Project description:The emergence of porcine epidemic diarrhea (PED) in commercial swine in North America and growing concerns about the potential for the introduction of African swine fever (ASF) from China, the European Union, or other affected regions has put a spotlight on the possible role of contaminated feed and feed ingredients in the introduction and transmission of viral swine pathogens. This paper systematically reviews the scientific literature regarding whether non-animal origin ingredients of commercial swine feed could introduce or transmit viral pathogens of swine into or within the United States. The purpose of this review is to identify, evaluate, and summarize the relevant scientific knowledge, published through March 2018, and to identify information gaps and research needs, thereby making the available evidence more accessible to policy makers, the swine industry, and the scientific community. A total of 26 documents were selected for the final review process, which included experimental studies, case reports, epidemiological investigations, and scientific opinion, among others. The review found that the scientific literature has addressed some critical experimental questions pertaining to transmission of swine viruses via feed and feed ingredients, but the current body of scientific knowledge lacks conclusive evidence of virus contamination of non-animal origin feed ingredients of commercial swine feed, particularly for imported commodities, and further investigation into the epidemiology of virus transmission via feed to swine under field conditions through natural feeding behavior is warranted. Additional studies of how imported ingredients of commercial swine feed are sourced, processed, transported and, thus, contaminated prior to importation into the United States are needed. Moving forward, studies designed to examine the likely source(s) of contamination and subsequent virus mitigation steps in processing and post-processing may be the most fruitful focus of research.

Project description:Feeds contribute highly to environmental impacts of livestock products. Therefore, formulating low-impact feeds requires data on environmental impacts of feed ingredients with consistent perimeters and methodology for life cycle assessment (LCA). We created the ECOALIM dataset of life cycle inventories (LCIs) and associated impacts of feed ingredients used in animal production in France. It provides several perimeters for LCIs (field gate, storage agency gate, plant gate and harbour gate) with homogeneously collected data from French R&D institutes covering the 2005-2012 period. The dataset of environmental impacts is available as a Microsoft® Excel spreadsheet on the ECOALIM website and provides climate change, acidification, eutrophication, non-renewable and total cumulative energy demand, phosphorus demand, and land occupation. LCIs in the ECOALIM dataset are available in the AGRIBALYSE® database in SimaPro® software. The typology performed on the dataset classified the 149 average feed ingredients into categories of low impact (co-products of plant origin and minerals), high impact (feed-use amino acids, fats and vitamins) and intermediate impact (cereals, oilseeds, oil meals and protein crops). Therefore, the ECOALIM dataset can be used by feed manufacturers and LCA practitioners to investigate formulation of low-impact feeds. It also provides data for environmental evaluation of feeds and animal production systems. Included in AGRIBALYSE® database and SimaPro®, the ECOALIM dataset will benefit from their procedures for maintenance and regular updating. Future use can also include environmental labelling of commercial products from livestock production.

Project description:Mycotoxins are toxic secondary fungal metabolites that can negatively affect animal productivity when ingested through feed. In order to assess mycotoxin contamination of poultry feed and feed ingredients vis-a-vis source tracking of feed contamination in Nigeria, 102 samples of feed (n = 30) and feed ingredients (n = 72) were collected from in-house mills of poultry farms across 12 states of Nigeria and analyzed for multiple mycotoxins using LC/MS-MS. One hundred and forty microbial metabolites were detected in the feed and feed ingredients. The most frequent mycotoxin in the feed was fumonisin B1, occurring in 97% of the samples at mean concentration of 1014 μg kg-1. AFB1 occurred in 83% of the feed samples at mean concentration of 74 μg kg-1 and in all feed ingredients except fish meal and other cereals (millet and rice). Feed samples analyzed in this study were contaminated with at least four mycotoxins: aflatoxins and fumonisin co-occurring in 80% of the samples. Peanut cake and maize contributed the most to the levels of aflatoxin and fumonisin, respectively, in the feed. Consequently, there is a need to explore other cereal- and protein-based ingredients for compounding feeds in order to reduce the risk associated with high mycotoxin (e.g. aflatoxin) intake in poultry.

Project description:Borna disease (BD), caused by the neurotropic RNA virus, Borna Disease virus, is an affliction ranging from asymptomatic to fatal meningoencephalitis across naturally and experimentally infected warmblooded (mammalian and bird) species. More than 100 years after the first clinical descriptions of Borna disease in horses and studies beginning in the 1980's linking Borna disease virus to human neuropsychiatric diseases, experimentally infected rodents have been used as models for examining behavioral, neuropharmacological, and neurochemical responses to viral challenge at different stages of life. These studies have contributed to understanding the role of CNS viral injury in vulnerability to behavioral, developmental, epileptic, and neurodegenerative diseases and aided evaluation of the proposed and still controversial links to human disease.

