Project description:The developmental functions of miR156-SPL regulatory network have been extensively studied in Arabidopsis, but the downstream genes regulated by each SPL have not been well characterized. In this study, Next Generation Sequencing-based transcriptome analysis was performed on roots of wild type (WT) and miR156 overexpression (miR156OE) plants. One of the SPL genes, SPL10, which represses lateral root growth in Arabidopsis, was significantly downregulated in miR156OE plants. A transcription factor, AGAMOUS-like MADS box protein 79 (AGL79), was also significantly downregulated in the miR156OE plants, but was upregulated in the SPL10 overexpression (SPL10OE) Arabidopsis plants. In addition, SPL10 was found to bind to the core consensus SPL binding sequences in AGL79 gene. Moreover, analyses of complementation lines revealed a linear relationship between SPL10 and AGL79 in regulating Arabidopsis plant development. In addition, it was observed that plant phenotypes are AGL79 dose-dependent, with higher expression causing narrow leaf shape, less number of leaves and early flowering time, whereas relatively lower AGL79 overexpression produce plants with more rosette leaves and more lateral branches. Our findings revealed direct binding of SPL10 to AGL79 promoter, which further suggests a role for miR156/SPL10 module in plant lateral root growth by directly regulating AGL79.

Project description:Background and aimsRoot diameter, especially apical diameter, plays an important role in root development and function. The variation in diameter between roots, and along roots, affects root structure and thus the root system's overall foraging performance. However, the effect of diameter variation on root elongation, branching and topological connections has not been examined systematically in a population of high-order roots, nor along the roots, especially for mature plants grown in the field.MethodsA method combining both excavation and analysis was applied to extract and quantify root architectural traits of adult, field-grown maize plants. The relationships between root diameter and other root architectural characteristics are analysed for two maize cultivars.Key resultsThe basal diameter of the lateral roots (orders 1-3) was highly variable. Basal diameter was partly determined by the diameter of the bearing segment. Basal diameter defined a potential root length, but the lengths of most roots fell far short of this. This was explained partly by differences in the pattern of diameter change along roots. Diameter tended to decrease along most roots, with the steepness of the gradient of decrease depending on basal diameter. The longest roots were those that maintained (or sometimes increased) their diameters during elongation. The branching density (cm(-1)) of laterals was also determined by the diameter of the bearing segment. However, the location of this bearing segment along the mother root was also involved - intermediate positions were associated with higher densities of laterals.ConclusionsThe method used here allows us to obtain very detailed records of the geometry and topology of a complex root system. Basal diameter and the pattern of diameter change along a root were associated with its final length. These relationships are especially useful in simulations of root elongation and branching in source-sink models.

Project description:The development of crops with better growth under suboptimal phosphorus availability would improve food security in developing countries while reducing environmental pollution in developed countries. We tested the hypothesis that maize (Zea mays) phenotypes with greater lateral root branching density have greater phosphorus acquisition from low phosphorus soils. Recombinant inbred lines with either 'many short' (MS) or 'few long' (FL) lateral root phenotypes were grown under high and low phosphorus conditions in greenhouse mesocosms and in the field. Under low phosphorus in mesocosms, lines with the MS phenotype had 89% greater phosphorus acquisition and 48% more shoot biomass than FL lines. Under low phosphorus in the field, MS lines had 16% shallower rooting depth (D95), 81% greater root length density in the top 20 cm of the soil, 49% greater shoot phosphorus content, 12% greater leaf photosynthesis, 19% greater shoot biomass, and 14% greater grain yield than FL lines. These results are consistent with the hypothesis that the phenotype of many, shorter lateral roots improves phosphorus acquisition under low phosphorus availability and merits consideration for genetic improvement of phosphorus efficiency in maize and other crops.

Project description:Gibberellins (GAs) are phytohormones that regulate a wide range of developmental processes in plants. Levels of active GAs are regulated by biosynthetic and catabolic enzymes like the GA 2-oxidases (GA2oxs). In tomato (Solanum lycopersicum L.) C19 GA2oxs are encoded by a small multigenic family of five members with some degree of redundancy. In order to investigate their roles in tomato, the silencing of all five genes in transgenic plants was induced. A significant increase in active GA4 content was found in the ovaries of transgenic plants. In addition, the transgenic unfertilized ovaries were much bigger than wild-type ovaries (about 30 times) and a certain proportion (5-37%) were able to develop parthenocarpically. Among the GA2ox family, genes GA2ox1 and -2 seem to be the most relevant for this phenotype since their expression was induced in unfertilized ovaries and repressed in developing fruits, inversely correlating with ovary growth. Interestingly, transgenic lines exhibited a significant inhibition of branching and a higher content of active GA4 in axillary buds. This phenotype was reverted, in transgenic plants, by the application of paclobutrazol, a GA biosynthesis inhibitor, suggesting a role for GAs as repressors of branching. In summary, this work demonstrates that GA 2-oxidases regulate gibberellin levels in ovaries and axillary buds of tomato plants and their silencing is responsible for parthenocarpic fruit growth and branching inhibition.

