Construction of All-in-Focus Images Assisted by Depth Sensing.

ABSTRACT: Multi-focus image fusion is a technique for obtaining an all-in-focus image in which all objects are in focus to extend the limited depth of field (DoF) of an imaging system. Different from traditional RGB-based methods, this paper presents a new multi-focus image fusion method assisted by depth sensing. In this work, a depth sensor is used together with a colour camera to capture images of a scene. A graph-based segmentation algorithm is used to segment the depth map from the depth sensor, and the segmented regions are used to guide a focus algorithm to locate in-focus image blocks from among multi-focus source images to construct the reference all-in-focus image. Five test scenes and six evaluation metrics were used to compare the proposed method and representative state-of-the-art algorithms. Experimental results quantitatively demonstrate that this method outperforms existing methods in both speed and quality (in terms of comprehensive fusion metrics). The generated images can potentially be used as reference all-in-focus images.

Machine learning assisted construction of a shallow depth dynamic ansatz for noisy quantum hardware.

Project description:The development of various dynamic ansatz-constructing techniques has ushered in a new era, making the practical exploitation of Noisy Intermediate-Scale Quantum (NISQ) hardware for molecular simulations increasingly viable. However, such ansatz construction protocols incur substantial measurement costs during their execution. This work involves the development of a novel protocol that capitalizes on regenerative machine learning methodologies and many-body perturbation theoretical measures to construct a highly expressive and shallow ansatz within the variational quantum eigensolver (VQE) framework with limited measurement costs. The regenerative machine learning model used in our work is trained with the basis vectors of a low-rank expansion of the N-electron Hilbert space to identify the dominant high-rank excited determinants without requiring a large number of quantum measurements. These selected excited determinants are iteratively incorporated within the ansatz through their low-rank decomposition. The reduction in the number of quantum measurements and ansatz depth manifests in the robustness of our method towards hardware noise, as demonstrated through numerical applications. Furthermore, the proposed method is highly compatible with state-of-the-art neural error mitigation techniques. This resource-efficient approach is quintessential for determining spectroscopic and other molecular properties, thereby facilitating the study of emerging chemical phenomena in the near-term quantum computing framework.

Construction of anisotropic fluorescent nanofibers assisted by electro-spinning and its optical sensing applications.

Project description:Fixing the gap between "nano-scaled" pieces and "product-scale" materials, devices or machines is an ineluctable challenge that people have to tackle. Herein, we show that combining self-assembly and electrospinning processes results in the fabrication of anisotropic fluorescent nanofibers (PDI@PVDF) in which the well-defined rod-like perylene bisimide derivative assemblies are embedded in a highly oriented way along the axis of the poly(vinylidene fluoride) (PVDF) fiber. Compared to fragile individual PDI assemblies, the electrospinning anisotropic fluorescent PDI@PVDF nanofibers not only maintain high sensitivity for aniline vapour but also exhibit an unexpected short response time for both quenching and recovering. The results demonstrate that electrospinning assistance is a versatile and effective strategy to maintain the anisotropy of fluorescent nanomaterials, building a bridge between self-assembled nano-rods and practical materials.

Extended depth of focus for coherence-based cellular imaging.

Project description:Improving lateral resolution for cross-sectional optical coherence tomography (OCT) imaging is difficult due to the rapid divergence of light once it is focused to a small spot. To overcome this obstacle, we introduce a fiber optics system that generates a coaxially focused multimode (CAFM) beam for depth of focus (DOF) extension. We fabricated a CAFM beam OCT probe and show that the DOF is more than fivefold that of a conventional Gaussian beam, enabling cross-sectional imaging of biological tissues with clearly resolved cellular and subcellular structures over more than a 400 ?m depth range. The compact and straightforward design and high-resolution extended DOF imaging capabilities of this technique suggests that it will be very useful for endoscopic cross-sectional imaging of human internal organs in vivo.

50 Hz volumetric functional imaging with continuously adjustable depth of focus.

Project description:Understanding how neural circuits control behavior requires monitoring a large population of neurons with high spatial resolution and volume rate. Here we report an axicon-based Bessel beam module with continuously adjustable depth of focus (CADoF), that turns frame rate into volume rate by extending the excitation focus in the axial direction while maintaining high lateral resolutions. Cost-effective and compact, this CADoF Bessel module can be easily integrated into existing two-photon fluorescence microscopes. Simply translating one of the relay lenses along its optical axis enabled continuous adjustment of the axial length of the Bessel focus. We used this module to simultaneously monitor activity of spinal projection neurons extending over 60 µm depth in larval zebrafish at 50 Hz volume rate with adjustable axial extent of the imaged volume.

