High precision computer-generated moire profilometry.

ABSTRACT: Recently, a computer-generated moiré profilometry was proposed by our research group. It can effectively avoid the influence of the transient caused by moiré fringes' direct acquisition and generally owns a higher accuracy. But when the spatial spectrum of the captured deformed pattern is severely aliased caused by the measured object, the accuracy of this method may be affected to some extent due to the impure background light component extraction. So, a high precision computer-generated moiré profilometry based on background light component's accurate elimination is proposed. By adding an additional special phase-shifting sinusoidal grating to accurately extract valid information in the spatial domain and improve the sinusoidal feature of the pattern, the measurement precision can be improved effectively. Though the single-shot feature is broken, the real-time measuring feature is still maintained successfully. Experimental results show the feasibility and validity of the proposed method.

Color-encoded single-shot computer-generated Moire profilometry.

Project description:A color-encoded single-shot computer-generated Moiré profilometry (CSCGMP) is proposed. Two sinusoidal gratings with a π phase difference are encoded in red and blue channels respectively to combine a composite color grating. While this composite color grating is projected onto the measured object, the corresponding color deformed pattern can be captured. So two deformed patterns with a π phase difference are separated from its red and blue components respectively. After normalization and subtraction, the AC component of both separated deformed patterns can be extracted. If this AC component respectively multiplied by the two AC components of fringe patterns of reference plane with a π/2 phase difference prepared and saved on the computer in advance, two computer-generated Moiré fringes just respectively standing for sine and cosine of phase which is modulated by the height of the object relative to the reference plane are figured out. So the 3D shape of the measured object can be reconstructed with normal computer-generated Moiré profilometry. Both simulation and experimental results show the feasibility and validity of the proposed method. It has potential in real-time 3D measurement due to its single-shot feature.

A super-grayscale and real-time computer-generated Moire profilometry using video grating projection.

Project description:By using the time-division multiplexing characteristics of the projector and the integral exposure characteristics of the charge coupled device (CCD) camera, a super-grayscale and real-time computer-generated Moiré profilometry based on video grating projection is proposed. The traditional digital static grating is of 256-grayscale at most. If an expected super-grayscale grating with a maximum grayscale of 766 is designed and divided into three 256-grayscale fringe patterns with balanced grayscale as far as possible, they can be synthesized into a repeated playing video grating instead of the traditional static grating. When the video grating is projected onto the measured object, as long as the exposure time is set to three times the refresh cycle of the video grating, the super-grayscale deformed patterns in the 766-grayscale can be captured with a 10-bit CCD camera, so that the deformed patterns are realistic. The digital error in computer-generated Moiré profilometry is effectively reduced. In addition, this method can expand the linear range of the deformed pattern by 20% in computer Moiré profilometry. Therefore, the proposed method has the perspectives of high accuracy and real-time measurement. Theoretical analysis and experimental results demonstrate the validity and capability of the proposed method.

Computer Generated Holography with Intensity-Graded Patterns.

Project description:Computer Generated Holography achieves patterned illumination at the sample plane through phase modulation of the laser beam at the objective back aperture. This is obtained by using liquid crystal-based spatial light modulators (LC-SLMs), which modulate the spatial phase of the incident laser beam. A variety of algorithms is employed to calculate the phase modulation masks addressed to the LC-SLM. These algorithms range from simple gratings-and-lenses to generate multiple diffraction-limited spots, to iterative Fourier-transform algorithms capable of generating arbitrary illumination shapes perfectly tailored on the base of the target contour. Applications for holographic light patterning include multi-trap optical tweezers, patterned voltage imaging and optical control of neuronal excitation using uncaging or optogenetics. These past implementations of computer generated holography used binary input profile to generate binary light distribution at the sample plane. Here we demonstrate that using graded input sources, enables generating intensity graded light patterns and extend the range of application of holographic light illumination. At first, we use intensity-graded holograms to compensate for LC-SLM position dependent diffraction efficiency or sample fluorescence inhomogeneity. Finally we show that intensity-graded holography can be used to equalize photo evoked currents from cells expressing different levels of chanelrhodopsin2 (ChR2), one of the most commonly used optogenetics light gated channels, taking into account the non-linear dependence of channel opening on incident light.

