Wuacanthus (Acanthaceae), a new Chinese endemic genus segregated from Justicia (Acanthaceae).

ABSTRACT: A new genus, Wuacanthus Y.F. Deng, N.H. Xia & H. Peng (Acanthaceae), is described from the Hengduan Mountains, China. Wuacanthus is based on Wuacanthus microdontus (W.W.Sm.) Y.F. Deng, N.H. Xia & H. Peng, originally published in Justicia and then moved to Mananthes. The new genus is characterized by its shrub habit, strongly 2-lipped corolla, the 2-lobed upper lip, 3-lobed lower lip, 2 stamens, bithecous anthers, parallel thecae with two spurs at the base, 2 ovules in each locule, and the 4-seeded capsule. Phylogenetic analyses show that the new genus belongs to the Pseuderanthemum lineage in tribe Justicieae. Wuacanthus is closely related to Pseuderanthemum but differs from the latter by its shorter corolla tube and two minute spurs at the base of each anther-theca. W. microdontus is assessed with the status EN B2ab (iii) based on the IUCN Red List Categories and Criteria.

Characterization and comparative analysis of the plastome sequence from Justicia ventricosa (Lamiales: Acanthaceae).

Project description:Justicia ventricosa is a characteristic ethnic herb commonly used to treat Orthopedic pains. Here, to confirm its phylogenetic position and to develop molecular markers that can distinguish different Justicia species, we obtained and analyzed the plastome of Justicia ventricosa. The plastome was sequenced using the Illumina HiSeq sequencing platform, assembled with NOVOPlasty, and annotated with CPGAVAS2. The genome has a circular structure of 149,700 bp, containing a large single-copy region of 82,324 bp, a small single-copy region of 17,260 bp, and two reverse repeat regions of 25,058 bp each. It encodes 112 unique genes, including 76 protein-coding genes, eight ribosomal RNA genes, and 28 transfer RNA genes. Twenty cis-splicing genes were found. In total, we predicted 19 microsatellite repeats and 13 tandem repeat sequences. For distributed repeats, four were palindrome repeats and five were direct repeats. To find the highly variable intergenic spacer (IGS) regions, we calculated the K2P distances for IGS regions from four Justicia species. The K2P values ranged from 6.11 to 57.82. The largest K2P distances were found for ccsA-ndhD, petB-petD, psbK-psbI, and ycf4-cemA. Phylogenetic analysis results showed that J. ventricosa was most closely related to J. leptostachya. To determine how Justicia species adapt to the environment, we performed selection pressure analysis. Nine genes were found to have undergone positive selection. Lastly, we performed a genome-wise DNA barcode prediction, seven pairs of primers were found. The results provide valuable information that can be used for molecular marker development and bioprospecting in Justicia species.

Xochiquetzallia (Asparagaceae, Brodiaeoideae), a new genus segregated from the paraphyletic Dandya.

Project description:A new genus, Xochiquetzallia, for the Brodiaeoideae, Asparagaceae family is here proposed. A taxonomic analysis based on morphology highlights its synapomorphies. The characters that distinguish Xochiquetzallia are the absence of a pith in the gynophore and the presence of an entire stigma. The recognition of Dandya purpusii as a monotypic genus is supported by the development of a short floral tube (< 2 mm) and a pith in the gynophore, as well as a divided stigma shared with the other genera of the Milla clade, Bessera, Jaimehintonia, Petronymphe and Milla. A key to its taxonomic determination is given for both the Xochiquetzallia species and the Milla clade genera.

Complete Chloroplast Genome Sequence of Justicia flava: Genome Comparative Analysis and Phylogenetic Relationships among Acanthaceae.

