Fabrication of Hypericin Imprinted Polymer Nanospheres via Thiol-Yne Click Reaction.

ABSTRACT: To fabricate molecularly imprinted polymer nanospheres via click reaction, five different clickable compounds were synthesized and two types of click reactions (azide-alkyne and thiol-yne) were explored. It was found that molecularly imprinted polymer nanospheres could be successfully synthesized via thiol-yne click reaction using 3,5-diethynyl-pyridine (1) as the monomer, tris(3-mercaptopropionate) (tri-thiol, 5) as the crosslinker, and hypericin as the template (MIP?NSHs). The click polymerization completed in merely 4 h to produce the desired MIP?NSHs, which were characterized by FTIR, SEM, DLS, and BET, respectively. The reaction conditions for adsorption capacity and selectivity towards hypericin were optimized, and the MIP?NSHs synthesized under the optimized conditions showed a high adsorption capacity (Q = 6.03 ?mol•g-1) towards hypericin. The imprinting factors of MIP?NSHs towards hypericin, protohypericin, and emodin were 2.44, 2.88, and 2.10, respectively.

Fabrication of Core-Shell Magnetic Molecularly Imprinted Nanospheres towards Hypericin via Click Polymerization.

Project description:The core-shell structure molecularly imprinted magnetic nanospheres towards hypericin (Fe?O?@MIPs) were prepared by mercapto-alkyne click polymerization. The shape and size of nanospheres were characterized by dynamic light scattering (DLS) and transmission electron microscope (TEM). The nanospheres were analyzed by FTIR spectroscopy to verify the thiol-yne click reaction in the presence or absence of hypericin. The Brunauer?Emmet?Teller (BET) method was used for measuring the average pore size, pore volume and surface area. The Fe?O?@MIPs synthesized displayed a good adsorption capacity (Q = 6.80 µmol·g-1). In addition, so-prepared Fe?O?@MIPs showed fast mass transfer rates and good reusability. The method established for fabrication of Fe?O?@MIPs showed excellent reproducibility and has broad potential for the fabrication of other core-shell molecularly imprinted polymers (MIPs).

Visible light mediated metal-free thiol-yne click reaction.

Project description:The carbon-sulfur bond formation reaction is of paramount importance for functionalized materials design, as well as for biochemical applications. The use of expensive metal-based catalysts and the consequent contamination with trace metal impurities are challenging drawbacks of the existing methodologies. Here, we describe the first environmentally friendly metal-free photoredox pathway to the thiol-yne click reaction. Using Eosin Y as a cheap and readily available catalyst, C-S coupling products were obtained in high yields (up to 91%) and excellent selectivity (up to 60 : 1). A 3D-printed photoreactor was developed to create arrays of parallel reactions with temperature stabilization to improve the performance of the catalytic system.

Thiol-yne click reaction: an interesting way to derive thiol-provided catechols.

Project description:The hydrothiolation of activated alkynes is presented as an attractive and powerful way to functionalize thiols bearing catechols. The reaction was promoted by a heterogeneous catalyst composed of copper nanoparticles supported on TiO2 (CuNPs/TiO2) in 1,2-dichloroethane (1,2-DCE) under heating at 80 °C. The catalyst could be recovered and reused in three consecutive cycles, showing a slight decrease in its catalytic activity. Thiol derivatives bearing catechol moieties, obtained through a versatile Michael addition, were reacted with different activated alkynes, such as methyl propiolate, propiolic acid, propiolamide or 2-ethynylpyridine. The reaction was shown to be regio- and stereoselective towards anti-Markovnikov Z-vinyl sulfide in most cases studied. Finally, some catechol derivatives obtained were tested as ligands in the preparation of coordination polymer nanoparticles (CNPs), by taking the advantage of their different coordination sites with metals such as iron and cobalt.

ESI-MS Analysis of Thiol-yne Click Reaction in Petroleum Medium.

