Polybrominated Diphenyl Ethers: Structure Determination and Trends in Antibacterial Activity.

ABSTRACT: Antibacterial-guided fractionation of the Dictyoceratid sponges Lamellodysidea sp. and two samples of Dysidea granulosa yielded 14 polybrominated, diphenyl ethers including one new methoxy-containing compound (8). Their structures were elucidated by interpretation of spectroscopic data of the natural product and their methoxy derivatives. Most of the compounds showed strong antimicrobial activity with low- to sub-microgram mL(-1) minimum inhibitory concentrations against drug-susceptible and drug-resistant strains of Staphylococcus aureus and Enterococcus faecium, and two compounds inhibited Escherichia coli in a structure-dependent manner.

Time Trends of Polybrominated Diphenyl Ethers (PBDEs) in Antarctic Biota.

Project description:Polybrominated diphenyl ethers (PBDEs) are "emerged" contaminants that were produced and used as flame retardants in numerous consumer and industrial applications for decades until banned. They remain ubiquitously present in the environment today. Here, a unique set of >200 biotic samples from the Antarctic was analyzed for PBDEs, including phytoplankton, krill, fish, and fur seal milk, spanning several sampling seasons over 14 years. PBDE-47 and -99 were the dominant congeners determined in all samples, constituting >60% of total PBDEs. A temporal trend was observed for ∑7PBDE concentrations in fur seal milk, where concentrations significantly increased (R2 = 0.57, p < 0.05) over time (2000-2014). Results for krill and phytoplankton also suggested increasing PBDE concentrations over time. Trends of PBDEs in fur seal milk of individual seals sampled 1 or more years apart showed no clear temporal trends. Overall, there was no indication of PBDEs decreasing in Antarctic biota yet, whereas numerous studies have reported decreasing trends in the northern hemisphere. Similar PBDE concentrations in perinatal versus nonperinatal milk implied the importance of local PBDE sources for bioaccumulation. These results indicate the need for continued assessment of contaminant trends, such as PBDEs, and their replacements, in Antarctica.

Polybrominated diphenyl ethers, hydroxylated polybrominated diphenyl ethers, and measures of thyroid function in second trimester pregnant women in California.

Project description:Prenatal exposure to polybrominated diphenyl ethers (PBDEs) may disrupt thyroid function and contribute to adverse neurodevelopmental outcomes. We conducted a pilot study to explore the relationship between serum concentrations of lower-brominated PBDEs (BDE-17 to -154), higher-brominated PBDEs (BDE-183 to -209), and hydroxylated PBDE metabolites (OH-PBDEs) with measures of thyroid function in pregnant women. Concentrations of PBDEs, OH-PBDEs, thyroid-stimulating hormone (TSH), total thyroxine (T(4)), and free T(4) were measured in serum samples collected between 2008 and 2009 from 25 second trimester pregnant women in California. Median concentrations of lower-brominated PBDEs and OH-PBDEs were the highest reported to date in pregnant women. Median concentrations of BDE-47 and the sum of lower-brominated PBDEs (ΣPBDE(5)) were 43.1 ng/g lipid and 85.8 ng/g lipid, respectively, and the sum of OH-PBDEs (ΣOH-PBDE(4)) was 0.084 ng/mL. We observed a positive association between the weighted sum of chemicals known to bind to transthyretin (ΣTTR binders) and TSH levels. We also found positive associations between TSH and ΣPBDE(5), ΣOH-PBDE(4), BDE-47, BDE-85, 5-OH-BDE47, and 4'-OH-BDE49, and an inverse association with BDE-207. Relationships with free and total T(4) were weak and inconsistent. Our results indicate that PBDE exposures are elevated in pregnant women in California and suggest a relationship with thyroid function. Further investigation is warranted to characterize the risks of PBDE exposures during pregnancy.

Project description:Extensive use of polybrominated diphenyl ethers (PBDEs) has resulted in widespread environmental distribution and concerns remain about risks to human health and ecosystems from legacy PBDE contamination. Though bioremediation has been proven to be a cost-effective and efficient technology for treating halogenated pollutants in situ, application of bioremediation technologies at PBDE-contaminated sites is restricted by a lack of knowledge about functional PBDE-dehalogenating microbes. In the current study, we observed distinct PBDE-dehalogenation activity by three previously isolated Dehalococcoides mccartyi strains (CG1, CG4, and 11a5). PBDE debromination proceeded via distinct pathways in each D. mccartyi strain and involved previously characterized (TceA11a5, PcbA1, and PcbA4) and uncharacterized (11a5_e001) reductive dehalogenases (RDase), suggesting that this phenotype may be more widespread among the Dehalococcoides than is currently recognized.

Patterns and Trends of Polybrominated Diphenyl Ethers in Bald Eagle Nestlings in Minnesota and Wisconsin, USA.

Project description:We measured concentrations of up to 17 polybrominated diphenyl ethers (PBDEs) in plasma of 492 bald eagle (Haliaeetus leucocephalus) nestlings between 1995 and 2017 from 12 study areas in Wisconsin and Minnesota, USA. Geometric mean concentrations of the sum of 9 PBDE congeners (∑PBDE) measured across all years ranged from 2.88 to 10.8 µg/L, and nestlings in urban areas had higher concentrations than those in remote locations. Region-wide from 2006 through 2017, we found that ∑PBDEs declined by 3.8% annually and congeners BDE-47, -99, and -100 declined by 5.6 to 6.5%, whereas BDE-153 and -154 had no significant declines. When categorized by waterbody type, nestlings from Great Lakes and river study areas had higher concentrations of ∑PBDEs than those at inland lakes, but river study areas spanned the extremes. From 2006 to 2017, ∑PBDEs declined by 7.3% annually in Great Lakes nestlings and by 3.2% in nestlings along rivers, and increased by 32.7% at inland lakes. Using a longer dataset (1995-2015), we found that ∑PBDEs declined in Lake Superior nestlings by 3.3% annually. Our results show that PBDEs declined in bald eagle nestling plasma in most study areas since PBDE production was reduced, but that concentrations remain high near urban centers and that trends differ by congener, study area, and waterbody type. Aerobic biotransformation of polybrominated diphenyl ethers (PBDEs) by bacterial isolates.

Project description:Polybrominated diphenyl ethers (PBDEs) are flame retardants that have been used in consumer products and furniture for three decades. Currently, very little is known about their fate in the environment and specifically about their susceptibility to aerobic biotransformation. Here, we investigated the ability of the polychlorinated biphenyl (PCB) degrading bacteria Rhodococcus jostii RHA1 and Burkholderia xenovorans LB400 to transform mono- through hexa-BDEs at ppb levels. We also tested the PBDE transforming abilities of the related strain Rhodococcus sp. RR1 and the ether-degrading Pseudonocardia dioxanivorans CB1190. The two PCB-degrading strains transformed all of the mono- through penta-BDEs and strain LB400 transformed one of the hexa-BDEs. The extent of transformation was inversely proportional to the degree of bromination. Strains RR1 and CB1190 were only able to transform the less brominated mono- and di-BDE congeners. RHA1 released stoichiometric quantities of bromide while transforming mono- and tetra-BDE congeners. LB400 instead converted most of a mono-BDE to a hydroxylated mono-BDE. This is the first report of aerobic transformation of tetra-, penta,- and hexa-BDEs as well as the first report of stoichiometric release of bromide during PBDE transformation.
