Scattering Assisted Imaging.

ABSTRACT: Standard imaging systems provide a spatial resolution that is ultimately dictated by the numerical aperture (NA) of the illumination and collection optics. In biological tissues, the resolution is strongly affected by scattering, which limits the penetration depth to a few tenths of microns. Here, we exploit the properties of speckle patterns embedded into a strongly scattering matrix to illuminate the sample at high spatial frequency content. Combining adaptive optics with a custom deconvolution algorithm, we obtain an increase in the transverse spatial resolution by a factor of 2.5 with respect to the natural diffraction limit. Our Scattering Assisted Imaging (SAI) provides an effective solution to increase the resolution when long working distance optics are needed, potentially paving the way to bulk imaging in turbid tissues.

Surface-Enhanced Raman Scattering Imaging Assisted by Machine Learning Analysis: Unveiling Pesticide Molecule Permeation in Crop Tissues.

Project description:Surface-enhanced Raman scattering (SERS) imaging technology faces significant technical bottlenecks in ensuring balanced spatial resolution, preventing image bias induced by substrate heterogeneity, accurate quantitative analysis, and substrate preparation that enhances Raman signal strength on a global scale. To systematically solve these problems, artificial intelligence techniques are applied to analyze the signals of pesticides based on 3D and dynamic SERS imaging. Utilizing perovskite/silver nanoparticles composites (CaTiO3/Ag@BONPs) as enhanced substrates, enabling it not only to cleanse pesticide residues from the surface to pulp of fruits and vegetables, but also to investigate the penetration dynamics of an array of pesticides (chlorpyrifos, thiabendazole, thiram, and acetamiprid). The findings challenge existing paradigms, unveiling a previously unnoticed weakening process during pesticide invasion and revealing the surprising permeability of non-systemic pesticides. Of particular note is easy to overlook that the combined application of pesticides can inadvertently intensify their invasive capacity due to pesticide interactions. The innovative study delves into the realm of pesticide penetration, propelling a paradigm shift in the understanding of food safety. Meanwhile, this strategy provides strong support for the cutting-edge application of SERS imaging technology and also brings valuable reference and enlightenment for researchers in related fields.

Local scattering ultrasound imaging.

Project description:Ultrasonic imaging is a widely used tool for detection, localisation and characterisation of material inhomogeneities with important applications in many fields. This task is particularly challenging when imaging in a complex medium, where the ultrasonic wave is scattered by the material microstructure, preventing detection and characterisation of weak targets. Fundamentally, the maximum information that can be experimentally obtained from each material region consists of a set of reflected signals for different incident waves. However, these data are not directly accessible from the raw measurements, which represent a superposition of reflections from all scatterers in the medium. Here we show, that a complete set of transmitter-receiver data encodes sufficient information in order to achieve full spatio-temporal separation of transmitter-receiver data, corresponding to different local scattering areas. We show that access to the local scattering data can provide valuable benefits for many applications. More importantly, this technique enables fundamentally new approaches, exploiting the angular distribution of the scattering amplitude and phase of each local scattering region. Here we demonstrate how the local scattering directivity can be used to build the local scattering image, releasing the full potential and richness of the transmit-receive data. As a proof of concept, we demonstrate the detection of small inclusions in various highly scattering materials using numerical and experimental examples. The described principles are very general and can be applied to any research field where the phased array technology is employed.

Dynamic light scattering imaging.

Project description:We introduce dynamic light scattering imaging (DLSI) to enable the wide-field measurement of the speckle temporal intensity autocorrelation function. DLSI uses the full temporal sampling of speckle fluctuations and a comprehensive model to identify the dynamic scattering regime and obtain a quantitative image of the scatterer dynamics. It reveals errors in the traditional theory of laser Doppler flowmetry and laser speckle contrast imaging and provides guidance on the best model to use in cerebral blood flow imaging.

Tracing multiple scattering trajectories for deep optical imaging in scattering media.

Project description:Multiple light scattering hampers imaging objects in complex scattering media. Approaches used in real practices mainly aim to filter out multiple scattering obscuring the ballistic waves that travel straight through the scattering medium. Here, we propose a method that makes the deterministic use of multiple scattering for microscopic imaging of an object embedded deep within scattering media. The proposed method finds a stack of multiple complex phase plates that generate similar light trajectories as the original scattering medium. By implementing the inverse scattering using the identified phase plates, our method rectifies multiple scattering and amplifies ballistic waves by almost 600 times. This leads to a significant increase in imaging depth-more than three times the scattering mean free path-as well as the correction of image distortions. Our study marks an important milestone in solving the long-standing high-order inverse scattering problems.