Project description:Petroleum contains a large number of heteroatomic compounds, but today, most of them are not efficiently utilized. The constant development of the sustainability concept recalls for rethinking the usage of fossil resources with improved chemical utility. In order to initiate research aimed at involving active petroleum compounds in chemical transformations, a new analytical method for product detection is needed. Here, we study the click reaction of thiols with alkynes, leading to the formation of α-vinyl sulfides directly in the petroleum environment. The reaction was carried out using an (IMes)Pd(acac)Cl catalyst, which demonstrated tolerance to petroleum components. In this study, the concentration of thiols ranged from 1 M to 0.01 M (from 8% to 0.1%). To detect products at low concentrations, a special alkyne labeled with an imidazole moiety was used. This approach made it possible to observe the formation of vinyl sulfides by electrospray ionization mass spectrometry (ESI-MS), which provides an opportunity for further optimization of the reaction conditions and future developments for the direct involvement of oil components in chemical reactions.

A Facile Approach for Fabrication of Core-Shell Magnetic Molecularly Imprinted Nanospheres towards Hypericin.

Project description:By taking advantage of the self-polymerization of dopamine on the surface of magnetic nanospheres in weak alkaline Tris-HCl buffer solution, a facile approach was established to fabricate core-shell magnetic molecularly imprinted nanospheres towards hypericin (Fe₃O₄@PDA/Hyp NSs), via a surface molecular imprinting technique. The Fe₃O₄@PDA/Hyp NSs were characterized by FTIR, TEM, DLS, and BET methods, respectively. The reaction conditions for adsorption capacity and selectivity towards hypericin were optimized, and the Fe₃O₄@PDA/Hyp NSs synthesized under the optimized conditions showed a high adsorption capacity (Q = 18.28 mg/g) towards hypericin. The selectivity factors of Fe₃O₄@PDA/Hyp NSs were about 1.92 and 3.55 towards protohypericin and emodin, respectively. In addition, the approach established in this work showed good reproducibility for fabrication of Fe₃O₄@PDA/Hyp.

Thiol-Yne click chemistry of acetylene-enabled macrocyclization.

Project description:Macrocycles have fascinated scientists for over half a century due to their aesthetically appealing structures and broad utilities in chemical, material, and biological research. However, the efficient preparation of macrocycles remains an ongoing research challenge in organic synthesis because of the high entropic penalty involved in the ring-closing process. Herein we report a photocatalyzed thiol-yne click reaction to forge diverse sulfur-containing macrocycles (up to 35-membered ring) and linear C2-linked 1,2-(S-S/S-P/S-N) functionalized molecules, starting from the simplest alkyne, acetylene. Preliminary mechanistic experiments support a visible light-mediated radical-polar crossover dihydrothiolation process. This operationally straightforward reaction is also amenable to the synthesis of organometallic complexes, bis-sulfoxide ligand and a pleuromutilin antibiotic drug Tiamulin, which provides a practical route to synthesize highly valued compounds from the feedstock acetylene gas.

Sunlight-Mediated Thiol-Ene/Yne Click Reaction: Synthesis and DNA Transfection Efficiency of New Cationic Lipids.

Project description:The design of green synthetic reaction conditions is very challenging, especially for biomaterials, but worthwhile if the compounds can be easily synthesized in the aqueous medium. Herein, we report the development of sunlight-mediated thiol-ene/yne click reaction in the presence of a catalytic amount of tert-butyl hydroperoxide (TBHP) in an aqueous medium. The optimized reaction conditions were successfully applied to synthesize a series of small molecules and lipids in a single step in the aqueous medium. The synthetic cationic lipid/co-lipid formed positively charged stable nanosized liposomes that effectually bind with the genetic materials. The in vitro DNA transfection and cellular uptake assays showed that the synthesized cationic lipids have comparable efficiency to commercially available Lipofectamine 2000. This mild synthetic strategy can also be used for smart design of novel or improvement of prevailing lipid-based nonviral gene delivery systems. Such chemical transformations in the aqueous medium are more environment-friendly than other reported thiol-ene/yne click reactions performed in an organic solvent medium.