Modulating T Cell Activation Using Depth Sensing Topographic Cues.

Project description:This report examines how sensing of substrate topography can be used to modulate T cell activation, a key coordinating step in the adaptive immune response. Inspired by the native T cell-antigen presenting cell interface, micrometer scale pits with varying depth are fabricated into planar substrates. Primary CD4+ T cells extend actin-rich protrusions into the micropits. T cell activation, reflected in secretion of cytokines interleukin-2 and interferon gamma, is sensitive to the micropit depth. Surprisingly, arrays of micropits with 4 μm depth enhance activation compared to flat substrates but deeper micropits are less effective at increasing cell response, revealing a biphasic dependence in activation as a function of feature dimensions. Inhibition of cell contractility abrogates the enhanced activation associated with the micropits. In conclusion, this report demonstrates that the 3D, microscale topography can be used to enhance T cell activation, an ability that most directly can be used to improve production of these cells for immunotherapy.

Recovering Depth from Still Images for Underwater Dehazing Using Deep Learning.

Project description:Estimating depth from a single image is a challenging problem, but it is also interesting due to the large amount of applications, such as underwater image dehazing. In this paper, a new perspective is provided; by taking advantage of the underwater haze that may provide a strong cue to the depth of the scene, a neural network can be used to estimate it. Using this approach the depthmap can be used in a dehazing method to enhance the image and recover original colors, offering a better input to image recognition algorithms and, thus, improving the robot performance during vision-based tasks such as object detection and characterization of the seafloor. Experiments are conducted on different datasets that cover a wide variety of textures and conditions, while using a dense stereo depthmap as ground truth for training, validation and testing. The results show that the neural network outperforms other alternatives, such as the dark channel prior methods and it is able to accurately estimate depth from a single image after a training stage with depth information.

Depth correction of 3D NanoSIMS images using secondary electron pixel intensities.

Project description:Strategies that do not require additional characterization to be performed on the sample or the collection of additional secondary ion signals are needed to depth correct 3D SIMS images of cells. Here, we develop a depth correction strategy that uses the pixel intensities in the secondary electron images acquired during negative-ion NanoSIMS depth profiling to reconstruct the sample morphology. This morphology reconstruction was then used to depth correct the 3D SIMS images that show the components of interest in the sample. As a proof of concept, we applied this approach to NanoSIMS depth profiling data that show the 15N-enrichment and 18O-enrichment from 15N-sphingolipids and 18O-cholesterol, respectively, within a metabolically labeled Madin-Darby canine kidney cell. Comparison of the cell morphology reconstruction to the secondary electron images collected with the NanoSIMS revealed that the assumption of a constant sputter rate produced small inaccuracies in sample morphology after approximately 0.66 μm of material was sputtered from the cell. Nonetheless, the resulting 3D renderings of the lipid-specific isotope enrichments better matched the shapes and positions of the subcellular compartments that contained 15N-sphingolipids and 18O-cholesterol than the uncorrected 3D SIMS images. This depth correction of the 3D SIMS images also facilitated the detection of spherical cholesterol-rich compartments that were surrounded by membranes containing cholesterol and sphingolipids. Thus, we expect this approach will facilitate identifying the subcellular structures that are enriched with biomolecules of interest in 3D SIMS images while eliminating the need for correlated analyses or additional secondary ion signals for the depth correction of 3D NanoSIMS images.

Light assisted antibody immobilization for bio-sensing.

Project description:Ultrashort UV pulses at 258 nm with repetition rate of 10 kHz have been used to irradiate buffer solution of antibody. The tryptophan residues strongly absorb this radiation thus becoming capable to disrupt the disulfide bridges located next to them. Due to their high reactivity the opened bridges can anchor a gold plate more efficiently than other sites of the macromolecule giving rise to preferential orientations of the variable part of the antibody. UV irradiation has been applied to anchor antiIgG antibody to the electrode of a Quartz Crystal Microbalance (QCM) that lends itself as a sensor, the antibody acting as the bio-receptor. An increase of the QCM sensitivity and of the linear range has been measured when the antibody is irradiated with UV laser pulses. The photo-induced reactions leading to disulfide bridge breakage have been analyzed by means of a chemical assay that confirms our explanation. The control of disulfide bridges by UV light paves the way to important applications for sensing purpose since cysteine in combination with tryptophan can act as a hook to link refractory bio-receptors to surfaces.
