Eucalyptusdimers A-C, Dimeric Phloroglucinol-Phellandrene Meroterpenoids from Eucalyptus robusta.

ABSTRACT: Eucalyptusdimers A-C, three dimeric phellandrene-derived meroterpenoids featuring an unprecedented, fused skeleton between two phellandrene and two acylphloroglucinol subunits, along with one biogenetically related intermediate, (±)-eucalyprobusone A, were isolated from the fruits of Eucalyptus robusta. Their structures and absolute configurations were elucidated using spectroscopic data, X-ray crystallography, and electronic circular dichroism analysis. The isolated meroterpenoids were evaluated for their anti-inflammatory, acetylcholinesterase inhibitory, and protein tyrosine phosphatase 1B inhibitory effects.

New Formyl Phloroglucinol Meroterpenoids from the Leaves of Eucalyptus robusta.

Project description:Seven new formyl phloroglucinol meroterpenoids (FPMs), namely eucalrobusones J-P (1-7), as well as three known ones (8-10) were isolated from the leaves of Eucalyptus robusta. Their structures were elucidated by spectroscopic data analysis, and their absolute configurations were determined by applications of the Snatzke's helicity rule and the electron circular dichroism (ECD) calculation. These FPMs are diverse in coupling patterns between phloroglucinol and sesquiterpenoid units, forming novel polycyclic ring systems. Compound 1 possesses a new carbon skeleton that a 1-oxaspiro[5.6]dodecane core is formed through C-14 rather than C-4 of the aromadendrane moiety. Compound 2 features a novel 6/7/5 ring-fused 6-oxabicyclo[3.2.2]nonane skeleton. Compounds 3-5 are rare aristolane-based FPMs. By forming different oxo bridges, compound 3 is the first sample of FPM with benzo-dihydrofuran structure, and compound 4 possesses a novel 6/6/6/6/3-fused pentacyclic skeleton. Compounds 1, 6, and 8 exhibited significant antifungal activities against Candida glabrata with MIC50 values of 2.57, 1.95, and 2.49 μg/mL, respectively.

Structurally Diverse Polymethylated Phloroglucinol Meroterpenoids from Baeckea frutescens.

Project description:Phytochemical investigation of the MeOH extract of twigs and leaves of Baeckea frutescens led to the isolation of seven new polymethylated phloroglucinol meroterpenoids (PPMs), named baeckfrutones M-S (1-7). Their structures and absolute configurations were determined by spectroscopic analyses, chiral-phase HPLC analysis, and electronic circular dichroism (ECD) calculations. PPM 1 is a novel meroterpenoid possessing a 6/6/5/3 tetracyclic skeleton in PPMs, whereas 3 and 4 are the first hydroxytasmanone type phloroglucinol-monoterpene hybrids. (+)-2 and 7 displayed potent anti-inflammatory activity with IC50 values of 20.86?±?0.60 and 36.21?±?1.18 ??, respectively.

Acylphloroglucinols with acetylcholinesterase inhibitory effects from the fruits of Eucalyptus robusta.

Project description:Eleven new acylphloroglucinols, including six new formylated phloroglucinol-monoterpene meroterpenoids, eucalyprobusals A-F (1-6), one monomeric acylphloroglucinol, eucalyprobusone B (7), and four dimeric acylphloroglucinols, eucalyprobusones C-F (8-11) were purified from the fruits of Eucalyptus robusta. The establishment of the structures of 1-11 was achieved by a combination of NMR and HRESIMS data analyses, electron circular dichroism (ECD), and single-crystal X-ray diffraction. Compounds 6, 8, and an inseparable mixture of 10 and 11 were found to be potent AChE inhibitors with IC50 values of 3.22 ± 0.36, 3.82 ± 0.22, and 2.55 ± 0.28 μΜ, respectively. Possible interaction sites of 6, 8, 10, and 11 with AChE were investigated by means of molecular docking studies, and the results revealed that AChE residues Asn87, Ser125, Thr83, Tyr133, Tyr124, Tyr337, and Tyr341 played crucial roles in the observed activity of the aforementioned compounds.

Quantification and Localization of Formylated Phloroglucinol Compounds (FPCs) in <i>Eucalyptus</i> Species.

Project description:The Eucalyptus genus is a hyper-diverse group of long-lived trees from the Myrtaceae family, consisting of more than 700 species. Eucalyptus are widely distributed across their native Australian landscape and are the most widely planted hardwood forest trees in the world. The ecological and economic success of Eucalyptus trees is due, in part, to their ability to produce a plethora of specialized metabolites, which moderate abiotic and biotic interactions. Formylated phloroglucinol compounds (FPCs) are an important class of specialized metabolites in the Myrtaceae family, particularly abundant in Eucalyptus. FPCs are mono- to tetra-formylated phloroglucinol based derivatives, often with an attached terpene moiety. These compounds provide chemical defense against herbivory and display various bioactivities of pharmaceutical relevance. Despite their ecological and economic importance, and continued improvements into analytical techniques, FPCs have proved challenging to study. Here we present a simple and reliable method for FPCs extraction, identification and quantification by UHPLC-DAD-ESI-Q-TOF-MS/MS. The method was applied to leaf, flower bud, and flower samples of nine different eucalypt species, using a small amount of plant material. Authentic analytical standards were used to provide high resolution mass spectra and fragmentation patterns. A robust method provides opportunities for future investigations into the identification and quantification of FPCs in complex biological samples with high confidence. Furthermore, we present for the first time the tissue-based localization of FPCs in stem, leaf, and flower bud of Eucalyptus species measured by mass spectrometry imaging, providing important information for biosynthetic pathway discovery studies and for understanding the role of those compounds in planta.

