Isosakuranetin-5-O-rutinoside: a new flavanone with antidepressant activity isolated from Salvia elegans Vahl.

ABSTRACT: Ursolic acid (1) and a new flavanone, 5-O-(6-rhamnosylglucoside)-7-hydroxy-4'-methoxyflavanone (2), were isolated from the leaves of Salvia elegans Vahl. These natural products displayed antidepressant activity in mice as determined by means of a forced swimming test (FST) evaluation. Structural elucidation was carried out by chemical derivatization (acetylation) and spectroscopic analyses, such as 1H- and 13C-NMR and two-dimensional (2-D) COSY, heteronuclear multiple quantum coherence (HMQC), and heteronuclear multiple-bond correlation (HMBC) spectroscopy experiments.

A New Flavanone as a Potent Antioxidant Isolated from Chromolaena odorata L. Leaves.

Project description:Chromolaena odorata L. (Asteraceae) is one of the tropical plants which is widely used as traditional medicines for diabetes and soft tissue wounds treatment in some regions in East Indonesia. The present study was aimed at determining the bioactive compounds of C. odorata leaves. The methanol and ethyl acetate extracts of C. odorata leaves have the inhibitory activity against 2,2-diphenyl-1-picryl-hydrazyl (DPPH) and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radicals as well as ?-glucosidase rat intestine enzyme. A new flavanone was isolated from the methanol extract and elucidated as 5,3'-dihydroxy-7,6'-dimethoxyflavanone or, namely, odoratenin (1) together with two known compounds: isosakuranetin (2) and subscandenin (3). The antioxidant activity of odoratenin (1) exhibited very potent ABTS radical inhibitory activity with IC50 value of 23.74 ?M which is lower than that of trolox (IC50 31.32 ?M) as a positive control. The result showed that a new flavanone, odoratenin (1), should be potential as an antioxidant source.

Characterization of the complete chloroplast genome of Salvia tiliifolia Vahl (Lamiaceae).

Project description:Salvia tiliifolia Vahl is native to Central America and considered as an invasive plant in Mexico, the United States, Ethiopia, South Africa, and Australia. The complete chloroplast (cp) genome of S. tiliifolia was 150,836 bp in length, which contained a large single-copy (LSC) region of 82,129 bp, a small single-copy (SSC) region of 17,533 bp, and a pair of inverted repeats (IRs) region of 25,587 bp each. The genome harbored 130 genes, including 85 protein coding genes, 37 tRNA genes, and 8 ribosomal RNA genes. The overall GC content was 37.99%. Phylogenetic analysis indicated that S. tiliifolia is closely related to the species of Salvia chanryoenica.

Trypanocidal Activity of Flavanone Derivatives.

Project description:Chagas disease, also known as American trypanosomiasis, is classified as a neglected disease by the World Health Organization. For clinical treatment, only two drugs have been on the market, Benznidazole and Nifurtimox, both of which are recommended for use in the acute phase but present low cure rates in the chronic phase. Furthermore, strong side effects may result in discontinuation of this treatment. Faced with this situation, we report the synthesis and trypanocidal activity of 3-benzoyl-flavanones. Novel 3-benzoyl-flavanone derivatives were prepared in satisfactory yields in the 3-step synthetic procedure. According to recommended guidelines, the whole cell-based screening methodology was utilized that allowed for the simultaneous use of both parasite forms responsible for human infection. The majority of the tested compounds displayed promising anti-Trypanosoma cruzi activity and the most potent flavanone bearing a nitrofuran moiety was more potent than the reference drug, Benznidazole.

Composition and Antioxidant, Antienzymatic and Antimicrobial Activities of Volatile Molecules from Spanish Salvia lavandulifolia (Vahl) Essential Oils.

Project description:The current study describes the composition of Salvia lavandulifolia (Vahl) essential oils (SlEOs) obtained from plants cultivated in Murcia (Spain), as determined by gas chromatography. Relative and absolute concentrations, the enantiomeric ratios of chiral compounds and the in vitro antioxidant, antienzymatic and antimicrobial activities are described. The main components of the SlEOs were camphor, 1,8-cineole, camphene and α-pinene, and the main enantiomers were (+)-camphor and (-)-camphene. The activities against free radicals and the capacity to reduce and chelate metallic ions were measured. SlEO-3 showed the highest activity in ORAC, DPPH, ABTS and reducing power methods, while SlEO-1 exhibited the highest chelating power. The activity of lipoxygenase and acetylcholinesterase could be inhibited by all the SlEOs, being bornyl acetate and limonene the most active individual compounds against lipoxygenase and 1,8-cineole against acetylcholinesterase. SlEOs and some individual compounds inhibited Escherichia coli, Staphylococcus aureus and Candida albicans. These results increase our knowledge of SlEOs and, particularly, provide for the first time a complete characterization of SlEOs from Murcia, Spain, while proposing possible biotechnological uses for them.

Four New Depsides Isolated from Salvia miltiorrhiza and Their Significant Nerve-Protective Activities.

