Project description:In the furrow illusion (Anstis, 2012), the perceived path of a moving target follows the veridical path orientation when viewed foveally, but follows the orientation of the texture when viewed peripherally. These radically different motion percepts depending on whether the stimulus is viewed foveally or peripherally has led Anstis to conclude that the furrow illusion reveals "profound differences in the way that the periphery and fovea process visual motion." In the current study, we rather argue that the different percepts can be explained by reduced position acuity with eccentricity and therefore do not imply different ways of processing motion per se. If feature tracking, which is position-based, is involved in the perception of the veridical motion direction, then impairing the feature tracking motion system should strengthen the illusion. To reduce contribution of the feature tracking motion system, we used a crowding paradigm consisting in presenting many nearby targets. We found that under crowding conditions, the furrow illusion was stronger. We conclude that feature tracking was involved in the perception of the veridical motion direction, which is compatible with the hypothesis that the different motion percepts at fixation and in the periphery are due to a reduced position acuity with eccentricity affecting feature tracking, not to different ways of processing motion per se.

Project description:During cytokinesis, cleavage furrow invagination requires an actomyosin-based contractile ring and addition of new membrane. Little is known about how this actin and membrane traffic to the cleavage furrow. We address this through live analysis of fluorescently tagged vesicles in postcellularized Drosophila melanogaster embryos. We find that during cytokinesis, F-actin and membrane are targeted as a unit to invaginating furrows through formation of F-actin-associated vesicles. F-actin puncta strongly colocalize with endosomal, but not Golgi-derived, vesicles. These vesicles are recruited to the cleavage furrow along the central spindle and a distinct population of microtubules (MTs) in contact with the leading furrow edge (furrow MTs). We find that Rho-specific guanine nucleotide exchange factor mutants, pebble (pbl), severely disrupt this F-actin-associated vesicle transport. These transport defects are a consequence of the pbl mutants' inability to properly form furrow MTs and the central spindle. Transport of F-actin-associated vesicles on furrow MTs and the central spindle is thus an important mechanism by which actin and membrane are delivered to the cleavage furrow.

Project description:Cytokinesis is the process of physical cleavage at the end of cell division; it proceeds by ingression of an acto-myosin furrow at the equator of the cell. Its failure leads to multinucleated cells and is a possible cause of tumorigenesis. Here, we calculate the full dynamics of furrow ingression and predict cytokinesis completion above a well-defined threshold of equatorial contractility. The cortical acto-myosin is identified as the main source of mechanical dissipation and active forces. Thereupon, we propose a viscous active nonlinear membrane theory of the cortex that explicitly includes actin turnover and where the active RhoA signal leads to an equatorial band of myosin overactivity. The resulting cortex deformation is calculated numerically, and reproduces well the features of cytokinesis such as cell shape and cortical flows toward the equator. Our theory gives a physical explanation of the independence of cytokinesis duration on cell size in embryos. It also predicts a critical role of turnover on the rate and success of furrow constriction. Scaling arguments allow for a simple interpretation of the numerical results and unveil the key mechanism that generates the threshold for cytokinesis completion: cytoplasmic incompressibility results in a competition between the furrow line tension and the cell poles' surface tension.

Project description:The mitotic spindle determines the cleavage furrow site during metazoan cell division, but whether other mechanisms exist remains unknown. Here we identify a spindle-independent mechanism for cleavage furrow positioning in Drosophila neuroblasts. We show that early and late furrow proteins (Pavarotti, Anillin, and Myosin) are localized to the neuroblast basal cortex at anaphase onset by a Pins cortical polarity pathway, and can induce a basally displaced furrow even in the complete absence of a mitotic spindle. Rotation or displacement of the spindle results in two furrows: an early polarity-induced basal furrow and a later spindle-induced furrow. This spindle-independent cleavage furrow mechanism may be relevant to other highly polarized mitotic cells, such as mammalian neural progenitors.

Project description:In a pattern of horizontal lines containing ± 45° zigzagging phase-shifted strips, vivid illusory motion is perceived when the pattern is translated up or down at a moderate speed. Two forms of illusory motion are seen: [i] a motion "racing" along the diagonal interface between the strips and [ii] lateral (sideways) motion of the strip sections. We found the relative salience of these two illusory motions to be strongly influenced by the vertical spacing and length of the line gratings, and the period length of the zigzag strips. Both illusory motions are abolished when the abutting strips are interleaved, separated by a gap or when a real line is superimposed at the interface. Illusory motion is also severely weakened when equiluminant colored grating lines are used. Illusory motion perception is fully restored at < 20% luminance contrast. Using adaptation, we find that line-ends alone are insufficient for illusory motion perception, and that both physical carrier motion and line orientation are required. We finally test a classical spatiotemporal energy model of V1 cells that exhibit direction tuning changes that are consistent with the direction of illusory motion. Taking this data together, we constructed a new visual illusion and surmise its origin to interactions of spatial and temporal energy of the lines and line-ends preferentially driving the magnocellular pathway.

Project description:We report a novel visual illusion we call the Ring Rotation Illusion (RRI). When a ring of stationary points replaces a circular outline, the ring of points appears to rotate to a halt, although no actual motion has been displayed. Three experiments evaluate the clarity of the illusory rotation. Clarity decreased as the diameter of the circle and ring increased and increased as the number of points forming the ring increased. The optimal interstimulus interval (ISI) between the circle and ring was 90 ms when stimulus presentations lasted 100 ms but 0 ms with 500 ms presentations. We compare the RRI to the Motion Bridging Effect (MBE), a similar illusion in which a stationary ring of points replaces an initial ring of points that spins so rapidly it looks like a stationary outline. A rotation of the stationary ring is seen that usually matches the direction of the initial ring's invisible spin. Participants reported a slightly more frequent and clearer motion percept with the MBE than RRI. ISI manipulations had similar effects on the two illusions, but the effects of number of points and ring diameter were largely restricted to the RRI. We suggest that both the RRI and MBE motion percepts are produced by a visual heuristic that holds that the transition from an outline circle to a ring of points is plausibly explained by a rapid spin decelerating to a halt, but in the case of the MBE, an additional direction-sensitive mechanism contributes to this percept.

Project description:A recent analysis of magic tricks suggests the existence of a perceptual illusion where the space hidden behind an occluding object is experienced as empty in a strangely compelling way. Here, we show that this illusion of absence is not just a trivial consequence of the lack of retinal stimulation but rather the result of an active process of perceptual construction. The results of a simple experiment show that this perceptual illusion of absence can in turn trigger perceptual processes which generate an immediate perceptual impression of levitation via a percept-percept coupling. This suggests that magical illusions of levitation are partially driven by an immediate perceptual impression of floating in thin air. The perceptual mechanisms underlying the illusion of absence are hitherto unknown, but our results provide support for a potential explanation based on the generic view principle.

Project description:Visual crowding, as context modulation, reduce the ability to recognize objects in clutter, sets a fundamental limit on visual perception and object recognition. It's considered that crowding does not exist in the fovea and extensive efforts explored crowding in the periphery revealed various models that consider several aspects of spatial processing. Studies showed that spatial and temporal crowding are correlated, suggesting a tradeoff between spatial and temporal processing of crowding. We hypothesized that limiting stimulus availability should decrease object recognition in clutter. Here we show, for the first time, that robust contour interactions exist in the fovea for much larger target-flanker spacing than reported previously: participants overcome crowded conditions for long presentations times but exhibit contour interaction effects for short presentation times. Thus, by enabling enough processing time in the fovea, contour interactions can be overcome, enabling object recognition. Our results suggest that contemporary models of context modulation should include both time and spatial processing.
