattern inclination angle with respect to the fluid shear flow direction (? = 5, 10, 15, and 20°). The effects of the fluid shear stress magnitude (? = 0.5, 1, 1.5, and 2.0 dyn/cm(2)), ?, and P-selectin incubation concentration were quantified in terms of the rolling velocity and edge tracking length. Rolling cells tracked along the inclined edges of the patterned lines before detaching and reattaching on another line. The detachment of rolling cells after tracking along the edge was consistent with a Poisson process of history-independent interactions. Increasing the edge inclination angle decreased the edge tracking length in an exponential manner, contrary to the shear stress magnitude and P-selectin incubation concentration, which did not have a significant effect. On the basis of these experimental data, we constructed an empirical model that predicted the occurrence of the maximum lateral displacement at an edge angle of 7.5°. We also used these findings to construct a Monte Carlo simulation for the prediction of rolling trajectories of HL60 cells on P-selectin-patterned substrates with a specified edge inclination angle. The prediction of lateral displacement in the range of 200 ?m within a 1 cm separation length supports the feasibility of label-free cell separation via asymmetric receptor patterns in microfluidic devices.

Project description:The advancement of imaging systems has significantly ameliorated various technologies, including Intelligence Surveillance Reconnaissance Systems and Guidance Systems, by enhancing target detection, recognition, identification, positioning, and tracking capabilities. These systems can be countered by deploying obscurants like smoke, dust, or fog to hinder visibility and communication. However, these counter-systems affect the visibility of both sides of the cloud. In this sense, this manuscript introduces a new concept of a smoke cloud composed of engineered Janus particles to conceal the target image on one side while providing clear vision from the other. The proposed method exploits the unique scattering properties of Janus particles, which selectively interact with photons from different directions to open up the possibility of asymmetric imaging. This approach employs a model that combines a genetic algorithm with Discrete Dipole Approximation to optimize the Janus particles' geometrical parameters for the desired scattering properties. Moreover, we propose a Monte Carlo-based approach to calculate the image formed as photons pass through the cloud, considering highly asymmetric particles, such as Janus particles. The effectiveness of the cloud in disguising a target is evaluated by calculating the Probability of Detection (PD) and the Probability of Identification (PID) based on the constructed image. The optimized Janus particles can produce a cloud where it is possible to identify a target more than 50% of the time from one side (PID > 50%) while the target is not detected more than 50% of the time from the other side (PD < 50%). The results demonstrate that the Janus particle-engineered smoke enables asymmetric imaging with simultaneous concealment from one side and clear visualization from the other. This research opens intriguing possibilities for modern obscurant design and imaging systems through highly asymmetric and inhomogeneous particles besides target detection and identification capabilities in challenging environments.