Project description:The complete chloroplast genome of J. flava, an endangered medicinal plant in Saudi Arabia, was sequenced and compared with cp genome of three Acanthaceae species to characterize the cp genome, identify SSRs, and also detect variation among the cp genomes of the sampled Acanthaceae. NOVOPlasty was used to assemble the complete chloroplast genome from the whole genome data. The cp genome of J. flava was 150, 888bp in length with GC content of 38.2%, and has a quadripartite structure; the genome harbors one pair of inverted repeat (IRa and IRb 25, 500bp each) separated by large single copy (LSC, 82, 995 bp) and small single copy (SSC, 16, 893 bp). There are 132 genes in the genome, which includes 80 protein coding genes, 30 tRNA, and 4 rRNA; 113 are unique while the remaining 19 are duplicated in IR regions. The repeat analysis indicates that the genome contained all types of repeats with palindromic occurring more frequently; the analysis also identified total number of 98 simple sequence repeats (SSR) of which majority are mononucleotides A/T and are found in the intergenic spacer. The comparative analysis with other cp genomes sampled indicated that the inverted repeat regions are conserved than the single copy regions and the noncoding regions show high rate of variation than the coding region. All the genomes have ndhF and ycf1 genes in the border junction of IRb and SSC. Sequence divergence analysis of the protein coding genes showed that seven genes (petB, atpF, psaI, rpl32, rpl16, ycf1, and clpP) are under positive selection. The phylogenetic analysis revealed that Justiceae is sister to Ruellieae. This study reported the first cp genome of the largest genus in Acanthaceae and provided resources for studying genetic diversity of J. flava as well as resolving phylogenetic relationships within the core Acanthaceae.

Tryonia, a new taenitidoid fern genus segregated from Jamesonia and Eriosorus (Pteridaceae).

Project description:The Neotropical fern genera Eriosorus and Jamesonia have long been thought of as close relatives. Molecular phylogenetic studies have confirmed this notion but have also revealed that neither genus is monophyletic with respect to the other. As a result, all known species of Eriosorus were recently subsumed under the older generic name Jamesonia. Here, through an analysis of a four-gene plastid dataset, we show that several species traditionally treated in Eriosorus are in fact more closely related to other taenitidoid fern genera (namely Austrogramme, Pterozonium, Syngramma, and Taenitis) than they are to the large Jamesonia sensu lato clade. Tryonia Schuettp., J.Prado & A.T.Cochran gen. nov. is described to accommodate these species and four new combinations are provided. Tryonia is confined to southeastern Brazil and adjacent Uruguay; it is distinct (from most species of Jamesonia) in having stramineous rachises.

Adetogramma (Polypodiaceae), a new monotypic fern genus segregated from Polypodium.

Project description:Polypodiaceae is one of the most diverse and abundant families of ferns in tropical and subtropical forests. Despite multiple studies investigating its phylogeny and taxonomy, several generic boundaries within the family still need clarification. One of the most problematic circumscriptions is that of Polypodium L., and one species that still contributes to this uncertainty is Polypodium chrysolepis Hook. The main goal of this study was to use molecular and morphological data to clarify the relationships of P. chrysolepis inside the polygrammoid clade. Sequences from three plastid regions (cpDNA - rbcL, rps4 and rps4-trnS IGS) from fifty species belonging to thirty-two genera of Polypodiaceae were analyzed using maximum likelihood and Bayesian inference. Polypodium chrysolepis constitutes an isolated lineage among the neotropical polygrammoid ferns, close to Serpocaulon and the grammitids, and is recognized here in a new genus. It can be distinguished by its entire leaves with free veins and peltate, pedicellate, lanceolate paraphyses. A new combination, Adetogramma chrysolepis, is proposed and a new taxonomic treatment is presented; its conservation status was assessed using IUCN Red List Categories and Criteria.

Taxonomic revision of the Mesoamerican genus Spathacanthus (Justicieae, Acanthoideae, Acanthaceae).

Project description:Spathacanthus is a Mesoamerican genus that occurs in tropical and temperate regions from southern Mexico to Costa Rica; its taxonomy has not been updated for two decades. In view of the fact that a new species has been discovered and that the interspecific affinities in this genus have not been addressed to date, the present study aims to revise the genus Spathacanthus. Specimens of plants of this genus collected from across the distribution range and deposited in herbaria and digital databases were reviewed. In parallel, a cladistic analysis was carried out, based on morphological characters in order to examine relationships between species. Four species of Spathacanthus were recognised: one endemic to Costa Rica, another micro-endemic to Veracruz in Mexico, one more restricted to the forests of Mexico and Guatemala and the last one more widely distributed. Reflecting the previously limited knowledge of the group, many of the specimens that we studied had been misidentified. A key to differentiate these species is provided, supplemented with photographs, drawings and other illustrations, morphological descriptions, synonymy and ecological data. Results, presented here, extend the distribution range of some taxa and a distribution map is presented. The cladistic analysis recovered the genus as monophyletic, showing that S. hoffmannii and S. hahnianus are sister taxa and S. magdalenae was found to be more closely related to S. parviflorus. These plants are vulnerable to degradation and habitat loss.