Project description:Suboptimal nitrogen (N) availability is a primary constraint for crop production in developing countries, while in developed countries, intensive N fertilization is a primary economic, energy, and environmental cost for crop production. We tested the hypothesis that under low-N conditions, maize (Zea mays) lines with few but long (FL) lateral roots would have greater axial root elongation, deeper rooting, and greater N acquisition than lines with many but short (MS) lateral roots. Maize recombinant inbred lines contrasting in lateral root number and length were grown with adequate and suboptimal N in greenhouse mesocosms and in the field in the USA and South Africa (SA). In low-N mesocosms, the FL phenotype had substantially reduced root respiration and greater rooting depth than the MS phenotype. In low-N fields in the USA and SA, the FL phenotype had greater rooting depth, shoot N content, leaf photosynthesis, and shoot biomass than the MS phenotype. The FL phenotype yielded 31.5% more than the MS phenotype under low N in the USA. Our results are consistent with the hypothesis that sparse but long lateral roots improve N capture from low-N soils. These results with maize probably pertain to other species. The FL lateral root phenotype merits consideration as a selection target for greater crop N efficiency.

Project description:Salinity stress constrains lateral root (LR) growth and severely affects plant growth. Auxin signaling regulates LR formation, but the molecular mechanism by which salinity affects root auxin signaling and whether salt induces other pathways that regulate LR development remains unknown. In Arabidopsis thaliana, the auxin-regulated transcription factor LATERAL ORGAN BOUNDARY DOMAIN 16 (LBD16) is an essential player in LR development under control conditions. Here, we show that under high-salt conditions, an alternative pathway regulates LBD16 expression. Salt represses auxin signaling but, in parallel, activates ZINC FINGER OF ARABIDOPSIS THALIANA 6 (ZAT6), a transcriptional activator of LBD16. ZAT6 activates LBD16 expression, thus contributing to downstream cell wall remodeling and promoting LR development under high-salt conditions. Our study thus shows that the integration of auxin-dependent repressive and salt-activated auxin-independent pathways converging on LBD16 modulates root branching under high-salt conditions.

Project description:Analysis of different leaves from transgenic plants over expressing microRNA miR156. Time-dependent gene expression is essential for developmental timing control. MiR156 was characterized as a time-dependent expressed microRNA during leaf development. The level of miR156 is positive correlated to developmental time during leaf growth. We used microarray to identify genes regulated by miR156 in a time-dependent manner during leaf development. Results indicate that most time-negative genes were activated in mature leaves of miR156 over expressed plants compared with wild type. Keywords: gene expression array-based (RNA / in situ oligonucleotide) To identify the temporal regulation role of miR156 in different leaves, all leaves in the main tiller were collected separately from Md/Mh (miR156 over expressed lines) and wild type (WT) plants. The level of miR156 were checked in these samples, then three biological repeats were selected and these RNA samples of the oldest (L1) and youngest leaf (L4 or L5) were applied for microarray hybridization.

Project description:Plant forms display a wide variety of architectures, depending on the number of lateral branches, internode elongation and phyllotaxy. These are in turn determined by the number, the position and the fate of the Axillary Meristems (AMs). Mutants that affect AM determination during the vegetative phase have been isolated in several model plants. Among these genes, the GRAS transcription factor LATERAL SUPPRESSOR (Ls) plays a pivotal role in AM determination during the vegetative phase. Hereby we characterize the phylogenetic orthologue of Ls in Antirrhinum, ERAMOSA (ERA). Our data supported ERA control of AM formation during both the vegetative and the reproductive phase in snapdragon. A phylogenetic analysis combined with an analysis of the synteny of Ls in several species strongly supported the hypothesis that ERA is a phylogenetic orthologue of Ls, although it plays a broader role. During the reproductive phase ERA promotes the establishment of the stem niche at the bract axis but, after the reproductive transition, it is antagonized by the MADS box transcription factor SQUAMOSA (SQUA). Surprisingly double mutant era squa plants display a squa phenotype developing axillary meristems, which can eventually turn into inflorescences or flowers.

Project description:Organ size is a major agronomic trait that determines grain yield and biomass production in crops. However, the molecular mechanisms controlling organ size, especially in legumes, are poorly understood. Using forward genetic approaches in a Tnt1 insertion mutant population of the model legume Medicago truncatula, we identified SMALL LEAF AND BUSHY1 (SLB1), which is required for the control of organ size and lateral branching. Loss of function of SLB1 led to reduced leaf and flower size but increased lateral branch formation in M. truncatula. SLB1 encodes an F-box protein, an orthologue of Arabidopsis thaliana STERILE APETALA (SAP), that forms part of an SKP1/Cullin/F-box E3 ubiquitin ligase complex. Biochemical and genetic analyses revealed that SLB1 controls M. truncatula organ growth and lateral branching by modulating the stability of BIG SEEDS1 (BS1). Moreover, the overexpression of SLB1 increased seed and leaf size in both M. truncatula and soybean (Glycine max), indicating functional conservation. Our findings revealed a novel mechanism by which SLB1 targets BS1 for degradation to regulate M. truncatula organ size and shoot branching, providing a new genetic tool for increasing seed yield and biomass production in crop and forage legumes.