Project description:African swine fever virus (ASFV) causes a fatal infectious disease affecting domestic pigs and wild boars. ASFV is highly stable and easily transmitted by consumption of contaminated swine feed and pork products. Heat treatment of feed ingredients is a means to minimize the risk of contamination through swine feed consumption. The objectives of this study were to determine the thermal inactivation of ASFV in non-animal and animal origin feed ingredients. The rate of thermal inactivation is represented by decimal reduction time (DT) or time required to reduce ASFV per 1 log at temperature T. The mean D60, D70, D80 and D85 of meat and bone meal (MBM), soybean meal (SBM), and maize grain (MZ) are in the ranges 5.11-6.78, 2.19-3.01, 0.99-2.02, and 0.16-0.99 min, respectively. DT is used to compare the heat resistance of ASFV in the feed ingredient matrices. The mean DT of ASFV in MBM, SBM and MZ was not statistically significant, and the heat resistance of ASFV in MBM, SBM, and MZ was not different at 60, 70, 80, or 85 °C. The multiple DT was used to develop a DT model to predict DT at various inactivation temperatures. The DT models for MBM, SBM, and MZ are log DT = - [Formula: see text] + 2.69, log DT = - [Formula: see text] + 2.55, and log DT = - [Formula: see text] + 4.01. To expand and ease the field applications, a spreadsheet predicting the DT and the inactivation time (with 95% confidence interval) from these DT models is available to download.

Project description:BackgroundThe development of public health policy is inextricably linked with governance structure. In our increasingly globalized world, human migration and infectious diseases often span multiple administrative jurisdictions that might have different systems of government and divergent management objectives. However, few studies have considered how the allocation of regulatory authority among jurisdictions can affect disease management outcomes.MethodsHere we evaluate the relative merits of decentralized and centralized management by developing and numerically analyzing a two-jurisdiction SIRS model that explicitly incorporates migration. In our model, managers choose between vaccination, isolation, medication, border closure, and a travel ban on infected individuals while aiming to minimize either the number of cases or the number of deaths.ResultsWe consider a variety of scenarios and show how optimal strategies differ for decentralized and centralized management levels. We demonstrate that policies formed in the best interest of individual jurisdictions may not achieve global objectives, and identify situations where locally applied interventions can lead to an overall increase in the numbers of cases and deaths.ConclusionsOur approach underscores the importance of tailoring disease management plans to existing regulatory structures as part of an evidence-based decision framework. Most importantly, we demonstrate that there needs to be a greater consideration of the degree to which governance structure impacts disease outcomes.

Project description:Currently, clinical gene therapy is experiencing a renaissance, with new products for clinical use approved in Europe and clinical trials for multiple diseases reporting positive results, especially those using recombinant adeno-associated viral (rAAV) vectors. Amid this new success, it is prudent to recall that the field of gene therapy experienced tragic setbacks in 1999 and 2002 because of the serious adverse events related to retroviral and adenoviral gene delivery in two clinical trials that resulted in the death of two patients. In both cases, the toxicity observed in humans had been documented to occur in animal models. However, these toxicities were either undetected or underappreciated before they arose in humans. rAAVs have been tested extensively in animals and animal models of disease, largely without adverse events, except for transient elevation in liver enzymes in some patients. However, a small but growing number of murine studies have documented that adeno-associated viral gene delivery can result in insertional mutagenesis. Herein, the aggregate data are reviewed from multiple murine studies where genotoxicity associated with rAAV treatment has been observed. The data emphasize the need for a proactive position to evaluate the potential risks and possible solutions associated with AAV-mediated gene therapy.

Project description:Objective:The study obtained data on the potential of local feed ingredients, both in quantity, quality, and continuity and to observe the absorption power and ability of livestock to use feed designed in vitro. Materials and Methods:The method uses a survey method of potential sources of animal feed and calculates the carrying capacity of ruminants and nutritional analysis of feed ingredients with proximate analysis. Feed formulations were made based on proximate analysis results of four formulas (P1, P2, P3, and P4) and tested in vitro. Results:First, Nganjuk district has the highest of the local food potential with the production of agricultural waste and agricultural industries, reaching 802,341.94 tons/year. Second, the most top carrying capacity analysis reached in Tulungagung district, which reached 62,534 ST/year or 43% of the total population of ruminants. Third, the results of the study of the quality of local feed ingredients indicate that each type of feed material has the right and proper nutrition given to ruminants. Fourth, the in vitro testing included showing P1 feed that had a very significant effect (p < 0.01) on dry matter digestibility and digestibility of organic matter of cows, respectively, 74.69% and 73.39%. Conclusion:The in vitro technique of making animal feed can be developed in the areas that have the potential to produce agricultural waste and agricultural industries to increase the carrying capacity of livestock.