Computer-generated ovaries to assist follicle counting experiments.

Project description:Precise estimation of the number of follicles in ovaries is of key importance in the field of reproductive biology, both from a developmental point of view, where follicle numbers are determined at specific time points, as well as from a therapeutic perspective, determining the adverse effects of environmental toxins and cancer chemotherapeutics on the reproductive system. The two main factors affecting follicle number estimates are the sampling method and the variation in follicle numbers within animals of the same strain, due to biological variability. This study aims at assessing the effect of these two factors, when estimating ovarian follicle numbers of neonatal mice. We developed computer algorithms, which generate models of neonatal mouse ovaries (simulated ovaries), with characteristics derived from experimental measurements already available in the published literature. The simulated ovaries are used to reproduce in-silico counting experiments based on unbiased stereological techniques; the proposed approach provides the necessary number of ovaries and sampling frequency to be used in the experiments given a specific biological variability and a desirable degree of accuracy. The simulated ovary is a novel, versatile tool which can be used in the planning phase of experiments to estimate the expected number of animals and workload, ensuring appropriate statistical power of the resulting measurements. Moreover, the idea of the simulated ovary can be applied to other organs made up of large numbers of individual functional units.

How Well Do Computer-Generated Faces Tap Face Expertise?

Project description:The use of computer-generated (CG) stimuli in face processing research is proliferating due to the ease with which faces can be generated, standardised and manipulated. However there has been surprisingly little research into whether CG faces are processed in the same way as photographs of real faces. The present study assessed how well CG faces tap face identity expertise by investigating whether two indicators of face expertise are reduced for CG faces when compared to face photographs. These indicators were accuracy for identification of own-race faces and the other-race effect (ORE)-the well-established finding that own-race faces are recognised more accurately than other-race faces. In Experiment 1 Caucasian and Asian participants completed a recognition memory task for own- and other-race real and CG faces. Overall accuracy for own-race faces was dramatically reduced for CG compared to real faces and the ORE was significantly and substantially attenuated for CG faces. Experiment 2 investigated perceptual discrimination for own- and other-race real and CG faces with Caucasian and Asian participants. Here again, accuracy for own-race faces was significantly reduced for CG compared to real faces. However the ORE was not affected by format. Together these results signal that CG faces of the type tested here do not fully tap face expertise. Technological advancement may, in the future, produce CG faces that are equivalent to real photographs. Until then caution is advised when interpreting results obtained using CG faces.

Automatic imitation of human and computer-generated vocal stimuli.

Project description:Observing someone perform an action automatically activates neural substrates associated with executing that action. This covert response, or automatic imitation, is measured behaviourally using the stimulus-response compatibility (SRC) task. In an SRC task, participants are presented with compatible and incompatible response-distractor pairings (e.g., an instruction to say "ba" paired with an audio recording of "da" as an example of an incompatible trial). Automatic imitation is measured as the difference in response times (RT) or accuracy between incompatible and compatible trials. Larger automatic imitation effects have been interpreted as a larger covert imitation response. Past results suggest that an action's biological status affects automatic imitation: Human-produced manual actions show enhanced automatic imitation effects compared with computer-generated actions. Per the integrated theory for language comprehension and production, action observation triggers a simulation process to recognize and interpret observed speech actions involving covert imitation. Human-generated actions are predicted to result in increased automatic imitation because the simulation process is predicted to engage more for actions produced by a speaker who is more similar to the listener. We conducted an online SRC task that presented participants with human and computer-generated speech stimuli to test this prediction. Participants responded faster to compatible than incompatible trials, showing an overall automatic imitation effect. Yet the human-generated and computer-generated vocal stimuli evoked similar automatic imitation effects. These results suggest that computer-generated speech stimuli evoke the same covert imitative response as human stimuli, thus rejecting predictions from the integrated theory of language comprehension and production.

Perceptually motivated loss functions for computer generated holographic displays.

