Increased Autophagy of Rice Can Increase Yield and Nitrogen Use Efficiency (NUE).

ABSTRACT: Autophagy (self-eating), a conserved pathway in eukaryotes, which is designed to handle cytoplasmic material in bulk and plays an important role in the remobilization of nutrient, such as nitrogen (N) under suboptimal nutrient conditions. Here, we identified a core component of an autophagy gene in rice (Oryza sativa), OsATG8a, with increased expression levels under N starvation conditions. Overexpression of OsATG8a significantly enhanced the level of autophagy and the number of effective tillers in the transgenic rice. In addition, the transgenic lines accumulated more N in grains than in the dry remains and the yield was significantly increased under normal N conditions. Further N allocation studies revealed that the nitrogen uptake efficiency (NUpE) and nitrogen use efficiency (NUE) significantly increased. Otherwise, under suboptimal N conditions, overexpression of OsATG8a did not seem to have any effect on yield and NUE, but NUpE was still improved significantly. Based on our findings, we consider OsATG8a to be a great candidate gene to increase NUE and yield.

Project description:Reasonable nitrogen (N) and potassium (K) application rates can effectively improve fertilizer use efficiency, rice yield and quality. A two-year field experiment was conducted with combined application of three N rates (135, 180, and 225 kg ha-1, denoted as N1-N3) and four K rates (0, 90, 135, and 180 kg ha-1, denoted as K0-K3) using super indica hybrid rice cultivar Yixiangyou (YXY) 2115 to explore the effects of co-application of N and K on rice growth and development. The results indicated that the combined application of N and K had significantly interactive effects on dry matter (DM) accumulation, nutrients absorption, N harvest index (NHI), K harvest index (KHI), spikelets per panicle and most rice quality indexes. The highest total DM accumulation (17998.17-19432.47 kg ha-1) at maturity stage was obtained under N3K2. The effect of co-application of N and K on nutrients absorption and utilization varied between the two years and within each year. The highest total N and K accumulations at maturity stage were observed under N3K1 and N3K2, respectively, while the highest N recovery efficiency (NRE) and K recovery efficiency (KRE) were observed under N1K3. High expression levels of N and K metabolism-related genes in rice grains were observed under N3K2 or N3K3, consistent with N and K uptake. Co-application of N and K increased rice yield significantly and the highest yield (6745.02-7010.27 kg ha-1) was obtained under N2K2. As more N was gradually applied, rice appearance quality improved but milling, cooking and eating quality decreased. Although appropriate application of K could improve rice milling, cooking and eating quality, it reduced appearance quality. The optimal milling, cooking and eating quality were obtained under N1K2, while the best appearance quality was obtained under N3K0. Overall, a combination of 135-210 kg ha-1 N and 115-137 kg ha-1 K application rates was recommended for achieving relatively higher yield and better quality in rice production.

Project description:Overuse of nitrogen (N) fertilizer has led to low N use efficiency (NUE) and high N loss in single rice cropping systems in southeast China. Application of controlled-release urea (CRU) is considered as an effective N fertilizer practice for improving crop yields and NUE. Here, field experiments were conducted during 2015-2017 to assess the effects of two CRUs (resin-coated urea (RCU) and polyurethane-coated urea (PCU)) on rice yields, NUE and soil fertility at two sites (Lincheng town (LC) and Xintang town (XT)). Four treatments were established at each site: (1) control with no N application (CK), (2) split application of conventional urea (U, 270?kg?N ha-1), (3) single basal application of RCU (RCU, 216?kg?N ha-1), and (4) single basal application of PCU (PCU, 216?kg?N ha-1). The N application rate in the CRU treatment compared to the U treatment was reduced by 20%. However, the results showed that, compared to split application of urea, single basal application of CRU led to similar rice grain yields and aboveground biomass at both sites. No significant difference in the N uptake by rice plant was observed between the U and CRU treatments at either site. There were no significant differences in the N apparent recovery efficiency (NARE) among the U, RCU and PCU treatments, with the exception of that in XT in 2015. Compared to application of U, application of CRU increased the N agronomic efficiency (NAE) and N partial factor productivity (NPFP) by 17.4-52.6% and 23.4-29.8% at the LC site, and 15.0-84.1% and 23.2-33.4% at the XT site, respectively, during 2015-2017. Yield component analysis revealed that greater rice grain yield in response to N fertilizer was attributed mainly to the number of panicles per m2, which increased in the fertilized treatments compared to the CK treatment. The application of CRU did not affect the soil fertility after rice harvest in 2016. Overall, these results suggest that single basal application of CRU constitutes a promising alternative N management practice for reducing N application rates, time- and labor-consuming in rice production in southeast China.