Fabrication and characterization of thiol-triacrylate polymer via Michael addition reaction for biomedical applications.

Project description:Thiol-acrylate polymers have therapeutic potential as biocompatible scaffolds for bone tissue regeneration. Synthesis of a novel cyto-compatible and biodegradable polymer composed of trimethylolpropane ethoxylate triacrylate-trimethylolpropane tris (3-mercaptopropionate) (TMPeTA-TMPTMP) using a simple amine-catalyzed Michael addition reaction is reported in this study. This study explores the impact of molecular weight and crosslink density on the cyto-compatibility of human adipose derived mesenchymal stem cells. Eight groups were prepared with two different average molecular weights of trimethylolpropane ethoxylate triacrylate (TMPeTA 692 and 912) and four different concentrations of diethylamine (DEA) as catalyst. The materials were physically characterized by mechanical testing, wettability, mass loss, protein adsorption and surface topography. Cyto-compatibility of the polymeric substrates was evaluated by LIVE/DEAD staining® and DNA content assay of cultured human adipose derived stem cells (hASCs) on the samples over over days. Surface topography studies revealed that TMPeTA (692) samples have island pattern features whereas TMPeTA (912) polymers showed pitted surfaces. Water contact angle results showed a significant difference between TMPeTA (692) and TMPeTA (912) monomers with the same DEA concentration. Decreased protein adsorption was observed on TMPeTA (912) -16% DEA compared to other groups. Fluorescent microscopy also showed distinct hASCs attachment behavior between TMPeTA (692) and TMPeTA (912), which is due to their different surface topography, protein adsorption and wettability. Our finding suggested that this thiol-acrylate based polymer is a versatile, cyto-compatible material for tissue engineering applications with tunable cell attachment property based on surface characteristics.

Fabrication of Photoluminescent Quantum Dot Thiol-yne Nanocomposites via Thermal Curing or Photopolymerization.

Project description:Strong, flexible, and transparent materials have garnered tremendous interest in recent years as materials and electronics manufacturers pursue devices that are bright, flexible, durable, tailorable, and lightweight. Depending on the starting components, polymers fabricated using thiol-yne chemistry have been shown to be exceptionally strong and/or flexible, while also being amenable to modification by the incorporation of nanoparticles. In the present work, novel ligands were synthesized and used to functionalize quantum dots (QDs) of various diameters. The functionalized QDs were then incorporated into thiol-yne prepolymer matrices. These matrices were subsequently polymerized to form QD thiol-yne nanocomposite polymers. To demonstrate the versatility of the fabrication process, the prepolymers were either thermally cured or photopolymerized. The resulting transparent nanocomposites expressed the size-specific color of the QDs within them when exposed to ultraviolet irradiation, demonstrating that QDs can be incorporated into thiol-yne polymers without significantly altering QD expression. With the inclusion of QDs, thiol-yne nanocomposite polymers are promising candidates for use in numerous applications including as device display materials, optical lens materials, and/or sensor materials.

Using Stereochemistry to Control Mechanical Properties in Thiol-Yne Click-Hydrogels.

Project description:The stereochemistry of polymers has a profound impact on their mechanical properties. While this has been observed in thermoplastics, studies on how stereochemistry affects the bulk properties of swollen networks, such as hydrogels, are limited. Typically, changing the stiffness of a hydrogel is achieved at the cost of changing another parameter, that in turn affects the physical properties of the material and ultimately influences the cellular response. Herein, we report that by manipulating the stereochemistry of a double bond, formed in situ during gelation, materials with diverse mechanical properties but comparable physical properties can be obtained. Click-hydrogels that possess a high % trans content are stiffer than their high % cis analogues by almost a factor of 3. Human mesenchymal stem cells acted as a substrate stiffness cell reporter demonstrating the potential of these platforms to study mechanotransduction without the influence of other external factors.