Project description:Understanding and improving the perceived quality of reconstructed images is key to developing computer-generated holography algorithms for high-fidelity holographic displays. However, current algorithms are typically optimized using mean squared error, which is widely criticized for its poor correlation with perceptual quality. In our work, we present a comprehensive analysis of employing contemporary image quality metrics (IQM) as loss functions in the hologram optimization process. Extensive objective and subjective assessment of experimentally reconstructed images reveal the relative performance of IQM losses for hologram optimization. Our results reveal that the perceived image quality improves considerably when the appropriate IQM loss function is used, highlighting the value of developing perceptually-motivated loss functions for hologram optimization.

Advances in computer-generated holography for targeted neuronal modulation

Project description:Abstract. Genetically encoded calcium indicators and optogenetics have revolutionized neuroscience by enabling the detection and modulation of neural activity with single-cell precision using light. To fully leverage the immense potential of these techniques, advanced optical instruments that can place a light on custom ensembles of neurons with a high level of spatial and temporal precision are required. Modern light sculpting techniques that have the capacity to shape a beam of light are preferred because they can precisely target multiple neurons simultaneously and modulate the activity of large ensembles of individual neurons at rates that match natural neuronal dynamics. The most versatile approach, computer-generated holography (CGH), relies on a computer-controlled light modulator placed in the path of a coherent laser beam to synthesize custom three-dimensional (3D) illumination patterns and illuminate neural ensembles on demand. Here, we review recent progress in the development and implementation of fast and spatiotemporally precise CGH techniques that sculpt light in 3D to optically interrogate neural circuit functions.

A computer-generated animated face stimulus set for psychophysiological research.

Project description:Human faces are fundamentally dynamic, but experimental investigations of face perception have traditionally relied on static images of faces. Although naturalistic videos of actors have been used with success in some contexts, much research in neuroscience and psychophysics demands carefully controlled stimuli. In this article, we describe a novel set of computer-generated, dynamic face stimuli. These grayscale faces are tightly controlled for low- and high-level visual properties. All faces are standardized in terms of size, luminance, location, and the size of facial features. Each face begins with a neutral pose and transitions to an expression over the course of 30 frames. Altogether, 222 stimuli were created, spanning three different categories of movement: (1) an affective movement (fearful face), (2) a neutral movement (close-lipped, puffed cheeks with open eyes), and (3) a biologically impossible movement (upward dislocation of eyes and mouth). To determine whether early brain responses sensitive to low-level visual features differed between the expressions, we measured the occipital P100 event-related potential, which is known to reflect differences in early stages of visual processing, and the N170, which reflects structural encoding of faces. We found no differences between the faces at the P100, indicating that different face categories were well matched on low-level image properties. This database provides researchers with a well-controlled set of dynamic faces, controlled for low-level image characteristics, that are applicable to a range of research questions in social perception.

Comparing computer-generated and pathologist-generated tumour segmentations for immunohistochemical scoring of breast tissue microarrays.

Project description:Tissue microarrays (TMAs) have become a valuable resource for biomarker expression in translational research. Immunohistochemical (IHC) assessment of TMAs is the principal method for analysing large numbers of patient samples, but manual IHC assessment of TMAs remains a challenging and laborious task. With advances in image analysis, computer-generated analyses of TMAs have the potential to lessen the burden of expert pathologist review.In current commercial software computerised oestrogen receptor (ER) scoring relies on tumour localisation in the form of hand-drawn annotations. In this study, tumour localisation for ER scoring was evaluated comparing computer-generated segmentation masks with those of two specialist breast pathologists. Automatically and manually obtained segmentation masks were used to obtain IHC scores for thirty-two ER-stained invasive breast cancer TMA samples using FDA-approved IHC scoring software.Although pixel-level comparisons showed lower agreement between automated and manual segmentation masks (?=0.81) than between pathologists' masks (?=0.91), this had little impact on computed IHC scores (Allred; =0.91, Quickscore; =0.92).The proposed automated system provides consistent measurements thus ensuring standardisation, and shows promise for increasing IHC analysis of nuclear staining in TMAs from large clinical trials.