All Photons Imaging Through Volumetric Scattering.

Project description:Imaging through thick highly scattering media (sample thickness ≫ mean free path) can realize broad applications in biomedical and industrial imaging as well as remote sensing. Here we propose a computational "All Photons Imaging" (API) framework that utilizes time-resolved measurement for imaging through thick volumetric scattering by using both early arrived (non-scattered) and diffused photons. As opposed to other methods which aim to lock on specific photons (coherent, ballistic, acoustically modulated, etc.), this framework aims to use all of the optical signal. Compared to conventional early photon measurements for imaging through a 15 mm tissue phantom, our method shows a two fold improvement in spatial resolution (4db increase in Peak SNR). This all optical, calibration-free framework enables widefield imaging through thick turbid media, and opens new avenues in non-invasive testing, analysis, and diagnosis.

Observation of laser-assisted electron scattering in superfluid helium.

Project description:Laser-assisted electron scattering (LAES), a light-matter interaction process that facilitates energy transfer between strong light fields and free electrons, has so far been observed only in gas phase. Here we report on the observation of LAES at condensed phase particle densities, for which we create nano-structured systems consisting of a single atom or molecule surrounded by a superfluid He shell of variable thickness (32-340 Å). We observe that free electrons, generated by femtosecond strong-field ionization of the core particle, can gain several tens of photon energies due to multiple LAES processes within the liquid He shell. Supported by Monte Carlo 3D LAES and elastic scattering simulations, these results provide the first insight into the interplay of LAES energy gain/loss and dissipative electron movement in a liquid. Condensed-phase LAES creates new possibilities for space-time studies of solids and for real-time tracing of free electrons in liquids.

Imaging with nature: compressive imaging using a multiply scattering medium.

Project description:The recent theory of compressive sensing leverages upon the structure of signals to acquire them with much fewer measurements than was previously thought necessary, and certainly well below the traditional Nyquist-Shannon sampling rate. However, most implementations developed to take advantage of this framework revolve around controlling the measurements with carefully engineered material or acquisition sequences. Instead, we use the natural randomness of wave propagation through multiply scattering media as an optimal and instantaneous compressive imaging mechanism. Waves reflected from an object are detected after propagation through a well-characterized complex medium. Each local measurement thus contains global information about the object, yielding a purely analog compressive sensing method. We experimentally demonstrate the effectiveness of the proposed approach for optical imaging by using a 300-micrometer thick layer of white paint as the compressive imaging device. Scattering media are thus promising candidates for designing efficient and compact compressive imagers.

Stimulated Brillouin Scattering Microscopic Imaging.

Project description:Two-dimensional stimulated Brillouin scattering microscopy is demonstrated for the first time using low power continuous-wave lasers tunable around 780 nm. Spontaneous Brillouin spectroscopy has much potential for probing viscoelastic properties remotely and non-invasively on a microscopic scale. Nonlinear Brillouin scattering spectroscopy and microscopy may provide a way to tremendously accelerate the data aquisition and improve spatial resolution. This general imaging setup can be easily adapted for specific applications in biology and material science. The low power and optical wavelengths in the water transparency window used in this setup provide a powerful bioimaging technique for probing the mechanical properties of hard and soft tissue.

Imaging scattering orientation with spatial frequency domain imaging.

Project description:Optical imaging techniques based on multiple light scattering generally have poor sensitivity to the orientation and direction of microscopic light scattering structures. In order to address this limitation, we introduce a spatial frequency domain method for imaging contrast from oriented scattering structures by measuring the angular-dependence of structured light reflectance. The measurement is made by projecting sinusoidal patterns of light intensity on a sample, and measuring the degree to which the patterns are blurred as a function of the projection angle. We derive a spatial Fourier domain solution to an anisotropic diffusion model. This solution predicts the effects of bulk scattering orientation on the amplitude and phase of the projected patterns. We introduce a new contrast function based on a scattering orientation index (SOI) which is sensitive to the degree to which light scattering is directionally dependent. We validate the technique using tissue simulating phantoms, and ex vivo samples of muscle and brain. Our results show that SOI is independent of the overall amount of bulk light scattering and absorption, and that isotropic versus oriented scattering structures can be clearly distinguished. We determine the orientation of subsurface microscopic scattering structures located up to 600 μm beneath highly scattering (μ(') (s) = 1.5 mm(-1)) material.