Project description:By investigating of the roots of Salvia miltiorrhiza, which is one of the most widely used Chinese herbs, we used phytochemical methods successfully to obtain twelve depsides: four depsides (1⁻4) that were previously undescribed, along with eight known ones (5⁻12). Their structure characteristics were assessed by HR-ESIMS, CD, NMR (¹H, 13C, HSQC, HMBC) data analyses. These four newly isolated compounds (1⁻4), as well as the other eight compounds (5⁻12), show extraordinary protective effects on hydrogen peroxide-induced apoptosis in HS-SY5Y cells. Among them, depside 4 and depside 6 displayed more obviously protective effects than others.

A new prenylated flavanone from Derris trifoliata Lour.

Project description:A new flavanone, 4',5,7-trihydroxy-6,8-di-(2-hydroxy-3-methylbut-3-enyl)- flavanone, was isolated from the aerial parts of Derris trifoliate, together with eleven known compounds: rotenone, tephrosin, 12a-hydroxyrotenone, deguelin, 6a,12a-dehydro-rotenone, dehydrodeguelin, 7a-O-methyldeguelol, 7a-O-methylelliptonol, 5,7,3',4'-tetra-hydroxy-6,8-diprenylisoflavone, daidzein and 4'-hydroxy-7-methoxyflavanone. 7a-O-Methylelliptonol was isolated for the first time from the genus Derris. Their structures were characterized on the basis of spectral data. Eight of the isolated compounds were found to be significantly toxic to brine shrimp (LC(50) range 0.06-9.95 μg/mL). The new compound showed weak toxicity (LC(50) = 211.31 μg/mL).

7-Acetoxyhorminone from Salvia multicaulis Vahl. as Promising Inhibitor of 3-Hydroxy-3-methylglutaryl Coenzyme A (HMG-CoA) Reductase.

Project description:3-Hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase is a key enzyme involved in cholesterol biosynthesis and one of the most important targets for the treatment of hypercholesterolemia. A limited number of studies on the HMG-CoA reductase inhibitory potential of natural products are available. Thus, in the current study, we aimed to test the HMG-CoA reductase inhibitory capacity of extracts from the roots and aerial parts of Salvia multicaulis Vahl., through activity-guided isolation. Our findings revealed that the root extract prepared with dichloromethane-acetone (1:1) showed the highest inhibition (71.97 ± 0.37%) at 100 µg/mL. The extract was then initially fractionated by column chromatography and the obtained fractions were monitored by thin layer chromatography. Fractions which were similar to each other were combined and a total of 15 fractions were obtained. Further conventional chromatographic studies were carried out on the active fractions. Based on these fractions, 10 known compounds, comprising 9 terpenes and 1 steroid derivative in total, were isolated and their structures were verified by a combination of IT-TOF-MS, and 1D and 2D NMR techniques. According to the enzyme inhibition data of the identified compounds, 7-acetoxyhorminone exerted the highest inhibition (84.15 ± 0.10%, IC50 = 63.6 ± 1.21 µg/mL). The molecular docking experiments on 7-acetoxyhorminone and horminone indicated that both compounds strongly bind to the active site of the enzyme.

Salvia elegans, Salvia greggii and Salvia officinalis Decoctions: Antioxidant Activities and Inhibition of Carbohydrate and Lipid Metabolic Enzymes.

Project description:Salvia elegans Vahl., Salvia greggii A. Gray, and Salvia officinalis L. decoctions were investigated for their health-benefit properties, in particular with respect to antioxidant activity and inhibitory ability towards key enzymes with impact in diabetes and obesity (α-glucosidase, α-amylase and pancreatic lipase). Additionally, the phenolic profiles of the three decoctions were determined and correlated with the beneficial properties. The S. elegans decoction was the most promising in regard to the antioxidant effects, namely in the scavenging capacity of the free radicals DPPH•, NO• and O₂•⁻, and the ability to reduce Fe3+, as well as the most effective inhibitor of α-glucosidase (EC50 = 36.0 ± 2.7 μg/mL vs. EC50 = 345.3 ± 6.4 μg/mL and 71.2 ± 5.0 μg/mL for S. greggii and S. officinalis, respectively). This superior activity of the S. elegans decoction over those of S. greggii and S. officinalis was, overall, highly correlated with its richness in caffeic acid and derivatives. In turn, the S. officinalis decoction exhibited good inhibitory capacity against xanthine oxidase activity, a fact that could be associated with its high content of flavones, in particular the glycosidic forms of apigenin, scutellarein and luteolin.

Antioxidant and α-glucosidase inhibitory activities of compound isolated from Stachytarpheta jamaicensis (L) Vahl. leaves.

Project description:Stachytarpheta jamaicensis is one of the folk medicines used for the treatment of diabetes in Ambon, Indonesia, but there are limited studies on the bioactivities of its constituents. This study aims to assess the antioxidant and antidiabetic activities of four extracts of S. jamaicensis leaves extracted using several solvents. Bioassay guided fractionation on each extract establishes for exploring S. jamaicensis leaves active compounds. The antioxidant was evaluated using the DPPH and ABTS methods, while the α-glucosidase inhibitory was carried out in vitro assay. The results showed that the methanol extract of S. jamaicensis leaves displays inhibition of DPPH, ABTS and α-glucosidase activity compared to other solvent extracts. Furthermore, 6β-hydroxyipolamiide was successfully isolated from the methanol extract of S. jamicensis leaves which was reported to have α-glucosidase inhibitory activity with an IC50 of 539.17 μg/mL. Based on the results, S. jamaicensis could be recommended as an antioxidant and antidiabetic agent.