The characteristics of the complete chloroplast genome for <i>Eucalyptus robusta</i> (Myrtaceae).

Project description:Eucalyptus robusta is a great tree of Myrtaceae. It is mainly distributed in Southeast provinces of China and Southeast Asian countries. There is no study on the genome of E. robusta far. Here we report the complete chloroplast genome of E. robusta, assembled from whole-genome high-throughput sequencing data, as a resource for future studies on the taxonomy and evolution of E. robusta. The complete chloroplast genome of E. robusta is 160,201 bp in length with a typical quadripartite structure, consisting of a large single-copy region (LSC, 88,905 bp), a single-copy region (SSC, 18,498 bp) and a pair of inverted repeats (IRs, 26,399 bp). It was predicted to contain a total of 128 genes, with an overall GC content of 36.86%. Phylogenetic analysis placed E. robusta closest to Eucalyptus globulus.

Structure Revision of Formyl Phloroglucinol Meroterpenoids: A Unified Approach Using NMR Fingerprinting and DFT NMR and ECD Analyses.

Project description:NMR fingerprints are valuable tools for analyzing complex natural product mixtures and identifying incorrectly assigned structures in the literature. Our diagnostic NMR fingerprints for formyl phloroglucinol meroterpenoids revealed discrepancies in the structures reported for eucalyprobusal C (1a) and eucalypcamal K (2a). NMR fingerprinting PCA analyses identified 1a as an oxepine-diformyl phloroglucinol and 2a as an oxepine 3-acyl-1-formyl phloroglucinol, contrary to their initial assignments as pyrano-diformyl and pyrano 3-acyl-1-formyl phloroglucinols, respectively. Extensive reinterpretation of their reported one- and two-dimensional NMR data, coupled with GIAO DFT-calculated 1H and 13C NMR chemical shift and DP4+ analyses, supported the unequivocal reassignment of eucalyprobusal C to 1b and eucalypcamal K to 2b. The absolute configurations of the revised oxepine-containing phloroglucinol meroterpenoids were confirmed via the reinterpretation of their reported ROESY and NOESY NMR data, along with comparative TDDFT-calculated and experimental ECD spectra.

Euglobal-IIIa, a novel acylphloroglucinol-sesquiterpene derivative from Eucalyptus robusta: absolute structure and cytotoxicity

Project description:Euglobal-IIIa (1), a novel acylphloroglucinol-sesquiterpene derivative, and a known analogue, have been isolated from leaves of Eucalyptus robusta. The structures was elucidated by extensive spectroscopic data and by comparison with data reported in literature, while the absolute configuration of 1 was determined by the X-ray diffraction analysis. Compound 1 exhibited comparable cytotoxicity with that of cisplatin against five human cancer cell lines HL-60, SMMC-7721, A-549, MCF-7, and SW480 with IC50 values of 15.7, 15.5, 17.6, 14.3, and 21.8 µM, respectively. Biomimetic Synthesis and Chemical Proteomics Reveal the Mechanism of Action and Functional Targets of Lysine-Reactive Phloroglucinol Meroterpenoids

Project description:Natural products have long been an enduring source of drug leads and chemical probes that have led to the development of numerous medicines. Despite their scarcity, the discovery of natural products that modulate protein function through covalent interactions with lysine residues holds immense potential to unlock new therapeutic interventions and advance our understanding of the intricate biological processes governed by these modifications. Phloroglucinol meroterpenoids comprise one of the largest classes of natural products displaying a vast array of biological activities. However, their mechanism of action and molecular targets have remained largely unexplored. In this study, we present an in-depth experimental and computational investigation into the synthesis, physicochemical and kinetic parameters, molecular mechanism of action, and functional cellular targets of selected phloroglucinol meroterpenoids. We leverage synthetic clickable analogues of natural products to observe disparate proteome-wide reactivity by in-gel fluorescence scanning and cell imaging. By implementing sample multiplexing and a redesigned desthiobiotin-based probe, we streamline a quantitative activity-based protein profiling protocol for mapping proteome-wide reactivity and ligandability of proteinaceous lysines directly in human cells. Using this platform, we identify numerous lysine-phloroglucinol meroterpenoid interactions in breast cancer cells that occur at functional sites on proteins from diverse structural and functional classes. Lastly, we demonstrate that phloroglucinol meroterpenoids perturb diverse biochemical functions through stereoselective and site-specific modification of lysines in proteins involved in glycolysis, lipid metabolism, and mitochondrial respiration. These findings underscore the broad potential of phloroglucinol meroterpenoids for targeting functional lysines in the human proteome.
